  • A cisplatina (CP) é um quimioterápico amplamente utilizado para o tratamento de vários tipos de tumores sólidos, contudo a falta de seletividade afeta células saudáveis, provocando diversos efeitos citotóxicos, incluindo a hepatotoxicidade. Embora os mecanismos subjacentes à hepatotoxicidade não estejam totalmente esclarecidos, evidências apontam para uma importante participação do processo inflamatório, no qual o aumento da expressão de citocinas pró-inflamatórias, redução de citocinas anti-inflamatórias e infiltração de células imunes, são determinantes no agravamento e progressão dos danos teciduais. Recentemente, alguns estudos têm reportado os efeitos hepatoprotetores mediados pela prática regular de exercícios físicos aeróbicos, contudo ainda não está claro qual intensidade é mais eficaz em ampliar esses efeitos protetores, especialmente em lesões hepáticas agudas. Assim, este estudo tem como objetivo comparar os efeitos entre o pré-condicionamento com o treinamento intervalado de alta intensidade (HIIT) e treinamentos tradicionais contínuos de leve (LIT) e moderada (MIT) intensidades sobre marcadores inflamatórios em ratas wistar com hepatotoxicidade induzida por CP. Para tanto, 35 ratas wistar foram divididas em cinco grupos (n=7 em cada grupo): controle e sedentários (C+S); tratados com CP e sedentário (CP+S); tratados com CP e submetidos ao LIT (CP+LIT); tratados com CP e submetidos ao MIT (CP+MIT) e tratados com CP e submetidos ao HIIT (CP+HIIT). Os protocolos de treinamentos consistiram de corrida em esteira motorizada, 5 dias por semana, por 8 semanas, antes do tratamento com CP. Ao final das 8 semanas de treinamento, as ratas receberam uma única injeção de CP (5 mg/kg i.p) ou solução salina, e 7 dias depois da injeção foram eutanasiadas. Amostras do fígado foram coletadas para avaliar a expressão do fator de transcrição nuclear kappa B (NF-ΚB), do receptor toll like 4 (TLR4), fator de necrose tumoral alfa (TNF-α), interleucina (IL)-1α, IL-1β, IL-6, IL-8 e IL-10, o número de células ED-1 positivas e marcadores de macrófagos M1 (iNOS) e M2 (arginase) no tecido hepático. Os nossos resultados indicam que o HIIT foi o único protocolo de exercício capaz de prevenir o aumento de todas as citocinas pró-inflamatórias analisadas e reduzir o número de células ED-1 positivas, pelo menos em parte através da atenuação da ativação da via TLR4/NF-κB. Adicionalmente, o pré-condicionamento com o HIIT potencializou as expressões da citocina anti-inflamatória IL-10 e do marcador de macrófagos do tipo M2, ao passo em que reduziu a expressão do marcador de macrófagos do tipo M1 no tecido hepático. Desse modo, o presente estudo sugere que o pré-condicionamento físico com o HIIT é mais eficaz em promover efeitos hepatoprotetores do que os protocolos de pré- condicionamentos com LIT e MIT, regulando importantes marcadores inflamatórios através da modulação da via de sinalização TLR4/NF-ΚB no tecido hepático de ratas tratadas com CP.

  • Cisplatin (CP) is a chemotherapy agent widely used to treat various types of solid tumors. However, its lack of selectivity affects healthy cells, causing several cytotoxic effects, including hepatotoxicity. Although the underlying mechanisms of hepatotoxicity are not fully understood, evidence suggests a significant involvement of the inflammatory process, in which increased expression of pro-inflammatory cytokines, reduction of anti-inflammatory cytokines, and immune cell infiltration are determinants in the exacerbation and progression of tissue damage. Recently, some studies have reported the hepatoprotective effects mediated by regular aerobic physical exercise. However, it is still unclear which intensity is most effective in enhancing these protective effects, especially in acute liver injuries. Therefore, this study aims to compare the impacts between preconditioning with high-intensity interval training (HIIT) and traditional continuous training of light (LIT) and moderate (MIT) intensities on inflammatory markers in Wistar rats with CP-induced hepatotoxicity. For this purpose, 35 Wistar female rats were divided into five groups (n=7 in each group): control and sedentary (C+S); treated with CP and sedentary (CP+S); treated with CP and subjected to LIT (CP+LIT); treated with CP and subjected to MIT (CP+MIT); and treated with CP and subjected to HIIT (CP+HIIT). The training protocols consisted of treadmill running, 5 days a week, for 8 weeks before the CP treatment. At the end of the 8-week training period, the rats received a single injection of CP (5 mg/kg i.p) or saline, and 7 days after the injection, they were euthanized. Liver samples were collected to evaluate the expression of nuclear factor kappa B (NF-κB), toll-like receptor 4 (TLR4), tumor necrosis factor alpha (TNF-α), interleukin (IL)-1α, IL-1β, IL-6, IL-8, and IL-10, the number of ED-1 positive cells, and M1 (iNOS) and M2 (arginase) macrophages markers in the hepatic tissue. Our results indicate that HIIT was the only exercise protocol capable of preventing the increase in all analyzed pro-inflammatory cytokines and reducing the number of ED-1 positive cells, at least in part through attenuating the TLR4/NF-κB signaling pathway. Additionally, preconditioning with HIIT enhanced the expression of the anti-inflammatory cytokine IL-10 and the M2 macrophage marker while reducing the expression of the M1 macrophage marker in hepatic tissue. Thus, this study suggests that preconditioning with HIIT is more effective in promoting hepatoprotective effects than LIT and MIT preconditioning protocols, regulating important inflammatory markers through modulation of the TLR4/NF-κB signaling pathway in the hepatic tissue of female rats treated with CP.

  • Objetivo: Avaliar os efeitos do treinamento físico e da dieta restrita em carboidratos e hiperproteica sobre a modulação da irisina plasmática e muscular e alterações morfológicas e funcionais no tecido adiposo branco e marrom de ratos Wistar machos em um modelo de obesidade. Métodos: 40 ratos Wistar machos foram divididos em dois grupos: Grupo controle (n=6) e Grupo com dieta hipercalórica (HC) para indução da obesidade (n=34) durante 8 semanas. Após esse período, 6 animais de cada grupo foram eutanasiados para confirmação da obesidade. Em seguida, os animais obesos foram divididos em 4 grupos: Grupo submetido à dieta HC e sedentário (n=7); Grupo submetido à dieta HC e treinado (n=7); Grupo submetido à dieta hiperproteica e restrita em carboidratos (HP) e sedentário; Grupo submetido à dieta hiperproteica e restrita em carboidratos (HP) e treinados (n=7). Após o período de 12 semanas, os animais foram eutanasiados e os tecidos adiposos subcutâneo, epididimal e marrom, assim como o tecido muscular e o sangue torácico foram coletados para posteriores análises. Avaliou-se a composição corporal, modulação sérica e tecidual de irisina/FNDC5, morfologia do músculo gastrocnêmio e dos tecidos adiposos subcutâneo (TASC) e marrom (TAM). Foram avaliados também parâmetros de atividade mitocondrial e metabolismo energético no TASC e TAM e marcadores inflamatórios, de dano oxidativo e atividade das enzimas antioxidantes no TASC, TAE e TAM. As análises
    estatísticas foram realizadas através do teste ANOVA two-way (2 dietas x 2 condições de treinamento), seguida pelo pós-teste de Bonferroni. Os dados foram apresentados como média±erro padrão. Resultados: A dieta HP e o treinamento físico influenciaram positivamente os parâmetros de composição corporal, morfologia do TASC e do músculo, parâmetros de metabolismo do TASC e parâmetros inflamatórios dos tecidos avaliados. No entanto, o efeito do treinamento físico foi superior à dieta HP por proteger os animais treinados de alterações no balanço redox e no perfil inflamatório de forma mais acentuada, além de determinar o aumento na concentração sérica de irisina. Conclusão: No modelo estudado, os efeitos da mudança na composição dietética e do treinamento físico foram determinantes para proteger os animais das alterações provocadas pela obesidade sobre parâmetros de composição corporal, morfologia e metabolismo tecidual, perfil inflamatório e oxidativo dos tecidos adiposos. Nossos achados reforçam a importância do treinamento físico
    regular ao determinar maior concentração de irisina circulante e modulação positiva no balanço redox e perfil inflamatório de todos os tecidos avaliados.

  • Objective: To evaluate the effects of physical training and a carbohydrate-restricted and high-protein diet on the modulation of plasma and muscle irisin and morphological and functional changes in white and brown adipose tissue of male Wistar rats in an obesity model. Methods: 40 male Wistar rats were divided into two groups: Control group (n=6) and Group with a high-calorie diet (HC) to induce obesity (n=34) for 8 weeks. After this period, 6 animals from each group were euthanized to confirm obesity. Then, the obese animals were divided into 4 groups: Group submitted to the HC diet and sedentary (n=7); Group submitted to the HC diet and trained (n=7); Group submitted to a high-protein, carbohydrate-restricted
    (HP) diet and sedentary; Group submitted to a high-protein and carbohydrate-restricted diet (HP) and trained (n=7). After a period of 12 weeks, the animals were euthanized and the subcutaneous, epididymal and brown adipose tissues, as well as muscle tissue and thoracic blood were collected for further analysis. Body composition, serum and tissue modulation of irisin/FNDC5, morphology of the gastrocnemius muscle and subcutaneous (TASC) and brown (TAM) adipose tissues were evaluated. Mitochondrial activity and energy metabolism parameters in TASC and TAM and inflammatory markers, oxidative damage and antioxidant enzyme activity in TASC, TAE and TAM were also evaluated.Statistical analyzes were performed using the two-way ANOVA test (2 diets x 2 training conditions), followed by the Bonferroni post-test. Data were presented as mean±standard error. Results: The HP diet and physical training positively influenced body composition parameters, TASC and muscle morphology, TASC metabolism parameters and inflammatory parameters of the evaluated tissues. However, the effect of physical training was superior to the HP diet by protecting the trained animals from changes in the redox balance and inflammatory profile in a more pronounced way, in addition to determining the increase in the serum concentration of irisin. Conclusion: In the model studied, the effects of changes in dietary composition and physical training were decisive in protecting animals from changes caused by obesity on parameters of body composition, tissue morphology and metabolism, and the inflammatory and oxidative profile of adipose tissues. Our findings reinforce the importance of regular physical training by determining a higher concentration of circulating irisin and positive modulation in the redox balance and inflammatory profile of all tissues evaluated.

  • Introdução: Infecções de pele e tecidos moles são um importante problema de saúde pública mundial, tanto pela morbidade associada às suas lesões bem como por serem porta de entrada para infecções mais invasivas, que frequentemente culminam no óbito do paciente acometido. Staphylococcus aureus (S. aureus) é a principal causa desse tipo de infecção, de modo que a sua significativa adaptabilidade e o desenvolvimento de resistência são os principais fatores ligados à sua propagação e aos desafios no seu tratamento. Nesse contexto, a terapia fotodinâmica antimicrobiana (TFA) com produtos naturais tem se mostrado uma boa alternativa no combate às infecções causadas por microrganismos resistentes a antibióticos, pois agregam benefícios inerentes a esses compostos aos danos causados pelas espécies reativas de oxigênio induzidas pela TFA. A própolis verde brasileira possui efeitos conhecidos como agente antimicrobiano e imunomodulador. Contudo, não se sabe ao certo se tais atividades seriam mantidas se utilizada no contexto da TFA. Objetivos: O objetivo do presente estudo foi caracterizar a própolis verde brasileira como fotossensibilizador para TFA, avaliar os seus efeitos antimicrobianos e imunomoduladores in vivo e desenvolver microemulsão como proposta terapêutica de bioproduto para o tratamento de infecções intradérmicas causadas por bactérias resistentes a antibióticos. Materiais e Métodos: Inicialmente, foram realizados estudos in vitro para se caracterizar o potencial fotodinâmico antimicrobiano da própolis verde brasileira. Para tal, realizou-se espectrometria de absorção molecular UV-Vis para avaliar os comprimentos de onda com potencial para ativar a própolis verde. Posteriormente, cepas de referência de Staphylococcus aureus Resistente à Meticilina (MRSA ATCC 43300) e Staphylococcus aureus Intermediário à Vancomicina (VISA ATCC 700699) foram expostas a concentrações variadas de própolis verde: 1µg/mL, 5µg/mL, 10µg/mL, 50µg/mL e 100µg/mL e estimuladas por luz LED azul, verde ou vermelha. Em seguida, avaliou-se o potencial zeta das bactérias expostas à própolis verde brasileira a fim de caracterizar um possível local de ação da própolis. Logo após, cromatografia líquida de alta eficiência com detector de arranjo de diodos acoplado à espectrometria de massas em tandem, juntamente com análise clássica de redes moleculares, foram realizadas para identificar potenciais moléculas bioativas com atividade fotodinâmica. Finalmente, foram realizadas análises de citotoxicidade em eritrócitos e em células nucleadas. Após essa etapa, avaliou-se os efeitos antimicrobianos e imunomoduladores da utilização da própolis verde brasileira como fotossensibilizador contra S. aureus in vivo, em modelo de infecção intradérmica em camundongos. Nesse momento, 24 camundongos Balb/c foram infectados por via intradérmica nas orelhas com 1,5x108 unidades formadoras de colônias de MRSA 43300. Após a infecção, foram separados em 4 grupos (6 animais por grupo) e tratados com o veículo, apenas própolis verde brasileira, somente luz LED azul ou com protocolo aPDT (própolis verde brasileira + luz LED azul). Por fim, desenvolveu-se microemulsão como um sistema de entrega transdérmica da própolis verde brasileira, caracterizando-se o sistema microemulsionado e realizando os testes de estabilidade acelerada. Resultados: Observou-se que a própolis verde brasileira exibe um pico de absorção pronunciado e fotorresponsividade aumentada quando exposta à luz azul na faixa de 400nm e 450nm. Esta característica revela atividade fotodinâmica significativa contra MRSA e VISA em concentrações a partir de 5 µg/mL. Além disso, a própolis compreende compostos como a curcumina e outros flavonóides provenientes da flavona, que possuem potencial para atividade fotodinâmica e outras funções antimicrobianas. Os testes com potencial zeta não apresentaram diferenças significativas quando as bactérias foram expostas à própolis verde brasileira, o que sugere potencial internalização do composto pelas bactérias. Além disso, a própolis verde brasileira não apresentou toxicidade em eritrócitos e células nucleadas. In vivo, a TFA com própolis verde brasileira reduziu a carga bacteriana no local da infecção. Também foi capaz de inibir a perda de peso decorrente da infecção, bem como modular a resposta inflamatória através de maior recrutamento de células neutrófilos para o tecido infectado. Finalmente, a TFA induziu aumento das citocinas IL-17A e IL-12p70 no linfonodo retromaxilar drenante. Por fim, a microemulsão com própolis verde brasileira apresentou tamanho de partícula, índice de polidispersividade e potencial zeta promissores, além de possuir estabilidade, verificada nos testes de estabilidade preliminares, sendo mantida a atividade fotodinâmica antimicrobiana induzida por luz azul. Conclusões: A própolis verde brasileira apresenta atividade fotodinâmica quando estimulada por luz LED azul, sendo capaz de induzir a morte de diferentes cepas de S. aureus in vitro, mas também por reduzir a carga bacteriana e modular a resposta inflamatória em modelo murino de infecção intradérmica. Além disso, o desenvolvimento de uma microemulsão com a própolis verde brasileira mostra-se promissor, uma vez que apresenta características físico-químicas de interesse e estabilidade viabilizando o desenvolvimento de protocolos que não demandem a injeção local do fotossensibilizador, não gerando dor e incômodo, o que pode diminuir o abandono terapêutico.

  • Introduction: Skin and soft tissue infections are an important global public health problem, both due to the morbidity associated with their lesions and because they are a gateway to more invasive infections, which often culminate in the death of the affected patient. Staphylococcus aureus is the main cause of this type of infection, so its significant adaptability and the development of resistance are the main factors linked to its spread and the challenges in its treatment. In this context, antimicrobial photodynamic therapy (aPDT) with natural products has proven to be a good alternative in combating infections caused by antibiotic-resistant microorganisms, as they add benefits inherent to these compounds to the damage caused by reactive oxygen species induced by aPDT. Brazilian green propolis has known effects as an antimicrobial and immunomodulatory agent. However, it is not known for certain that such activities would be maintained if used in the context of aPDT. Objective: The objective of the present study was to characterize Brazilian green propolis as a photosensitizer for aPDT, evaluate its antimicrobial and immunomodulatory effects in vivo and develop microemulsion as a therapeutic bioproduct proposal for the treatment of intradermal infections caused by antibioticresistant bacteria. Materials and Methods: Initially, in vitro studies were carried out to characterize the antimicrobial photodynamic potential of Brazilian green propolis. To this end, UV-Vis molecular absorption spectrometry was carried out to evaluate the wavelengths with the potential to activate green propolis. Subsequently, reference strains of Methicillin-Resistant Staphylococcus aureus (MRSA ATCC 43300) and VancomycinIntermediate Staphylococcus aureus (VISA ATCC 700699) were exposed to varying concentrations of green propolis: 1µg/mL, 5µg/mL, 10µg/mL, 50µg/mL mL and 100µg/mL and stimulated by blue, green or red LED light. Next, the zeta potential of bacteria exposed to Brazilian green propolis was evaluated in order to characterize a possible site of propolis action. Soon after, high performance liquid chromatography with diode array detector coupled to tandem mass spectrometry, together with classical molecular network analysis, were performed to identify potential bioactive molecules with photodynamic activity. Finally, cytotoxicity analyzes were performed on erythrocytes and nucleated cells. After this step, the antimicrobial and immunomodulatory effects of using Brazilian green propolis as a photosensitizer against S. aureus in vivo were evaluated, in an intradermal infection model in mice. At that time, 24 Balb/c mice were infected intradermally in the ears with 1.5x108 colony-forming units of MRSA 43300. After infection, they were separated into 4 groups (6 animals per group) and treated with the vehicle, only propolis Brazilian green, only blue LED light or with aPDT protocol (Brazilian green propolis + blue LED light). Finally, microemulsion was developed as a transdermal delivery system for Brazilian green propolis, characterizing the microemulsified system and carrying out accelerated stability tests. Results: It was observed that Brazilian green propolis exhibits a pronounced absorption peak and increased photoresponsivity when exposed to blue light in the range of 400nm and 450nm. This characteristic reveals significant photodynamic activity against MRSA and VISA at concentrations from 5 µg/mL. Furthermore, propolis comprises compounds such as curcumin and other flavonoids from flavones, which have potential for photodynamic activity and other antimicrobial functions. Tests with zeta potential did not show significant differences when the bacteria were exposed to Brazilian green propolis, which suggests potential internalization of the compound by the bacteria. Furthermore, Brazilian green propolis did not present toxicity to erythrocytes and nucleated cells. In vivo, TFA with Brazilian green propolis reduced the bacterial load at the site of infection. It was also able to inhibit weight loss resulting from infection, as well as modulate the inflammatory response through greater recruitment of polymorphonuclear cells/neutrophils to the infected tissue. Finally, the microemulsion with Brazilian green propolis showed promising particle size, polydispersity index and zeta potential, in addition to having stability, verified in preliminary stability tests, with antimicrobial photodynamic activity induced by blue light being maintained. Conclusions: Brazilian green propolis presents photodynamic activity when stimulated by blue LED light, being able to induce the death of different strains of S. aureus in vitro, but also to reduce the bacterial load and modulate the inflammatory response in a murine model of intradermal infection. Furthermore, the development of a microemulsion with Brazilian green propolis shows promise, as it presents interesting physical-chemical characteristics and stability, enabling the development of protocols that do not require local injection of the photosensitizer, without generating pain and discomfort, which can reduce therapeutic abandonment.

  • A cisplatina (CP) é um antineoplásico amplamente utilizado no tratamento de diversos tipos

    de tumores sólidos, contudo o seu uso é limitado principalmente pela elevada

    nefrotoxicidade, a qual é manifestada pela injúria renal aguda (IRA). A fisiopatologia da

    IRA é complexa e envolve, dentre outros mecanismos, a ativação de diversas vias de

    sinalização apoptótica. Por outro lado, alguns estudos têm demonstrado efeitos protetores do

    exercício físico aeróbico sobre a IRA. Para além disso, o estresse mecânico é conhecido por

    aumentar a produção de IL-11, uma citocina pleiotrópica com efeitos anti apoptóticos e anti

    necróticos.Assim, o objetivo do presente estudo foi comparar o impacto do treinamento

    intervalado de alta intensidade (HIIT) com os treinamentos contínuos de leve (LIT) e

    moderada (MIT) intensidades sobre os níveis renais de IL-11 e expressões de marcadores

    apoptóticos no tecido renal de ratas com nefrotoxicidade induzida por CP. Para tanto, 35

    ratas wistar foram divididas em cinco grupos (n=7 em cada grupo): controle e sedentários

    (C+S); tratados com CP e sedentário (CP+S); tratados com CP e submetidos ao LIT

    (CP+LIT); tratados com CP e submetidos ao MIT (CP+MIT) e tratados com CP e submetidos

    ao HIIT (CP+HIIT). Os protocolos de treinamentos consistiram de corrida em esteira

    avaliar o nitrogênio ureico sérico (BUN), expressão gênica do receptor de TNF 1 (TNFR1)

    e 2 (TNFR2), caspase-3, MAPK(p38) (MAPK14), p53, Bax, Bak, Bcl-2 e Bcl-xL, níveis

    renais de IL-11 e p53, e imunolocalização de caspase-3 clivada, Bax, Bcl-2 e MAPK(p38)

    no tecido renal. Os dados revelam que todos os grupos treinados demonstraram um aumento

    significativo nos níveis renais de IL-11 associado à redução da expressão local de

    marcadores pró-apoptóticos e aumento dos marcadores antiapoptóticos. Tais efeitos foram

    dependentes da intensidade do treinamento, e, portanto, mais pronunciados com o HIIT.

    Como conclusão, o presente estudo sugere que o HIIT parece proporcionar renoproteção

    superior aos treinamentos contínuos de intensidades leve e moderada, modulando

    beneficamente as vias de sinalização apoptóticas, e consequentemente atenuando a IRA

    induzida por CP neste modelo experimental. Além disso, tal efeito pode estar relacionado,

    pelo menos em parte, ao aumento dos níveis renais de IL-11 induzido pelo treinamento,

    também de maneira dependente da sua intensidade.

  • Cisplatin (CP) is an antineoplastic widely used in the treatment of different types of solid tumors, but its use is mainly limited by high nephrotoxicity, which is manifested by acute kidney injury (AKI). The pathophysiology of ARF is complex and involves, among other controls, the activation of several apoptotic signal pathways. On the other hand, some studies have shown protective effects of aerobic exercise on ARF. Furthermore, mechanical stress is known to increase the production of IL-11, a pleiotropic cytokine with anti-apoptotic and anti- necrotic effects. Thus, the aim of the present study was to compare the impact of high intensity interval training (HIIT) with continuous light (LIT) and moderate intensity (MIT) training on renal IL-11 levels and expressions of apoptotic markers in tissue renal rate of CP-induced nephrotoxicity. For this purpose, 35 female Wistar rats were divided into five groups (n=7 in each group): control and sedentary (C+S); treated with CP and sedentary (CP+S); treated with CP and approved to LIT (CP+LIT); CP-treated and MIT-approved (CP+MIT) and CP-treated and HIIT-approved (CP+HIIT). Training protocols consisted of running on a motorized treadmill, 5 days a week, for 8 weeks. At the end of the 8 weeks of training, the rates received a single injection of CP (5 mg/kg) or saline solution, and 7 days after the injection they were euthanized. Serum samples and animal rinses were collected to assess serum urea urea (BUN), TNF receptor 1 (TNFR1) and 2 (TNFR2) gene expression, caspase-3, MAPK(p38) (MAPK14), p53, Bax, Bak, Bcl-2 and Bcl-xL, renal IL-11 and p53 levels, and immunolocalization of cleaved caspase-3, Bax, Bcl-2 and MAPK(p38) in renal tissue. The data reveal that all trained groups experience a significant increase in renal IL-11 levels associated with reduced local expression of pro-apoptotic markers and increased anti-apoptotic markers. Such effects were dependent on training intensity, and therefore more pronounced with HIIT. In conclusion, the present study suggests that HIIT seems to provide superior renoprotection to continuous training at mild and moderate intensities, beneficially modulating apoptotic signal pathways, and consequently attenuating ARF induced by CP in this experimental model. Furthermore, such an effect may be related, at least in part, to the increase in the renal levels of IL-11 induced by training, also dependent on its intensity.

  • Aborto é a interrupção da gestação antes da 22ª semana gestacional, ou o feto pesando menos de 500g. Podendo ser de maneira habitual ou induzida, possuindo uma multicausalidade, que englobam infecções genitais. As infecções do trato genital ascendem para cavidade uterina sendo responsável por 15% dos abortos precoces e 66% nos abortos tardios. As respostas inflamatórias exacerbadas oriundas do processo infeccioso acometem a homeostase gestacional, contribuindo com o perfil pró-inflamatório e modificando o estado oxidativo. Este trabalho teve como objetivo principal avaliar os marcadores de estresse oxidativo no contexto do aborto espontâneo associado as infecções por microrganismos de importância ginecológica. Foram utilizadas amostras biológicas de tecido placentário de mulheres que evoluíram com aborto espontâneo e mulheres que tiveram parto a termo. Ao avaliar as enzimas do estresse oxidativo, foram identificados níveis elevados de marcadores da peroxidação lipídica (TBARS e proteína carbonilada) no grupo aborto, bem como o aumento da atividade antioxidante pela CAT, e redução da atividade da GPx, além de uma resposta não alterada da SOD. Entretanto, o perfil da peroxidação lipídica foi acentuado no grupo aborto infectado por UP. E os níveis da GPx foram reduzidos no grupo aborto infectado por UP, comparado ao controle infectado, enquanto SOD e CAT não apresentaram diferenças; a razão SOD/CAT esteve reduzida e SOD/GPx elevada nos grupos de UP, comparado aos sem infecção. Na razão da expressão gênica, o aborto contribuiu com a elevação de Mn SOD/GPx e infecção de UP reduziu a expressão de CuZn SOD/GPx e Mn SOD/GPx no grupo aborto e controle infectado, comparado aos grupos sem infecção. Em seguida, houve aumento dos níveis de nitrito no grupo aborto infectado por UP. Este trabalho demonstrou que a infecção por UP pode contribuir para alterar o perfil oxidante, sugerindo, assim, que o desequilíbrio do estresse oxidativo pode contribuir com a ocorrência do aborto espontâneo associado a infecção bacteriana.

  • Abortion is the termination of pregnancy before the 22nd gestational week or when the fetus weighs less than 500g. It can occur spontaneously or be induced and has multiple causes, including genital infections. Genital tract infections can ascend to the uterine cavity and account for 15% of early abortions and 66% of late abortions. The exacerbated inflammatory responses resulting from the infectious process disrupt gestational homeostasis, contributing to a pro-inflammatory profile and altering the oxidative state. The main objective of this study was to evaluate oxidative stress markers in the context of spontaneous abortion associated with infections by gynecologically significant microorganisms. Biological samples of placental tissue from women who experienced spontaneous abortion and women who had full-term deliveries were used. When evaluating oxidative stress enzymes, elevated levels of lipid peroxidation markers (TBARS and carbonylated protein) were identified in the abortion group, along with increased antioxidant activity by CAT and reduced GPx activity, while SOD showed an unchanged response. However, the profile of lipid peroxidation was accentuated in the abortion group infected with UP. GPx levels were reduced in the UP-infected abortion group compared to the infected control, while SOD and CAT showed no differences. The SOD/CAT ratio was reduced and the SOD/GPx ratio was increased in the UP groups compared to the non-infected groups. In terms of gene expression ratio, abortion contributed to an increase in Mn SOD/GPx, while UP infection reduced the expression of CuZn SOD/GPx and Mn SOD/GPx in both the abortion and infected control groups compared to the non-infected groups. Furthermore, nitrite levels were increased in the UP-infected abortion group. This study demonstrated that UP infection may contribute to altering the oxidative profile, suggesting that an imbalance in oxidative stress could contribute to the occurrence of spontaneous abortion associated with bacterial infection.

  • A espécie vegetal a ser estudada é a Commiphora Leptophloeos (Mart.) J.B. Gillet, conhecida popularmente como umburana fêmea do gênero Commiphora, família Burseraceae, é uma espécie endêmica da região do povoado de Palmeiras, distrito da cidade de Contendas do Sincorá – BA. É utilizada tradicionalmente segundo a população local como fortificante, cicatrizante de feridas e contra tosse, bronquite, gastrite úlcera e afecções urinárias. O estudo teve como objetivo avaliar o potencial medicinal dessa planta como anti-inflamatório, antinociceptivo, antioxidante e ação toxicológica. O extrato etanólico foi preparado por maceração e após evaporação foi obtido o extrato bruto com o rendimento de 2,2%. A prospecção fitoquímica foi realizada por CCD com reveladores específicos e perfil cromatográfico avaliado por CG-EM com destaque para a identificação de: estigmasterol, lupeol, ácido protocatecuico, lanosterol, oleato de etila e linoleato de etila. O ensaio antioxidante foi realizado através da avaliação da capacidade antioxidante total, pelo método do fosfomolibdato, no qual o extrato da planta apresentou capacidade antioxidante. Para os ensaios de nocicepção e inflamação foram utilizados camundongos machos adultos Balb/c. O efeito antinociceptivo foi avaliado pelos testes: Contorção abdominal induzido por ácido acético 0,6%, onde todas as concentrações (25, 50, 100 e 200 mg/kg) promoveram redução significativa das respostas nociceptivas (p < 0,05). Já a concentração de 100mg/kg do extrato etanólico demonstrou ter seus efeitos antinociceptivos mediados pela via colinérgica (p < 0,05). Injeção intraplantar de formalina 1,5%, na fase 1 e 2, todas as concentrações (50, 100 e 200 mg/kg) reduziram o número de “flinches”, em comparação ao controle negativo (p < 0,05), quando comparadas ao grupo controle positivo morfina, as concentrações de 50 e 200 mg/kg não apresentaram diferença estatística (p < 0,05), o edema de pata formado não obteve redução significativa em nenhuma concentração testada (p < 0,05). No Von Frey eletrônico por injeção intraplantar de carragenina (Cg), todas as concentrações (50, 100, 200 mg/kg) foram capazes de reduzir significativamente a hipernocicepção (p < 0,05). Para a determinação da atividade anti-inflamatória foram realizados testes de: Migração de neutrófilos para a cavidade peritoneal, onde todas as concentrações (50, 100, 200 mg/kg) apresentaram redução significativa da migração de neutrófilos em (p < 0,05); Determinação dos níveis de nitrito, todas as concentrações (50, 100, 200 mg/kg) apresentaram redução significativa (p < 0,05). Avaliação da permeabilidade vascular por azul de Evans, que mostrou uma significativa redução do extravasamento do corante no líquido peritoneal em todas as concentrações testadas (50, 100, 200 mg/kg), (p < 0,05). A determinação da toxicidade foi realizada com Artemia salina Leach e a classificação de toxicidade foi altamente tóxica. A citotoxicidade foi investigada por meio do ensaio de MTT, onde as concentrações de 25, 50 e 100ug/kg conseguiram diminuir a viabilidade das células tumorais (p < 0,05). Os resultados permitem considerar que a C. leptophloeos (Mart.) é uma promissora fonte natural para a identificação de novos compostos bioativos e agentes terapêuticos, bem como uma espécie promissora para uso quanto fitoterápico.

  • The plant species to be studied is Commiphora leptophloeos (Mart.) J.B. Gillet, popularly known as umburana fêmea of the genus Commiphora, family Burseraceae, is an endemic species of the region of the village of Palmeiras, district of the city of Contendas do Sincorá - BA. According to the local population, it is traditionally used as a tonic, healing wounds and against coughs, bronchitis, gastritis, ulcers and chronic conditions. The study aimed to evaluate the medicinal potential of this plant as an antiinflammatory, antinociceptive, antioxidant and toxicological action. The ethanolic extract was prepared by maceration and after evaporation the crude extract was obtained with a yield of 2.2%. The phytochemical prospection was carried out by CCD with specific developers and the chromatographic profile evaluated by GC-MS, highlighting the identification of: stigmasterol, lupeol, protocatechuic acid, lanosterol, ethyl oleate and ethyl linoleate. The antioxidant assay was carried out by evaluating the total antioxidant capacity, using the phosphomolybdate method, in which the plant extract showed antioxidant capacity. For nociception and inflammation assays, adult male Balb/c mice were used. The antinociceptive effect was evaluated by the tests: Abdominal writhing induced by 0.6% acetic acid, where all concentrations (25, 50, 100 and 200 mg/kg) promoted a significant reduction in nociceptive responses (p < 0.05). The concentration of 100mg/kg of the ethanolic extract demonstrated that its antinociceptive effects were mediated by the cholinergic pathway (p < 0.05). Intraplantar injection of 1.5% formalin, in phase 1 and 2, all concentrations (50, 100 and 200 mg/kg) reduced the number of flinches, compared to the negative control (p < 0.05), when incidents to the morphine positive control group, the concentrations of 50 and 200 mg/kg did not show statistical difference (p < 0.05), the paw edema formed did not obtain a significant reduction in any experimental concentration (p < 0.05). In electronic Von Frey by intraplantar injection of carrageenan (Cg), all concentrations (50, 100, 200 mg/kg) were able to significantly reduce hypernociception (p < 0.05). For the experiment of anti-inflammatory activity, tests were performed: Migration of neutrophils to the peritoneal cavity, where all concentrations (50, 100, 200 mg/kg) established a significant reduction in neutrophil migration in (p < 0.05) ; The determination of nitrite levels, all concentrations (50, 100, 200 mg/kg) showed a significant reduction (p < 0.05). Evaluation of vascular permeability by Evans blue, which showed a significant reduction in dye leakage into the peritoneal fluid at all doses (50, 100, 200 mg/kg), (p < 0.05). Toxicity innovation was performed with Artemia salina Leach and the toxicity classification was highly toxic. Cytotoxicity was investigated using the MTT assay, where concentrations of 25, 50 and 100ug/kg were shown to decrease tumor cell viability (p < 0.05). The results allow us to consider that C. leptophloeos (Mart.) is a promising natural source for the identification of new bioactive compounds and therapeutic agents, as well as a promising species for use as a herbal medicine

  • As Infecções Sexualmente Transmissíveis (IST) são definidas como um grupo de doenças causadas por microrganismos transmitidos principalmente através do contato sexual sem o uso de preservativo. Além do HIV, outras IST bacterianas são consideradas epidêmicas no Brasil, destacando-se a gonorreia (Neisseria gonorrhoeae), infecções por micoplasmas (Mycoplasma genitalium, Mycoplasma hominis, Ureaplasma urealyticum, Ureaplasma parvum) e por Chlamydia trachomatis. Durante a infecção por IST células imunológicas produzem uma série de mediadores químicos como: fator de necrose tumoral alfa (TNF-α), interleucinas IL1-β, IL-6, IL-10, dentre outros, que atuam orquestrando a resposta imunológica. Objetivo: definir a assinatura de em jovens homens que fazem sexo com homens e mulheres transsexuais, correlacionando os resultados com a prevalência de IST por Neisseria gonorrhoeae, Chlamydia trachomatis e Mycoplasmas. Metodologia: Trata-se de um estudo de corte transversal que faz parte de um projeto piloto intitulado “Estudo PrEP 15-19” (Título: Estudo da efetividade da Profilaxia Pré-Exposição [PrEP]. Foram coletadas amostras clínicas em duplicata de cada participante, no momento da primeira visita ao serviço. Para isso, foram obtidas por fricção de swab na  mucosa uretral peniana, oral e anal, sendo armazenadas em 5 mL de meio de transporte. A quantificação dos microrganismos foi realizada por meio de técnica de quantificação absoluta. As dosagens das citocinas IL-1β, IL-6, IL-10 e fator de necrose tumoral TNF-α foram realizadas utilizando-se kit de ELISA eBioscience. Resultados: Nossa amostra foi composta predominantemente por homens que fazem sexo com homens, pretos ou pardos, com idade entre 18 e 19 anos. Destes, 17,36% apresentaram uma ou mais infecções. Houve associação entre ensino médio completo com IST (p 0,045). Dentre as IST estudadas destaca-se a prevalência de 85,3% de micoplasmas. Na avaliação do perfil de citocinas secretados entre infectados e não infectados não encontramos associação estatisticamente significativa. Testes de matriz foram realizados entre os infectados com associação entre maiores valores de TNF-α e IL-6. Na população de não infectados, observa-se associação de maneira geral entre as citocinas, ou seja, os participantes com níveis menores apresentaram redução de todas as citocinas estudadas. Neste estudo tivemos a prevalência de 4,73% de coinfecções, todas com pelo menos um micoplasma. 50% das coinfecções encontrada foram do gênero Mycoplasma hominis e Ureaplasma urealyticum. Na avaliação da assinatura de citocinas entre mono e coinfectados e encontramos maiores níveis de IL-1β em indivíduos monoinfectados quando comparados aos co-infectados e de TNF-α nos monoinfectados, quando comparados com não infectados e coinfectados. Na observação do heatmap com clusters não supervisionado notam-se dois grupos, um com maior potencial inflamatório,e outro com menor potencial, com destaque para M. hominis e U. urealyticum  com alto potencial de resposta inflamatória.  Na Análise Principal Component fica evidenciada a associação entre infecção por M. hominis e secreção de IL1-β. Conclusão: Nosso estudo não encontrou diferença estatística entre jovens infectados e não infectados, entretanto, a infecção por micoplasma e a coinfecção por gonorreia e clamidia demonstram associação com o aumento de citocinas.

  • Sexually Transmitted Infections (STIs) are defined as a group of diseases caused by microorganisms transmitted mainly through sexual contact without the use of condoms. In addition to HIV, other bacterial STIs are considered epidemic in Brazil, especially gonorrhea (Neisseria gonorrhoeae), mycoplasma infections (Mycoplasma genitalium, Mycoplasma hominis, Ureaplasma urealyticum, Ureaplasma parvum) and Chlamydia trachomatis. During STI infection, immune cells produce a series of chemical mediators such as: tumor necrosis factor alpha (TNF-α), interleukins IL1-β, IL-6, IL-10, among others, which act by orchestrating the immune response. Objective: to define the signature of in young men who have sex with men and transsexual women, correlating the results with the prevalence of STIs due to Neisseria gonorrhoeae, Chlamydia trachomatis and Mycoplasmas. Methodology: This is a cross-sectional study that is part of a pilot project entitled “PrEP Study 15-19” (Title: Study of the effectiveness of Pre-Exposure Prophylaxis [PrEP]. Clinical samples were collected in duplicate from each participant , at the time of the first visit to the service. For this, they were obtained by rubbing a swab on the penile, oral and anal urethral mucosa, being stored in 5 mL of transport medium. The quantification of microorganisms was performed using the absolute quantification technique IL-1β, IL-6, IL-10 and TNF-α tumor necrosis factor assays were performed using an eBioscience ELISA kit. Results: Our sample consisted predominantly of black men who have sex with men or brown, aged between 18 and 19 years. Of these, 17.36% had one or more infections. There was an association between complete high school education and STI (p 0.045). Among the studied STIs, the prevalence of 85.3% stands out of mycoplasmas.In the evaluation of the profile of cytokines secreted between infected and non-infected, we found no statistically significant association. Matrix tests were performed among those infected with an association between higher TNF-α and IL-6 values. In the non-infected population, there is a general association between cytokines, that is, participants with lower levels showed a reduction in all cytokines studied. In this study, we had a prevalence of 4.73% of coinfections, all with at least one mycoplasma. 50% of the coinfections found were Mycoplasma hominis and Ureaplasma urealyticum. In the evaluation of the cytokine signature between mono- and co-infected individuals, we found higher levels of IL-1β in monoinfected individuals when compared to co-infected individuals and of TNF-α in monoinfected individuals, when compared to non-infected and co-infected individuals. In observing the heatmap with unsupervised clusters, two groups can be seen, one with a greater inflammatory potential, and the other with a lower potential, with emphasis on M. hominis and U. urealyticum with a high potential for an inflammatory response. The Principal Component Analysis shows the association between M. hominis infection and IL1-β secretion. Conclusion: Our study found no statistical difference between infected and uninfected young people, however, mycoplasma infection and co-infection with gonorrhea and chlamydia demonstrate an association with increased cytokines.

  • Objetivo: Avaliar os efeitos do treinamento físico terapêutico ou preventivo sobre parâmetros morfológicos e funcionais no tecido adiposo marrom em um modelo animal de envelhecimento e obesidade. Métodos: Foram utilizados 24 ratos Wistar machos (idade inicial = 4 meses e idade final = 14 meses) que foram aleatoriamente distribuídos em três grupos experimentais envelhecidos e obesos (n= 8/grupo): sedentário (EOS), exercício terapêutico (EOT) e exercício preventivo (EOP). Avaliou-se parâmetros de adiposidade corporal, e no tecido adiposo marrom (TAM) a atividade das enzimas antioxidantes (catalase, glutationa peroxidase-GPx e superóxido dismutase-SOD-1), TBARS e nitritos totais, parâmetros de morfologia e a expressão gênica de marcadores do perfil redox e função tecidual. Resultados: O exercício preventivo reduziu a massa de tecido adiposo subcutâneo abdominal dos animais envelhecidos e obesos. O peso do tecido adiposo abdominal visceral total (TAV) foi reduzido nos grupos exercitados EOT e EOP. No TAM, os protocolos de exercício, terapêutico e preventivo, determinaram o aumento do número de adipócitos por campo, redução do número de adipócitos uniloculares e aumento do número de adipócitos multiloculares. O percentual de vacúolos lipídicos por campo no TAM foi reduzido apenas no grupo EOP. Os níveis de TBARS e nitritos totais foram reduzidos pelos protocolos de exercícios terapêuticos e preventivo, sendo a redução maior no grupo EOP. A atividade da enzima antioxidante catalase, foi maior nos animais dos grupos EOT e EOP. A atividade da enzima GPx aumentou apenas nos animais treinados preventivamente apesar de a imunomarcação para GPx apresentar-se maior tanto no grupo EOT quanto EOP. A análise da expressão dos genes que codificam as enzimas Catalase, GPx-1 e SOD-1 no tecido adiposo marrom revelou que tanto o exercício terapêutico quanto o preventivo aumentaram a expressão gênica das enzimas antioxidantes, Catalase e GPx-1, sendo maior o efeito do treinamento preventivo. Não foram observadas diferenças entre os grupos na expressão gênica da enzima SOD-1. A expressão do gene que codifica a proteína UCP-1 no tecido adiposo marrom aumentou nos grupos EOP e EOT, enquanto que as expressões da PGC-1a e SIRT 1 aumentaram apenas no EOP. Conclusão: Os protocolos de exercícios terapêutico e preventivo determinaram melhora nos parâmetros de morfologia, função e estresse oxidativo no tecido adiposo marrom de ratos envelhecidos e com obesidade induzida por dieta. Os efeitos benéficos pelo protocolo preventivo foram mais evidentes. e sinalizam para o papel da regularidade da prática de exercícios para a atenuação ou reversão das alterações no tecido adiposo marrom no modelo animal estudado.

  • Objective: To evaluate the effects of therapeutic or preventive physical training on morphological and functional parameters in brown adipose tissue in an animal model of aging and obesity. Methods: Twenty-four male Wistar rats (initial age = 4 months and final age = 14 months) were randomly distributed into three elderly and obese experimental groups (n= 8/group): sedentary (EOS), therapeutic exercise (EOT) and preventive exercise (EOP). Body adiposity parameters were evaluated and, in brown adipose tissue (BAT), the activity of antioxidant enzymes, TBARS and total nitrites, morphology parameters, and gene expression of redox profile markers and tissue function. Results: Preventive exercise reduced the mass of abdominal subcutaneous adipose tissue in aged and obese animals. Total visceral abdominal adipose tissue (TAV) weight reduces in the EOT and EOP exercised groups. In the TAM, the exercise protocols, therapeutic and preventive, determined the increase in the number of adipocytes per field, reduction in the number of unilocular adipocytes, and increase in the number of multilocular adipocytes. The TAM's percentage of lipid vacuoles per field was reduced only in the EOP group. TBARS and total nitrite levels were reduced by therapeutic and preventive exercise protocols, with the reduction being more remarkable in the EOP group. The activity of the antioxidant enzyme catalase was higher in animals from the EOT and EOP groups. The activity of the GPx enzyme increased only in the preventively trained animals, although the immunostaining for GPx was higher in both the EOT and EOP groups. The analysis of the expression of the genes that encode the enzymes Catalase, GPx-1, and SOD-1 in brown adipose tissue revealed that both therapeutic and preventive exercise increased the gene expression of the antioxidant enzymes, Catalase and GPx-1, with a more significant effect of preventive training. No differences were observed between groups in SOD-1 enzyme gene expression. The gene encoding the UCP-1 protein expression in brown adipose tissue increased in the EOP and EOT groups, while the expressions of PGC-1a and SIRT 1 increased only in the EOP. Conclusion: The therapeutic and preventive exercise protocols determined improved morphology, function, and oxidative stress parameters in the brown adipose tissue of aged rats with diet-induced obesity. The preventive protocol's beneficial effects were more evident, indicating the role of regular exercise practice in attenuating or reversing alterations in brown adipose tissue in the studied animal model.

  • Introdução: A obesidade é caracterizada como uma condição crônica e complexa, que contribui para impactos negativos na saúde humana. Em sua gênese atuam fatores genéticos, ambientais, metabólicos e comportamentais que podem determinar redução na qualidade de vida, predispondo a alterações metabólicas e cardovasculares . Diante dessa problemática, o exercício físico é uma importante estratégia para redução da adiposidade corporal e prevenção de distúrbios cardiovasculares associados à obesidade. Objetivo: Avaliar o impacto do treinamento físico nas alterações do perfil oxidativo e nos parâmetros morfofuncionais cardiovasculares em modelo animal de obesidade induzida por dieta hipercalórica. Métodos: Foram utilizados 24 ratos Wistar machos com peso entre 150 e 200g, cujo protocolo deste estudo foi aprovado pelo Comitê de Ética no Uso de Animais (CEUA/IMS-UFBA) sob parecer número 053/2017. Inicialmente, os animais experimentais foram submetidos a dieta controle normocalórica (DC, n=10) ou dieta hipercalórica (DH, n=14) para indução da obesidade durante 8 semanas. Após esse período, os animais do grupo dieta controle (DC) ou do grupo dieta hipercalórica (DH) foram subdivididos em 2 grupos experimentais cada, os quais foram treinados em esteira motorizada (DCT, n=5; DHT, n=7) ou mantidos sedentários (DCS, n=5; DHS, n=7) por 12 semanas. Ao final do experimento, os depósitos de tecido adiposo abdominal foram dissecados e pesados e o ventrículo e a aorta removidos para análise histológica e de estresse oxidativo. Sangue torácico coletado para análises bioquímicas e hormonais. Os cortes histológicos foram corados com Hematoxilina-Eosina (HE) e Tricrômio de Masson (TM) e analisados pelo sistema informatizado (ImageJ). Resultados: O consumo da dieta hipercalórica determinou aumento do consumo alimentar e calórico, do peso e da adiposidade corporal, alterou a tolerância a glicose. O protocolo treinamento aplicado reduziu o peso corporal, índice de adiposidade e os tecido adiposo abdominais dos animais submetidos à dieta hipercalórica. Não foram observados alterações significativas sobre a glicemia de jejum, insulina, HOMA-IR, HOMA-b e QUICK. A pressão arterial sistólica aumentou nos animais sedentários que consumiram a dieta hipercalórica e foi reduzida pelo protocolo de treinamento físico aproximadamente em 12%. Os níveis de CK/MB foram reduzidos em animais treinados em comparação com animais sedentários submetidos a uma dieta hipercalórica. Os animais submetidos a dieta hipercalórica apresentaram maiores áreas na espessura da túnica íntima da aorta, consequentemente, esses animais tiveram menores lúmens aórticos; enquanto os animais treinados apresentaram menores espessuras e maiores lúmens. As áreas dos cardiomiócitos e a capilarização demonstraram adaptações cardíacas patológicas nos animais sedentários que consumiram dieta hipercalórica, enquanto nos animais treinados notou-se adaptações cardíacas fisiológicas com aumento de cardiomiócitos e de neocapilarização. Os animais sedentários submetidos a dieta hipercalórica apresentaram maiores áreas com fibrose enquanto protocolo de treinamento reduziu essas áreas. O treinamento aplicado neste estudo aumentou os valores de catalase e GPX; enquanto os valores de peroxidação lipídica (TBARS) foram reduzidos nos animais treinados. Os níveis de nitritos totais cardíaco foram maiores no grupo (DHS) e nos grupos treinados independente da dieta. Conclusão: O protocolo de treinamento determinou melhora na adiposidade corporal, e parâmetros morfofuncionais e oxidativos cardiovasculares no modelo animal de obesidade induzida por dieta.

  • Introduction: Obesity is characterized as a chronic and complex condition that contributes to negative impacts on human health. In its genesis, genetic, environmental, metabolic and behavioral factors act that can determine a reduction in quality of life, predisposing the population to metabolic and cardiovascular diseases. Faced with this problem, physical exercise is an important strategy for reducing body adiposity and preventing cardiovascular disorders associated with obesity. Objective: To evaluate the impact of physical exercise on changes in the oxidative profile and cardiovascular morphofunctional parameters in an animal model of obesity induced by a high-calorie diet. Methods: We used 24 male Wistar rats weighing between 150 and 200g, whose study protocol was approved by the Ethics Committee on the Use of Animals (CEUA/IMS-UFBA) under opinion number 053/2017. Initially, the experimental animals were subjected to a normocaloric control diet (DC, n=10) or a hypercaloric diet (DH, n=14) to induce obesity for 8 weeks. After this period, the animals in the control diet group (CD) or the hypercaloric diet group (HD) were subdivided into 2 experimental groups each, which were trained on a motorized treadmill (DCT, n=5; DHT, n=7) or maintained sedentary (DCS, n=5; DHS, n=7) for 12 weeks. At the end of the experiment, the abdominal adipose tissue deposits were dissected and weighed and the ventricle and aorta were removed for histological analysis. Chest blood was collected for biochemical and hormonal analysis. The histological sections were stained with Hematoxylin-Eosin (HE) and Masson's Tricromion (TM) and analyzed by the computerized system (ImageJ). Results: Consumption of a high-calorie diet resulted in an increase in food and caloric intake, body weight and adiposity, and altered glucose tolerance. The training protocol applied reduced the body weight, adiposity index, and abdominal adipose tissue of animals submitted to the high-calorie diet. No significant changes were observed in fasting blood glucose, insulin, HOMA-IR, HOMA-b, and Quick. Systolic blood pressure increased in sedentary animals that consumed the high-calorie diet and was reduced by the physical training protocol by approximately 12%. CK/MB levels were reduced in trained animals compared to sedentary animals subjected to a high-calorie diet. Animals submitted to a high-calorie diet had larger areas in the thickness of the aortic tunica intima, consequently, these animals had smaller aortic lumens; while trained animals had smaller thicknesses and larger lumens. The areas of cardiomyocytes and capillarization demonstrated pathological cardiac adaptations in sedentary animals that consumed a high-calorie diet, while in trained animals’ were noted increased cardiomyocyte area and new vascularization in both diets. Sedentary animals submitted to a high-calorie diet showed larger areas with fibrosis, while the training protocol reduced these areas. The training applied in this study increased catalase and GPX values; while lipid peroxidation values (TBARS) had lower rates in trained animals. Cardiac total nitrite levels were higher in the (DHS) group and in the trained groups regardless of diet. Conclusion: The training protocol determined an improvement in body adiposity, oxidative profile and cardiovascular morphofunctional parameters in the animal model of diet-induced obesity.

  • O Diabetes Mellitus tipo 1 (DMT1) é uma doença crônica não transmissível na qual a insulina é produzida insuficientemente devido a destruição das células β no pâncreas. As mudanças geradas por esta condição aumentam o risco de infecções por estes indivíduos. Staphylococcus aureus resistente a meticilina (MRSA) é um dos principais patógenos causadores de infecções em indivíduos com diabetes. Neste trabalho, tivemos como objetivo investigar os efeitos da aplicação da terapia fotodinâmica (PDT), utilizando a curcumina como fotossensibilizante (FS), no tratamento de infecção intradérmica em camundongos com DMT1. Camundongos C57bl/6, foram pesados e, em jejum, tiveram a glicemia mensurada. O DMT1 foi induzido pela injeção intraperitoneal de uma dose única de estreptozotocina (STZ) (180 mg/kg). Cinco dias após a administração da STZ a glicemia dos animais foi mensurada para confirmar o estabelecimento do DMT1. Quinze dias após a indução do DMT1, os animais foram pesados e então infectados por MRSA (ATCC 43300) pela via intradérmica na orelha esquerda. Para a infecção, foi quantificada a concentração de 1,5 x 107 UFC da cepa MRSA ATCC 4330. Vinte e quatro horas após a infecção, os tratamentos experimentais foram realizados. Uma solução contendo 100 μg de curcumina foi fotoativada ex vivo com o emprego de luz de LED (450 nm) obtendo uma fluência de 13.5 J/cm2 . Após a fotoativação, a solução foi inoculada no mesmo sítio onde foi realizada a infecção. Os tratamentos para controle consistiram na aplicação do FS não-fotoativado ou salina estéril. A eutanásia dos animais foi realizada 24 h após os tratamentos e a orelha esquerda e o linfonodo retromaxilar esquerdo foram cirurgicamente coletados. O linfonodo foi macerado em salina para análise da carga bacteriana, dosagem de citocinas por ELISA, dosagem de nitritos e nitratos e contagem de células. A orelha foi utilizada para a confecção de lâminas que foram coradas por H&E ou marcadas com anticorpos anti-mieloperoxidase (MPO) por imuno-histoquímica. As lâminas foram fotomicrografadas e contagens de células e análises de expressão de MPO no tecido foram realizadas. O grupo PDT apresentou menor carga bacteriana no linfonodo quando comparado aos controles (p-valor <0,05). Além da menor carga bacteriana, o grupo PDT também apresentou altos teores de nitratos e nitritos no linfonodo quando comparado aos controles (p-valor >< 0,001). O recrutamento celular para o sitio infeccioso apresentou-se similar entre os grupos, entretanto, a expressão da MPO no grupo PDT foi menos intensa comparada ao grupo salina (p-valor< 0,001). Quando analisada a produção de citocinas, o grupo PDT apresentou menores concentrações de IL-1, IL-12 e IL-10 (p-valor <0,01; p-valor ><0,05; p-valor >< 0,05, respectivamente), quando comparado ao grupo salina. Este estudo mostra, pela primeira, vez o potencial terapêutico da PDT usando curcumina no tratamento de uma infecção intradérmica causada por S. aureus em camundongos com DMT1.

  • Type 1 Diabetes Mellitus (T1DM) is a chronic non-communicable disease in which insulin is insufficiently produced due to the destruction of β cells in the pancreas. The changes generated by this condition increase the risk of infections for these individuals. Methicillin-resistant Staphylococcus aureus (MRSA) is one of the main pathogens causing infections in individuals with diabetes. In this work, we aimed to investigate the effects of the application of photodynamic therapy (PDT), using curcumin as a photosensitizer (PS), in the treatment of intradermal infection in mice with T1DM. C57bl/6 mice were weighed and, in a fasting state, their blood glucose was measured. DMT1 was induced by intraperitoneal injection of a single dose of streptozotocin (STZ) (180 mg/kg). Five days after STZ administration, the animals' blood glucose was measured to confirm the establishment of DMT1. Fifteen days after DMT1 induction, the animals were weighed and then infected intradermally in the left ear. For infection, the concentration of 1.5 x 107 CFU of the MRSA ATCC 4330 strain was quantified. Twentyfour hours after infection, experimental treatments were performed. A solution containing 100 μg of curcumin was photoactivated ex vivo using LED light (450 nm) obtaining a fluence of 13.5 J/cm2 . After photoactivation, the solution was inoculated at the same site where the infection was carried out. Control treatments consisted of the application of non-photoactivated PS or sterile saline. The animals were euthanized 24 h after the treatments and the left ear and left retromaxillary lymph node were surgically collected. The lymph node was macerated in saline for analysis of bacterial load, measurement of cytokines by ELISA, measurement of nitrites and nitrates, and cell counts. The ear was used to make slides that were stained by H&E or stained with anti-myeloperoxidase (MPO) antibodies by immunohistochemistry. Slides were photomicrographed and cell counts and tissue MPO expression analyzes were performed. The PDT group had a lower bacterial load in the lymph node when compared to controls (p-value <0.05). In addition to the lower bacterial load, the PDT group also had higher levels of nitrates and nitrites in the lymph node when compared to controls (p-value >< 0.001). Cell recruitment to the infectious site was similar between the groups, however, the expression of MPO in the PDT group was less intense compared to the saline group (p-value < 0.001). When analyzing the production of cytokines, the PDT group showed lower production of IL-1, IL-12, and IL-10 (p-value <0.01; p-value ><0.05; p-value >< 0.05, respectively), when compared to the saline group. This study shows, for the first time, the therapeutic potential of PDT using curcumin in the treatment of an intradermal infection caused by S. aureus in mice with T1DM.

  • Objetivo: Investigar os efeitos do treinamento físico aeróbico realizado terapeuticamente ou ao longo da vida sobre alterações histológicas, no balanço redox, inflamação, e metabolismo no músculo esquelético e  ventrículo cardíaco provocadas pela associação entre envelhecimento e obesidade in vivo e in vitro. 

    Métodos: O experimento foi dividido em estudo piloto e estudo principal. O estudo piloto buscou identificar a idade em que as alterações musculares e de performance física se iniciam em resposta ao envelhecimento natural. Dessa forma, 41 ratos Wistar machos com 4 meses de idade foram distribuídos aleatoriamente em 6 grupos definidos pela idade à eutanásia, sendo estes 4 (n = 5), 8 (n = 5), 12 (n = 7), 14 (n = 8), 18 (n = 8) e 20 meses (n = 8). Ao longo do protocolo experimental, os animais foram pesados semanalmente, e a performance física avaliada mensalmente através do teste de corrida máximo em esteira motorizada para ratos. Após a definição da idade na qual o declínio muscular se iniciou (14 meses), foi avaliada a influência do treinamento físico sobre as alterações observadas. Para tal, 32 ratos Wistar machos com 4 meses de idade foram distribuídos aleatoriamente em dois grupos: grupo sedentário (n = 16) e grupo treinado (n = 16). Ao completarem 8 meses de idade, 8 animais de cada grupo foram eutanasiados, formando os grupos jovens sedentário e treinado. Os demais animais seguiram até completarem 14 meses de idade, e constituíram os grupos envelhecidos sedentário e treinado. O treinamento foi realizado em esteira motorizada para ratos, a moderada intensidade, em dias alternados e duração de 1h por sessão. Os animais foram eutanasiados 24h após a última sessão de treinamento. Foram coletadas amostras de sangue, tecidos adiposos e músculos, que foram devidamente mensurados e armazenados em ultra freezer -80 °C, ou fixados. As amostras congeladas foram utilizadas para quantificação dos perfis inflamatório e oxidativo, e as amostras fixadas para estudos histológicos. O estudo principal constitui-se de um estudo in vivo e outro in vitro. O protocolo in vivo foi realizado com 32 ratos Wistar machos que foram distribuídos aleatoriamente em quatro grupos (n = 8/grupo): grupo envelhecido sedentário (ESed), envelhecido sedentário alimentado com dieta hiperlipídica (DHL) (ESed+DHL), envelhecido treinado terapeuticamente e alimentado com DHL (ETT+DHL) e envelhecido treinado ao longo da vida e alimentado com DHL (ETL+DHL). Após 24h da última sessão de treinamento físico, todos os animais foram eutanasiados e em seguida foram coletados o músculo gastrocnêmio e ventrículo cardíaco para estudos histológicos, imuno-histoquímicos, do balanço redox, inflamação e metabolismo. O músculo gastrocnêmio foi utilizado para análise da expressão de mRNA e microRNA por qPCR. Os depósitos de tecidos adiposos abdominais foram coletados e pesados pera determinação da composição corporal. Para os estudos in vitro, foram utilizados macrófagos primários obtidos do exsudato peritoneal de 6 camundongos BALB/c machos com 4 meses de idade, elicitados através da administração de tioglicolato (3% via IP), e mioblastos primários isolados do músculo gastrocnêmio de um rato Wistar macho com 4 meses de idade. As culturas de macrófagos e mioblastos foram expostas ao soro dos animais do estudo in vivo durante 24h. As culturas foram usadas para análise da viabilidade celular por MTT, dosagem de nitritos totais e peróxido de hidrogênio e análise da expressão de genes envolvidos na resposta inflamatória e metabolismo muscular. Todas as variáveis de interesse deste estudo foram submetidas ao teste de normalidade Shapiro-Wilk. As comparações entre grupos do teste piloto foram feitas com o teste ANOVA para medidas repetidas (análise temporal) e ANOVA one-way, seguida pelo pós-teste de Tukey. Para as análises entre grupos jovens e envelhecidos, sedentários e treinados, foi adotado delineamento fatorial (2x2) utilizando o teste ANOVA two-way. As análises dos estudos in vivo e in vitro foram feitas através dos testes T de Student e ANOVA one-way.

    Resultados: Os resultados do estudo piloto indicam que o declínio da função muscular se inicia aos 14 meses de idade, evidenciado pela redução da performance física, peso da massa magra e do músculo gastrocnêmio e capacidade antioxidante do músculo. Por outro lado, a rotina de treinamento físico até os 14 meses melhorou
    a composição corporal, perfil inflamatório, status redox e protegeu o músculo contra redução da área seccional das fibras. Os resultados do estudo in vivo evidenciaram um desfecho desfavorável em resposta à associação entre envelhecimento e obesidade induzida por dieta sobre os parâmetros musculares e cardíacos morfologia, fibrose, inflamação, estresse oxidativo e metabolismo. Além disso, foi observado no
    músculo gastrocnêmio o aumento da expressão do miRNA-21 e genes envolvidos na resposta pró-inflamatória e catabólica, e redução da expressão de genes envolvidos na resposta anti-inflamatória, antioxidante e anabólica. O treinamento físico, em especial o protocolo de treinamento ao longo da vida, atenuou os achados decorrentes da associação entre envelhecimento e obesidade sobre os parâmetros musculares e cardíacos, reduziu a expressão do miR-21 e aumentou a expressão do miR-486. Nos estudos in vitro, os macrófagos e mioblastos expostos ao soro dos animais envelhecidos e obesos apresentaram aumento do perfil pró-inflamatório e catabólico, respectivamente, ao passo que o soro dos animais treinados, especialmente aqueles que realizaram o treinamento ao longo da vida, determinou redução do perfil próinflamatório e catabólico das culturas celulares.

    Conclusão: No modelo animal estudado, a associação entre sedentarismo, envelhecimento e obesidade induzida por dieta hiperlipídica determinou prejuízos musculares especialmente nos perfis inflamatório, oxidativo e metabólico. Em contrapartida, a prática regular de exercício físico, especialmente ao longo de toda a vida, atenuou as alterações provocadas pela associação entre envelhecimento e obesidade em todos os parâmetros estudados, in vivo e in vitro. Nossos achados reforçam a superioridade do protocolo de treinamento ao longo da vida como ferramenta eficaz contra os efeitos da idade, mesmo quando associado à hábitos alimentares não saudáveis.

  • Objective: To investigate the effects of aerobic physical training performed therapeutically or throughout life on histological changes, redox balance, inflammation, and metabolism in skeletal muscle and cardiac ventricle caused by the association between aging and obesity in vivo and in vitro.

    Methods: The experiment was divided into a pilot study and a main study. The pilot study aimed to identify the age at which changes in muscle and physical performance begin in response to natural aging. Thus, 41 male Wistar rats aged 4 months were randomly distributed into 6 groups defined by age at euthanasia, these being 4 (n = 5), 8 (n = 5), 12 (n = 7), 14 (n = 8), 18 (n = 8) and 20 months (n = 8). Throughout the experimental protocol, the animals were weighed weekly, and physical performance was assessed monthly using the maximal running test on a motorized treadmill for rats. After defining the age at which muscle decline began (14 months), the influence of physical training on the changes observed was evaluated. To this end, 32 4-month-old male Wistar rats were randomly distributed into two groups: sedentary group (n = 16) and trained group (n = 16). Upon reaching 8 months of age, 8 animals from each group were euthanized, forming the young sedentary and trained groups. The remaining animals continued until they reached 14 months of age, and constituted the sedentary and trained aged groups. Training was carried out on a motorized treadmill for rats, at moderate intensity, on alternate days and lasting 1 hour per session. The animals were euthanized 24 hours after the last training session. Blood, adipose tissue and muscle samples were collected, duly measured and stored in an ultra-freezer -80 °C, or fixed. The frozen samples were used to quantify the inflammatory and oxidative profiles, and the samples were fixed for histological studies. The main study consisted of an in vivo and an in vitro study. The in vivo protocol was carried out with 32 male Wistar rats that were randomly distributed into four groups (n = 8/group): aged sedentary group (ESed), aging sedentary group fed a high-fat diet (HFD) (ASed+HFD), aging trained therapeutically and fed with HFD (ATT+HFD) and aging lifelong trained and fed with HFD (ALT+DHL). 24 hours after the last physical training session, all animals were euthanized and then the gastrocnemius muscle and cardiac ventricle were collected for histological, immunohistochemical, redox balance, inflammation and metabolism studies. The gastrocnemius muscle was used to analyze mRNA and microRNA expression by qPCR. Abdominal adipose tissue deposits were collected and weighed to determine body composition. For in vitro studies, primary macrophages obtained from the peritoneal exudate of 6 4-month-old male BALB/c mice, elicited through the administration of thioglycolate (3%), and primary myoblasts isolated from the gastrocnemius muscle of a 4 month old male rat were used. Cultures of macrophages and myoblasts were exposed to serum from the animals from the in vivo study for 24h. The cultures were used to analyze cell viability by MTT, total nitrites and hydrogen peroxide and to analyze the expression of genes involved in the inflammatory response and muscle metabolism. All variables of interest in this study were subjected to the Shapiro-Wilk normality test. Comparisons between pilot test groups were made using the ANOVA test for repeated measures (temporal analysis) and one-way ANOVA, followed by the Tukey post-test. For analyzes between young and aged, sedentary and trained groups, a factorial design (2x2) was adopted using the two-way ANOVA test. Analyzes of in vivo and in vitro studies were performed using Student's T test and one-way ANOVA test.

    Results: The results of the pilot study indicate that the decline in muscle function begins at 14 months of age, evidenced by the reduction in physical performance, lean mass and gastrocnemius muscle weight and antioxidant capacity of the muscle. On the other hand, the physical training routine up to 14 months improved body composition, inflammatory profile, redox status and protected the muscle against reduction in the cross-sectional area of the fibers. The in vivo study showed an unfavorable outcome in response to the association between aging and diet-induced obesity on muscle and cardiac parameters, morphology, fibrosis, inflammation, oxidative stress and metabolism. Furthermore, an increase in the expression of miRNA-21 and genes involved in the pro-inflammatory and catabolic response, and a reduction in the expression of genes involved in the antioxidant and anabolic response were observed in the gastrocnemius muscle. Physical training, especially the lifelong training protocol, attenuated the findings resulting from the association between aging and obesity on muscular and cardiac parameters, reduced the expression of miR-21 and increased the expression of miR-486. In in vitro studies, macrophages and myoblasts exposed to serum from aging and obese animals showed an increase in the pro-inflammatory and catabolic profile, respectively, while serum from trained animals, especially those that had undergone lifelong training, determined reduction of the pro-inflammatory and catabolic profile of cell cultures.

    Conclusion: In the animal model studied, the association between sedentary lifestyle, aging and obesity induced by a high-fat diet determined muscle damage, especially in the inflammatory, oxidative and metabolic profiles, and these changes appear to be associated with the positive modulation of miR-21. On the other hand, regular physical exercise, especially throughout life, attenuated the changes caused by the association between aging and obesity in all parameters studied, in vivo and in vitro. Our findings reinforce the importance of adopting exercise routines throughout life as an effective tool against the effects of age, especially when associated with unhealthy eating habits.

  • A nefropatia diabética está frequentemente associada ao aumento do estresse oxidativo pela geração de espécies reativas de oxigênio (ERO), as quais podem provocar uma sequência de respostas pró-inflamatórias, pró-apotóticas e disfunção endotelial nos rins. Novas estratégias terapêuticas são necessárias para atenuar e/ou prevenir a progressão da nefropatia diabética. Diante disso, este estudo avaliou o impacto de dois protocolos de exercício nas alterações oxidativas, inflamatórias e apoptóticas induzidas pelo diabetes mellitus tipo 1 (DM1) no tecido renal de ratas. Para tal, 30 ratas Wistar foram utilizadas e divididas em 5 grupos: CS - ratas controle sedentárias (n=6), CT – ratas controle treinadas (n=6), DS – ratas diabéticas sedentárias (n=6), DT – ratas diabéticas treinadas (n=6) e DPT – ratas diabéticas previamente treinadas (n=6). Apenas o grupo DPT foi submetido a quatro semanas de exercício antes da indução do diabetes. Em seguida, o DM1 foi induzido nos grupos DS, DT e DPT com estreptozotocina (40mg/kg, iv), e os grupos DPT e DT foram submetidos a 8 semanas de exercício. O grupo CT também foi submetido a 8 semanas de exercício após administração de citrato (0,1 M, iv). O tecido renal foi removido e destinado aos estudos de estresse oxidativo, imuno-histoquímica, ELISA e Western blot. Os nossos dados mostram que o exercício físico reduziu os níveis de TBARS e aumentou a atividade da superóxido dismutase no tecido renal de ratas submetidas ao treinamento físico. Contudo, somente o exercício prévio aumentou a atividade da enzima glutationa peroxidase. Além disso, o exercício reduziu a caspase-3, o p-p38 e a expressão de macrófagos M1 e aumentou a expressão de macrófagos M2 e Bcl-2, bem como os níveis IL-10 no tecido renal. Somente o exercício iniciado antes da indução do diabetes reduziu os níveis de TNF-alfa no grupo DPT. Em conclusão, nossos dados demonstram que o exercício físico de intensidade moderada melhorou o status redox, a inflamação e apoptose. Contudo, o exercício iniciado previamente a indução do diabetes promoveu efeitos adicionais por aumentar a atividade da glutationa peroxidase e reduzir os níveis de TNF-alfa no tecido renal das ratas Wistar

  • Diabetic nephropathy is often associated with increased oxidative stress by the generation of reactive oxygen species (ROS), which can provoke a sequence of pro-inflammatory, pro-apototic responses and endothelial dysfunction in the kidneys. New therapeutic strategies are needed to attenuate and/or prevent the progression of diabetic nephropathy. Therefore, this study evaluated the impact of two exercise protocols on oxidative, inflammatory and apoptotic changes induced by type 1 diabetes mellitus (DM1) in the renal tissue of female rats. For this, 30 female Wistar rats were used and divided into 5 groups: SC - sedentary control rats (n=6), TC - trained control rats (n=6), SD - sedentary diabetic rats (n=6), TD - rats trained diabetic rats (n=6) and PTD – previously trained diabetic rats (n=6). Only the PTD group was submitted to four weeks of exercise before the induction of diabetes. Then, DM1 was induced in the SD, TD and PTD groups with streptozotocin (40mg/kg, iv), and the PTD and TD groups underwent 8 weeks of exercise. The TC group also was submitted 8 weeks of exercise after administration of citrate (0.1 M, iv). Kidney tissue was removed for oxidative stress, immunohistochemistry, ELISA and Western blot studies. Our data show that exercise training reduced TBARS levels and increased superoxide dismutase activity in the renal tissue of rats of the TD and PTD groups. However, only previous exercise increased the activity of the enzyme glutathione peroxidase in the PTD group. Furthermore, exercise reduced the expression of caspase-3, p-p38 and M1 macrophages and increased M2 macrophages and Bcl-2, as well as IL-10 levels in kidney tissue of the TD and PTD groups. Only exercise initiated before diabetes induction reduced TNF-alpha levels in the DPT group. In conclusion, our data demonstrate that moderate intensity aerobic exercise improved redox status, inflammation and apoptosis. However, exercise initiated prior to the induction of diabetes promoted additional effects by increasing glutathione peroxidase activity and reducing TNF-alpha levels in the kidney tissue of female Wistar rats.

  • INTRODUÇÃO/OBJETIVO: O envelhecimento em mulheres está associado as alterações decorrentes do climatério, que culmina em modificações na composição corporal, alterações metabólicas e hormonais, que apresentam maior susceptibilidade ao desenvolvimento de doenças crônicas não transmissíveis (DCNT). Dentre os agravos crônicos de maior incidência nesta população, o diabetes mellitus (DM) e a hipertensão arterial sistêmica (HAS) se destacam. Essas doenças, apesar dos diferentes órgãos e sistemas envolvidos, têm um fator de risco comum: em geral, inflamação clinicamente silenciosa de baixo grau que contribui para a patogênese e progressão de doenças relacionadas à idade. Portanto, este trabalho teve como finalidade o estudo ampliado sobre as interações dos parâmetros antropométricos, bioquímicos e inflamatórios associados ao risco na gravidade dessas doenças em mulheres idosas na pós-menopausa. MÉTODOS: O estudo envolveu a avaliação de 126 mulheres idosas, tendo diagnóstico positivo de hipertensão arterial e diabetes mellitus. Os grupos foram divididos em riscos proposto pela Sociedade Europeia de Cardiologia. Dados antropométricos foram coletados com a análise de bioimpedância, enquanto os dados inflamatórios e bioquímicos foram realizados a partir da coleta sanguínea. Os soros obtidos foram avaliados por ensaios enzimáticos. As análises estatísticas foram realizadas através do GraphPad Prism, e as correlações foram analisadas através do software R Studio. RESULTADOS: A circunferência da cintura e a razão circunferência da cintura/altura foram maiores no grupo risco muito grave quando comparado ao grupo de risco leve. A massa magra (p< 0,011) diminui com o aumento do risco de gravidade. As pressões sistólica (p< 0,0002) e diastólica (p< 0,0054) aumentaram em paralelo com o aumento da gravidade em relação aos grupos. Os marcadores bioquímicos aumentaram com o risco de gravidade: glicose, 7 triglicerídeos, colesterol, LDLc e VLDLc, exceto o HDLc. Com relação aos dados inflamatórios foi possível observar que as citocinas inflamatórias IL-1β, IFN-γ, TNF-α aumentaram em conjunto com a gravidade. Entretanto, a citocina antiinflamatória IL-10 (p=0,0293) diminuiu no grupo de risco muito grave. As razões entre as citocinas mostraram padrões diferentes entre os grupos. O grupo de idosas de risco muito grave apresentou menores valores para as razões IL-10/IL1β (p< 0,05), IL-10/IL-17 (p< 0,05) e IL-10/TNF-ɑ (p< 0,01) quando comparado com as idosas de risco grave. Os padrões de correlações paras as variáveis bioquímicas, inflamatórias e de composição corporal são distintas entre os grupos. Entretanto, o grupo de risco muito grave, observa menores forças de correlação de alguns parâmetros e o surgimento de outras interações. CONCLUSÃO: Os resultados mostraram ser extensamente alterados no grupo de risco muito grave, onde o perfil inflamatório perde sua responsividade à medida que aumenta a gravidade. Este é o primeiro trabalho que mostra uma visão ampliada da complexa interação entre os diferentes parâmetros avaliados em mulheres idosas hipertensas e diabéticas.

  • INTRODUCTION/OBJECTIVE: Aging in women is associated with changes resulting from the climacteric, which culminates in changes in body composition, metabolic and hormonal changes, which are more susceptible to the development of chronic non-communicable diseases (NCDs). Among the chronic diseases with the highest incidence in this population, diabetes mellitus (DM) and systemic arterial hypertension (SAH) stand out. These diseases, despite the different organs and systems involved, have a common risk factor: in general, clinically silent low-grade inflammation that contributes to the pathogenesis and progression of age-related diseases. Therefore, this work aimed to carry out an expanded study on the interactions of anthropometric, biochemical and inflammatory parameters associated with the risk of the severity of these diseases in elderly postmenopausal women. METHODS: The study involved the evaluation of 126 elderly women with a positive diagnosis of arterial hypertension and diabetes mellitus. The groups were divided into risks proposed by the European Society of Cardiology. Anthropometric data were collected with bioimpedance analysis, while inflammatory and biochemical data were obtained from blood collection. The sera obtained were evaluated by enzymatic assays. Statistical analyzes were performed using GraphPad Prism, and correlations were analyzed using R Studio software. RESULTS: Waist circumference and waist circumference/height ratio were higher in the very severe risk group when compared to the mild risk group. Lean body mass (p< 0.011) decreases with increasing risk of severity. Systolic (p< 0.0002) and diastolic (p< 0.0054) pressures increased in parallel with the increase in severity in relation to groups. Biochemical markers increased with the risk of severity: glucose, triglycerides, cholesterol, LDLc and VLDLc, except HDLc. Regarding the inflammatory data, it was possible to observe that the inflammatory cytokines IL-1β, IFN-γ, TNF-α increased together with the severity. However, the anti-inflammatory cytokine IL10 (p=0.0293) decreased in the very severe risk group. Cytokine ratios showed different patterns between groups. The group of elderly women at very severe 9 risk had lower values for the ratios IL-10/IL-1β (p< 0.05), IL-10/IL-17 (p< 0.05) and IL-10/TNF- ɑ (p< 0.01) when compared with elderly women at severe risk. The patterns of correlations for biochemical, inflammatory and body composition variables are different between groups. However, the very severe risk group observes lower correlation forces for some parameters and the emergence of other interactions. CONCLUSION: The results showed to be extensively altered in the very severe risk group, where the inflammatory profile loses its responsiveness as the severity increases. This is the first work that shows an expanded view of the complex interaction between the different parameters evaluated in elderly hypertensive and diabetic women.

  • A Serotonina (5-hidroxitriptamina ou 5-HT) modula respostas imunológicas em células imunes, sendo que essa ação ocorre por meio do sistema serotoninérgico. Este neurotransmissor monoaminérgico atua sobre as células imunes, coordenando as respostas imunológicas de macrófagos, células Natural killer (NK), células dendríticas e linfócitos, seja inibindo-as ou estimulando-as. Esse papel imunomodulador ocorre principalmente em células com ação pró-inflamatória, porém não são conhecidos os efeitos da 5-HT especificamente sobre diferentes estímulos imunes, em tecidos específicos. Com isso, o presente estudo teve como objetivo avaliar o efeito da 5-HT, na resposta imunológica induzida por inoculação de Staphylococcus aureus e Escherichia coli, em cultura de macrófagos peritoneais e esplênicos de camundongos tratados ou não com a serotonina. Para isso, foram utilizados camundongos da linhagem C57BL6. O primeiro grupo foi o de animais tratados com 5-HT in vivo, onde os camundongos passaram por uma cirurgia intracerebroventricular (ICV). O segundo grupo de animais sham, realizou a cirurgia ICV, no entanto, foi administrado apenas ácido ascórbico 0,1% (veículo). Após a cirurgia, os macrófagos peritoneais e esplênicos foram extraídos dos animais e seguiram para incubação durante 24 horas. O grupo sham foi dividido em dois subgrupos: macrófagos tratados com 5-HT in vitro e os macrófagos não tratados. Os macrófagos tratados in vitro receberam diferentes dosagens de 5-HT (10-3 M, 10-6 Me 10-8 M) e foram novamente incubados por 24 horas, enquanto o subgrupo de macrófagos não tratados não passou por nenhum tipo de contado com 5-HT. Após o período de respectiva incubação, os macrófagos tratados in vivo, in vitro e os não tratados foram infectados com E. coli e S. aureus, LPS (controle positivo) ou salina (controle negativo) e incubados novamente por 3 horas. Após a coleta, foi realizado a expressão gênica das citocinas TNF-α, IL-1β, IL-6 nas células, bem como a dosagem de espécies reativas de oxigênio e de óxido nítrico do sobrenadante. De maneira geral, macrófagos obtidos de animais tratado in vivo com 5-HT e macrófagos tratados in vitro com 10-3 M e 10-6 M de 5-HT apresentaram menores expressões de marcadores inflamatórios quando comparado com macrófagos sem nenhum tipo de tratamento, principalmente para as infecções utilizando E. coli e S. aureus. O mesmo efeito não foi observado com o tratamento in vitro utilizando 10-8 M de 5-HT. Portanto, os resultados do presente estudo nos levam a conclusão que esse hormônio pode estar envolvido na inibição de componentes que regulam a inflamação, de maneira dose dependente e de acordo ao tipo de infecção ao qual este estiver exposto. Além disso, a serotonina também pode ser utilizada futuramente como um agente terapêutico no tratamento de doenças inflamatórias. No entanto, mais estudos necessitam ser realizados para avaliar aprofundadamente os mecanismos envolvidos nesse processo.

  • Serotonin (5-hydroxytryptamine or 5-HT) modulates immune responses in immune cells, and this action occurs through the serotonin system. This monoaminergic neurotransmitter acts on immune cells, coordinating the immune responses of macrophages, Natural killer (NK) cells, dendritic cells, and lymphocytes, either inhibiting or stimulating them. This immunomodulatory role occurs mainly in cells with pro-inflammatory action, especially the effects of 5-HT on different immunological stimuli in specific tissues that are not known. Thus, the present study aimed to evaluate the effect of 5-HT on the immune response induced by inoculation of Staphylococcus aureus and Escherichia coli in the culture of peritoneal and splenic macrophages from mice treated or not with serotonin. For this, mice of the C57BL6 strain were used. The first animal was treated with 5-HT in vivo, where the ventricular group mice were operated on by intracerebral surgery (ICV). The second group of sham animals experienced an ICV surgery; however, only 0.1% ascorbic acid (vehicle) was administered. After surgery, peritoneal and splenic macrophages were extracted from the animals and incubated for 24 hours. The sham group was divided into two subgroups: macrophages treated with 5-HT in vitro and untreated macrophages. The macrophages treated in vitro received different doses of 5-HT (10-3 M and 10-8 M) and were again incubated for 24 hours, while the subgroup of untreated macrophages did not undergo any contact with 5-HT. After the incubation period, respectively, negative macrophages treated in vivo, in vitro, and untreated were infected with E. coli and S. aureus, LPS (positive control), or saline (control) and incubated again for 3 hours. After collection, the gene expression of the cytokines TNF-α, IL1β, and IL-6 was performed in the cells, as well as the reactive oxygen and nitric oxide species in the supernatant. In general, macrophages from animals treated in vivo with 5-HT and macrophages treated in vitro with 10-3 M and 10-6 M 5-HT showed lower inflamed marker faces when compared to macrophages without any treatment, mainly for infections using E. coli and S. aureus. The same effect was not observed with in vitro treatment using 10-8 M 5-HT. In addition, serotonin may also be used in the future as a therapeutic agent in the treatment of inflammatory diseases. However, studies are carried out for more studies carried out in this process.

  • Objetivo: Avaliar os efeitos das dietas hiperlipídicas com óleos de coco ou de linhaça restritas em carboidratos ou do exercício de moderada intensidade sobre parâmetros metabólicos e hepáticos em ratos obesos. Métodos: Foram utilizados 54 ratos machos Wistar que inicialmente receberam as dietas controle (n=6) ou hipercalórica (n=48) para indução da obesidade por 8 semanas. Após este período, os animais do grupo controle (n=6) e parte do grupo hipercalórica (n=6) foram eutanasiados e os demais foram subdivididos em seis grupos, com diferentes dietas e submetidos ou não ao exercício físico de moderada intensidade por 12 semanas: hipercalórica e sedentários (n=7); hipercalórica e exercitados (n=7); hiperlipídica com óleo de coco restrita em
    carboidratos e sedentários (n=7); hiperlipídica com óleo de coco restrita em carboidratos e exercitados (n=7), hiperlipídica com óleo de linhaça restrita em carboidratos e sedentários (n=7) e hiperlipídica com óleo de linhaça restrita em carboidratos e exercitados (n=7). Avaliou-se parâmetros dietéticos, cardiovasculares e de composição corporal, o perfil lipídico e inflamatório plasmático, a homeostase glicêmica, a resistência e sensibilidade à insulina. Além disso, avaliou-se o desenvolvimento de esteatose, inflamação, fibrose e
    estresse oxidativo no fígado. Resultados: O modelo experimental de obesidade induzida por dieta foi validado. A dieta hiperlipídica com óleo de coco restrita em carboidratos reduziu o consumo alimentar e calórico, mas
    aumentou a adiposidade corporal, os níveis pressóricos, glicêmicos e insulínicos, reduziu os níveis de IL-10, aumentou o colesterol no fígado, promoveu deposição macrovesicular de gordura inflamação, fibrose e estresse
    oxidativo no tecido hepático. A dieta hiperlipídica com óleo de linhaça restrita em carboidratos, embora tenha aumentado a ingestão calórica, não aumentou a massa e a adiposidade corporal, melhorou o perfil lipídico, reduziu os níveis de IL6 e IL-10, o estresse oxidativo e a deposição de macro e microvesículas
    de gordura no fígado, no entanto, promoveu inflamação hepática e hiperglicemia. O exercício promoveu efeitos benéficos sobre a maioria dos parâmetros metabólicos e hepáticos. Conclusão: O consumo da dieta hiperlipídica com óleo de coco restrita em carboidratos impactou negativamente a adiposidade corporal e os perfis metabólicos e inflamatórios sistêmico e hepático, enquanto que o consumo da dieta hiperlipídica com óleo de linhaça restrita em carboidratos e o exercício promoveram efeitos benéficos sobre a maioria dos parâmetros metabólicos e hepáticos analisados. Desta forma, sugere-se cautela na utilização destas dietas, mesmo quando associadas ao exercício, reforçando a necessidade da realização de mais estudos para determinar as recomendações seguras de inclusão desses óleos na dieta.

  • Objective: To evaluate the effects of high-fat diets with coconut or linseed oil carbohydrate-restricted and moderate-intensity exercise on metabolic and hepatic parameters in obese rats. Methods: Fifty-four male Wistar rats were used and initially received a control (n=6) or hypercaloric (n=48) diet for 8 weeks to induce obesity. After this period, the animals of the control group (n=6) and part of the hypercaloric group (n=6) were euthanized and the others were subdivided into six groups, with different diets and submitted or not to moderate-intensity physical exercise for 12 weeks: hypercaloric and sedentary (n=7); hypercaloric and exercised (n=7); high-fat diet with coconut oil carbohydrate-restricted and sedentary (n=7); high-fat diet with coconut oil carbohydrate-restricted and exercised (n=7), high-fat diet with linseed oil
    carbohydrate-restricted and sedentary (n=7) and high-fat diet with linseed oil carbohydrate-restricted and exercised (n=7). Dietary, cardiovascular, and body composition parameters were evaluated, as well as lipid and plasma inflammatory profile, glycemic homeostasis, insulin resistance and sensitivity. In addition, the development of steatosis, inflammation, fibrosis, and oxidative stress in liver were evaluated. Results: The experimental model of diet-induced obesity was validated. The high-fat diet with coconut oil carbohydrate-restricted reduced food and caloric intake, but increased body adiposity, blood pressure, glucose and insulin levels, reduced IL-10 levels, increased liver cholesterol, promoted macrovesicular fat deposition, inflammation, fibrosis and oxidative stress in liver tissue. The high-fat diet with linseed oil carbohydrate-restricted, although it increased caloric intake, did not increase body mass and adiposity, improved lipid profile, reduced IL6 and IL-10 levels, oxidative stress and the deposition of macro and microvesicles of fat in the liver, however, promoted hepatic inflammation and hyperglycemia. Exercise enabled beneficial effects on most of the metabolic and hepatic parameters analyzed. Conclusion: Consumption of the high-fat diet with coconut oil carbohydrate-restricted
    negatively impacted body adiposity and systemic and hepatic metabolic and inflammatory profiles, while consumption of the high-fat diet with linseed oil carbohydrate-restricted and exercise had beneficial effects on most of the metabolic and hepatic parameters analyzed. Thus, caution is suggested in the use of these diets, even when associated with exercise, reinforcing the need for further studies to determine safety recommendations for including these oils in the diet.

  • Introdução: Inflamação é uma resposta biológica complexa que envolve os sistemas imune, autonômico, vascular e somatossensorial, que cursa com a síntese de mediadores inflamatórios e indução de dor por ativação de nociceptores. Uma das causas mais comuns e impactantes de inflamação na saúde pública é a infecção por Staphylococcus aureus, principal causa de bacteremia, com piores desfechos clínicos hospitalares. Substâncias antioxidantes têm sido avaliadas quanto nesse contexto como alternativa terapêutica analgésica, antioxidante, anti-inflamatória, antitumoral e bactericida. Dentre elas, destacam-se os óleos essenciais de plantas aromáticas, como o β-cariofileno (BCP), e os ácidos graxos poli-insaturados, como o Ácido Docosahexanóico (DHA). Objetivo: Avaliar as atividades biológicas da associação BCP-DHA em modelos experimentais in vitro e in vivo de antinocicepção e inflamação. Materiais e métodos: Para tanto, os efeitos antinociceptivos foram avaliados em modelos animais através dos testes de contorção abdominal induzida por ácido acético, do edema de pata induzido por injeção intraplantar de formalina e da hipernocicepção de VonFrey. Para determinar os efeitos anti-inflamatórios, monócitos isolados do sangue periférico de homens adultos voluntários foram infectados com Staphylococcus aureus resistente à meticilina e incubados com tratamento, para posterior dosagem de citocinas e análise da expressão gênica. A capacidade antioxidante foi avaliada pelo método DPPH. Foi realizado o ensaio MTT para determinar a viabilidade celular das linhagens pulmonares A549 e BEAS frente ao tratamento com BCP-DHA e, em seguida, foram feitos os testes de adesão e migração com as doses das associações determinadas no teste anterior. Resultados: Efeitos antinociceptivos foram observados nos três modelos realizados ao comparar os grupos controle (salina) e tratados com BCP-DHA, obtendo significância estatística (p < 0,05) no teste de contorções abdominais induzidas por ácido acético, na avaliação dos flintches e lambidas de pata no modelo de injeção intraplantar de formalina e na hipernocicepção do teste de Von Frey. Houve redução significativa dos níveis de GM-CSF, TNFα, IL-1, IL-6 e IL-12, bem como aumento de IL-10 nos grupos submetidos ao tratamento com a associação BCP-DHA, assim como regulação negativa da expressão dos genes envolvidos na cascata de sinalização inflamatória intracelular (IL-2, IL-6, IRF7, NLRP3 e TYK2) em todos os grupos que receberam o tratamento, independente da presença de infecção. No teste do DPPH apenas o BCP apresentou sequestro significativo de radicais livre, com EC 50 = 2,82 ± 0,06 M. Houve atividade citotóxica significativa das 2 combinações de BCP-DHA testadas sobre linhagem tumoral A549, bem como importante inibição dos processos de adesão e migração celular. Observou-se subexpressão de genes envolvidos no ciclo celular, no reparo do DNA, da excisão de nucleotídeos, de bases mal pareadas e pós replicação, na manutenção de telômeros, oncogenes e supressores tumorais. Diante disso, nossos resultados sugerem fortemente que a associação de BCP-DHA surge como uma possível nova alternativa terapêutica com efeitos analgésico, anti-inflamatório, antiangiogênico, pró-apoptótico e antiproliferativo.

  • Introduction: Inflammation is a complex biological response involving the immune, autonomic, vascular and somatosensory systems, which involves the synthesis of inflammatory mediators and pain induction by activating nociceptors. One of the most common and impacting causes of inflammation in public health is infection by Staphylococcus aureus, the main cause of bacteremia, with worse in-hospital clinical outcomes. Antioxidant substances have been evaluated in this context as an analgesic, antioxidant, anti-inflammatory, antitumor, and bactericidal therapeutic alternatives. Among them, the essential oils of aromatic plants, such as β-caryophyllene (BCP), and polyunsaturated fatty acids, such as Docosahexaenoic acid (DHA) stand out. Objective: To evaluate the biological activities of the BCP-DHA association in vitro and in vivo experimental models of antinociception and inflammation. Materials and methods: For that, the antinociceptive effects were evaluated in animal models through tests of abdominal writhing induced by acetic acid, paw edema induced by intraplantar injection of formalin, and VonFrey hypernociception. To determine the anti-inflammatory effects, monocytes isolated from peripheral blood of adult male volunteers were infected with methicillin-resistant Staphylococcus aureus and incubated with treatment for subsequent cytokine measurement and gene expression analysis. The antioxidant capacity was evaluated by the DPPH method. The MTT assay was performed to determine the cell viability of the A549 and BEAS lung lines against treatment with BCP-DHA and then, adhesion and migration tests were performed with the doses of the associations determined in the previous test. Results: Antinociceptive effects were observed in the three models performed when comparing the control (saline) and BCP-DHA treated groups, obtaining statistical significance (p < 0.05) in the test of abdominal writhing induced by acetic acid, in the evaluation of flinches and Paw licking in the intraplantar formalin injection model and in Von Frey test hypernociception. There was a significant reduction in the levels of GM-CSF, TNFα, IL-1, IL-6, and IL-12, as well as an increase in IL-10 in the groups submitted to treatment with the BCP-DHA association, as well as negative regulation of the expression of genes involved in the intracellular inflammatory signaling cascade (IL-2, IL-6, IRF7, NLRP3, and TYK2) in all groups that received treatment, regardless of the presence of infection. In the DPPH test, only BCP showed significant scavenging of free radicals, with EC 50 = 2.82 ± 0.06 M. There was the significant cytotoxic activity of the 2 combinations of BCP-DHA tested on the A549 tumor cell line, as well as important inhibition of cell adhesion and migration. There was underexpression of genes involved in the cell cycle, DNA repair, nucleotide excision, mismatched bases and post replication, and maintenance of telomeres, oncogenes, and tumor suppressors. Therefore, our results strongly suggest that the association of BCP-DHA emerges as a possible new therapeutic alternative with analgesic, anti-inflammatory, antiangiogenic, pro-apoptotic, and antiproliferative effects.

  • Introdução: As infecções por Staphylococcus aureus apresentam uma taxa de morbidade e mortalidade elevada, considerado um grande problema para a saúde pública. Existem poucas informações na literatura sobre o papel da dihidrotestosterona nas infecções causadas por S. aureus. O objetivo: Avaliar a influência da 5α-dihidrotestosterona (DHT) sobre a resposta de macrófagos peritoneais murinos (MPMs) e monócitos humanos do sangue periférico (hPMs) na resposta imunológica induzida por Staphylococcus aureus. Metodologia: Foi realizado um modelo in vitro oriundo de MPMs machos BALB/C Shams, orquiectomizados (OQX) e fêmeas e de hPBMs de homens e mulheres jovens em idade fértil, saudáveis, com níveis de esteroides sexuais dentro da faixa de normalidade. As células foram inoculadas com 10μl de S. aureus capa 80 ou com salina estéril (controle) por um período de 6h. As células dos machos OQX, fêmeas e dos hPBMs homens e mulheres também foram pré-tratadas com a concentração de 100μl (10-2M) de DHT por 24h antes da estimulação com S. aureus. As citocinas TNF-α, IL-1α, IL-1β, IL-6, IL-8, IL-10, IL-12, GM-CSF, os nitritos totais, peróxido de hidrogênio (H2O2)  foram mensurados em sobrenadante de cultura em MPMs e em hPBMs. Além disso, foi realizada a análise de 84 genes, por RT-PCR array, envolvidos na resposta imune contra S. aureus. Resultados: Em MPMs, as células inoculadas com S. aureus dos machos Sham apresentaram níveis mais elevados das citocinas inflamatórias e menores concentrações de IL-10, nitritos totais e H2O2 em comparação aos OQX. As células dos machos Sham inoculadas com S. aureus também apresentaram maiores concentrações de citocinas pró-inflamatórias e menores concentrações de IL-10, nitritos totais e H2O2 em relação as fêmeas. No tratamento com o DHT, a concentração de citocinas inflamatórias e a expressão de genes como TLR-2 e genes envolvidos pela via do NF-kB foram maiores nos OQX tratados com o hormônio em comparação aos OQX sem o pré-tratamento. Além disso, as concentrações de nitritos totais e H2O2 foram menores em células com o pré-tratamento, tanto em células dos machos OQX, quanto em fêmeas. Para o experimento com seres humanos, todos os sujeitos da pesquisa selecionados estavam saudáveis e os níveis dos hormônios sexuais estavam dentro dos padrões de normalidade para homens e mulheres em idade fértil (17β-estradiol, progesterona, hormônios folículo estimulante (FSH), luteinizante (LH) e testosterona). Os hPBMs inoculados com S. aureus de homens apresentaram maiores concentrações de citocinas inflamatórias e menores concentrações de IL-10, menores níveis de nitritos totais e H2O2 em comparação as mulheres. Em resposta ao tratamento, com o DHT, o hormônio inibiu as concentrações de nitritos totais e H2O2 tanto em homens quanto em mulheres e apresentaram maior expressão de TLR-2 e genes envolvidos pela via do NF-kB, corroborando com os resultados em murinos. Conclusão: Nossos resultados demonstraram que existe diferença entre os sexos na resposta a inoculação por S. aureus e que o DHT apresenta propriedades imunomoduladoras pró-inflamatórias em MPMs e em hPBMs. O hormônio estimulou maior liberação de citocinas inflamatórias, genes envolvidos nessa resposta e suprimiu a liberação de marcadores inflamatórios importantes para a fagocitose contra S. aureus. Em relação ao sexo, as fêmeas desempenharam uma melhor resposta de defesa imune contra esse patógeno.Este trabalho traz uma abordagem não apresentada na literatura, com dados sobre a atuação do DHT na resposta inflamatória contra S. aureus, podendo estes serem considerados resultados importantes para gerar novos estudos na área sobre os andrógenos e seu papel na resposta contra microrganismos, que é de grande interesse científico.

  • Introduction: Staphylococcus aureus infections have a high morbidity and mortality rate, considered a major problem for public health. There is little information about the role of dihydrotestosterone in infections caused by S. aureus. Objective: To evaluate the influence of 5α-dihydrotestosterone (DHT) on the response of murine peritoneal macrophages (MPMs) and human peripheral blood monocytes (hPMs) in the immune response induced by Staphylococcus aureus. Methodology: An in vitro model was performed from male orchiectomized (OQX) and female BALB/C Shams MPMs and from healthy young male and female MPMs of childbearing age, with sex steroid levels within the normal range. Cells were inoculated with 10μl of S. aureus cap 80 or with sterile saline (control) for a period of 6h. Cells from OQX males, females, male and female hPMs were also pre-treated with a concentration of 100μl (10-2M) of DHT for 24h before stimulation with S. aureus. Cytokines TNF-α, IL-1α, IL-1β, IL-6, IL-8, IL-12, IL-10, GM-CSF, total nitrites, hydrogen peroxide (H2O2) were measured in culture supernatant. in MPMs and in hPMs. In addition, 84 genes involved in the immune response against S. aureus were analyzed by RT-PCR array. Results: In MPMs, cells inoculated with S. aureus from Sham males showed higher levels of inflammatory cytokines and lower concentrations of IL-10, total nitrites and H2O2 compared to OQX. Cells from male Sham inoculated with S. aureus also showed higher concentrations of pro-inflammatory cytokines and lower concentrations of IL-10, total nitrites and H2O2 compared to females. In the treatment with DHT, the concentration of inflammatory cytokines and the expression of genes such as TLR-2 and genes involved in the NF-kB pathway were higher in OQX treated with the hormone compared to OQX without pretreatment. In addition, the concentrations of total nitrites and H2O2 were lower in pre-treatment cells, both in OQX male and female cells. For the human experiment, all selected research subjects were healthy and sex hormone levels were within the normal range for men and women of childbearing age (17β-estradiol, progesterone, follicle-stimulating hormones (FSH), luteinizing hormones ( LH) and testosterone). hPMs inoculated with S. aureus from men had higher concentrations of inflammatory cytokines and lower concentrations of IL-10, lower levels of total nitrites and H2O2 compared to women. In response to treatment, with DHT, the hormone inhibited the concentrations of total nitrites and H2O2 in both men and women, and we observed greater expression of TLR-2 and genes involved in the NF-kB pathway, similar to the results in mice. Conclusion: Our results showed that there is a difference between the sexes in the response to inoculation by S. aureus and that DHT has pro-inflammatory immunomodulatory properties in MPMs and in hPMs. The hormone stimulated greater release of inflammatory cytokines, genes involved in this response, and suppressed the release of important inflammatory markers for phagocytosis against S. aureus. Regarding sex, females performed a better immune defense response against this pathogen. This work presents an approach not presented in the literature, with data on the role of DHT in the inflammatory response against S. aureus, which can be considered important results to generate new studies in androgens and their role in the response against microorganisms, which is of great scientific interest.

  • Introdução. Os mecanismos pelos quais o exercício promove seus efeitos benéficos no pâncreas ainda não foram totalmente elucidados no diabetes mellitus tipo 1. Diante do exposto, o presente estudo visa comparar os efeitos do exercício físico iniciado previamente e após a indução do diabetes sobre alterações metabólicas e lesões pancreáticas, analisando aspectos morfofuncionais, as expressões de fatores de transcrição e de marcadores de inflamação, apoptose e estresse oxidativo no tecido pancreático, correlacionando essas alterações às modificações da função das células β produtoras de insulina. Métodos. Trinta ratas Wistar, com peso entre 290 a 310 g, divididas em 5 grupos: CS (controle sedentário, n=6), CT (controle treinado, n=6), DS (diabético sedentário, n= 6), DT (diabético treinado, n=6) e DPT (diabético previamente treinado, n=6). Apenas o grupo DPT foi submetido a 4 semanas de exercício antes da indução do diabetes por estreptozotocina (40 mg/kg, iv). Após confirmação do diabetes, os grupos DPT, DS e CT foram submetidos a 8 semanas de exercício. O tecido pancreático foi coletado para estudos de estresse oxidativo, imuno-histoquímico, ELISA e Western Blot. Resultados. O exercício aeróbico moderado melhorou o controle metabólico e reduziu o estresse oxidativo/nitrativo, inflamação, apoptose e a expressão de SIRT 1 nas ilhotas pancreáticas das ratas diabéticas dos grupos DT e DPT comparadas às ratas sedentárias do grupo DS (p<0,05). Ambos os grupos de animais diabéticos submetidos ao exercício apresentaram melhora do perfil lipídico marcada pela redução do colesterol total e triglicerídeos plasmático, LDL-c e VLDL-c com aumento do HDL-c levando à redução do índice aterogênico, comparados aos animais diabéticos sedentários (p<0,05). Apenas o grupo DPT apresentou redução da expressão protéica de Nrf2 (p<0,05) e HO-1 (p<0,05), redução dos níveis de IL-10 e aumento dos níveis de IL-6 e (p<0,05), maior capacidade física e prevenção da perda de peso e da diminuição da insulina pancreática comparados ao grupo DS. Os protocolos de exercício reduziram os níveis de TBRAS no pâncreas e no soro dos animais DT e DPT, a peroxidação lipídica, e a expressão de nitrotirosina, expressão gênica e atividade das enzimas antioxidants catalase e GPx, nitritos totais e TNF no pâncreas dos grupos DS e DPT em relação ao grupo DS (p<0,05). A expressão proteica do Nrf2 foi positivamente correlacionada com os níveis pancreáticos de TBARS (P < 0.0001) e nitritos (P < 0.01). Enquanto, a expressão proteica de HO-1 (P < 0.0006) foi positivamente correlacionada com os níveis de TBARS no pâncreas. O exercício também reduziu a apoptose, através da inibição do fatores pró-apoptóticos Bad, Bax e caspase-3 e aumento do marcador anti-apoptótico Bcl2 além de melhorar a expressão protéica e número de células positivas para SIRT1 nos grupos DS e DPT (p<0,05). Foi observada expressão de SIRT1 negativamente correlacionada aos níveis de TNF (P <0.0091) e TBARS (P <0.0386) no pâncreas. Conclusão. Nossos dados demonstram que o exercício moderado promoveu efeitos benéficos no pâncreas ao reduzir a peroxidação lipídica, processo inflamatório e apoptose em ratas com diabetes tipo 1.

  • Introduction. The mechanisms by which exercise promotes its beneficial effects in the pancreas have not yet been fully elucidated in type 1 diabetes mellitus. Aim. The study of exposure factors and physical exercise analyzed initiated previously aims at a diabetes treatment on metabolic and pancreatic changes, as expressions of presentation factors and injury markers, to oxidative stress in pancreatic tissue, correlating these changes to the functions of the pancreas. producer of insulin-producing cells. Methods. Thirty female Wistar rats, weighing between 290 and 310 g, divided into 5 groups: SC (sedentary control, n=6), TC (trained control, n=6), SD (sedentary diabetic, n=6), TD (trained diabetic, n=6) and PTD (previously trained diabetic, n=6). Only the D+PEx group was submitted to 4 weeks of exercise before the induction of diabetes by streptozotocin (40 mg/kg, iv). After confirming of diabetes, the PTD, TD and TC groups were submitted to 8 weeks of exercise. Pancreatic tissue was collected for studies of oxidative stress, immunohistochemical, ELISA and Western Blot. Results. Moderate aerobic exercise improved metabolic control and reduced oxidative/nitractive stress, inflammation, apoptosis and SIRT 1 expression in the pancreatic islets of diabetic rats in the TD and PTD groups compared to sedentary rats the SD group (p<0.05). Both groups of diabetic animals submitted to exercise showed an improvement in the lipid profile marked by a reduction in total cholesterol and plasma triglycerides, LDL-c and VLDL-c with an increase in HDL-c leading to a reduction in the atherogenic index, compared to sedentary diabetic animals (p<0.05). Only the PTD group had reduced Nrf2 (p<0.05) and HO-1 expression (p<0.05), reduced IL-10 levels and increased IL-6 levels and (p<0.05), greater physical capacity and prevented weight loss, decrease in pancreatic insulin, and increase in plasma TBARS levels. Exercise protocols reduced the lipid peroxidation and expression of nitrotyrosine, antioxidants enzymes (catalase and GPx), totals nitrites and TNF in the pancreas of SD and PTD groups compared to the SD group (p<0.05). Nrf2 protein expression was positively correlated with pancreatic levels of TBARS (P < 0.0001) and nitrites (P < 0.01). The HO-1 protein expression (P < 0.0006) was positively correlated with pancreas TBARS levels. Exercise also reduced apoptosis by inhibiting the pro-apoptotic factors Bad, Bax and caspase-3 and increasing the anti-apoptotic marker Bcl2, in addition to improving protein expression and number of SIRT1-positive cells in the DS and DPT groups (p <0.05). SIRT1 expression was observed to negatively correlated with TNF (P<0.0091) and TBARS (P<0.0386) levels in the pancreas. Conclusion. Our data demonstrate that moderate exercise promoted beneficial effects in the pancreas by reducing lipid peroxidation, inflammatory process and apoptosis in female rats with type 1 diabetes.

  • Objetivo: Avaliar os efeitos da dieta cetogênica hiperproteica sobre parâmetros relacionados à homeostase glicêmica e alterações hepatorrenais em ratos Wistar obesos submetidos ou não ao exercício. Métodos: Foram utilizados 40 ratos machos Wistar, pesando entre 150 e 200g que inicialmente receberam as dietas controle (n=6) ou hipercalórica (n=34) para indução da obesidade por 8 semanas. Após este período, os animais do grupo controle (n=6) e parte do grupo hipercalórica (n=6) foram eutanasiados, sendo os animais obesos remanescentes subdivididos em quatro grupos, recebendo dietas e submetidos ou não ao exercício físico de moderada intensidade por 12 semanas: hipercalórica e sedentários (n=7); hipercalórica e exercitados (n=7); cetogênica hiperproteica e sedentários (n=7); e cetogênica hiperproteica e exercitados (n=7). Ingestão alimentar, parâmetros de composição e adiposidade corporal, pressão arterial, perfil bioquímico e inflamatório sérico, homeostase glicêmica, resistência e sensibilidade à insulina foram avaliados. Além disso, avaliou-se a expressão de IR, AMPK, Akt, GSK3β e GLUT4 no músculo, a função renal e hepática, o perfil morfométrico, inflamatório e oxidativo dos rins, a deposição de gordura no fígado, a morfologia, o estresse oxidativo e a imunomarcação de insulina e glucagon no pâncreas. Resultados: O modelo de obesidade induzida por dieta foi validado. O tratamento da obesidade com a dieta cetogênica hiperproteica e/ou com o exercício por 12 semanas determinou redução da ingestão alimentar e calórica, do ganho de massa corporal e da adiposidade. Apesar do aumento na glicemia, a combinação da dieta e exercício reduziu insulina sérica, e atenuou os indicadores de resistência periférica. O exercício isolado aumentou expressão de glucagon e reduziu insulina no pâncreas, aumentou expressão do IR, GLUT4 e GSK3β no músculo. A dieta e o exercício alteraram os indicadores de inflamação sistêmica, o perfil lipídico sérico e os marcadores de função hepática, além de reduzirem a deposição macrovesicular de lipídios no fígado, melhorarem o perfil oxidativo no pâncreas e atenuarem a peroxidação lipídica no rim. A dieta cetogênica hiperproteica, por si só, promoveu alterações na função e inflamação renal, sem, induzir modificações estruturais significativas. Conclusão: A dieta cetogênica hiperproteica associada ou não ao exercício por 12 semanas teve impacto positivo sobre a redução da massa e adiposidade corporais e deposição de gordura no fígado, mas não permitiu constatação sobre a melhora da homeostase glicêmica, da resposta insulínica e do perfil lipídico. As alterações pancreáticas e hepatorrenais pareceram discretas e insuficientes para se determinar efeitos positivos ou negativos, assim, mais investigações são requeridas, sobretudo, das vias metabólicas relacionadas. Nesse sentido, sugere-se cautela na utilização desta dieta, mesmo quando associada ao exercício, visto que algumas lacunas foram evidenciadas, especialmente no que se refere à homeostase glicêmica e ao comprometimento da função renal devido à proteinúria.

  • Objective: To evaluate the effects of a high-protein ketogenic diet on parameters related to glycemic homeostasis and hepatorenal alterations in obese Wistar rats submitted or not to exercise. Methods: Forty male Wistar rats weighing between 150 and 200g were used, which initially received control (n=6) or hypercaloric (n=34) diets to induce obesity for 8 weeks. After this period, the animals of the control group (n=6) and part of the hypercaloric group (n=6) were euthanized, with the remaining obese animals subdivided into four groups, receiving diets and submitted or not to moderate-intensity physical exercise for 12 weeks: hypercaloric and sedentary (n=7); hypercaloric and exercised (n=7); high-protein ketogenic and sedentary (n=7); high-protein ketogenic and exercised (n=7). Food intake, body composition and adiposity parameters, blood pressure, serum biochemical and inflammatory profile, glycemic homeostasis, insulin resistance and sensitivity were evaluated. In addition, we evaluated the expression of IR, AMPK, Akt, GSK3β and GLUT4 in muscle, kidney and liver function, morphometric, inflammatory and oxidative profile of the kidneys, fat deposition in the liver, morphology, oxidative stress and immunostaining of insulin and glucagon in the pancreas. Results: The diet-induced obesity model was validated. The treatment of obesity with a high-protein ketogenic diet and/or exercise for 12 weeks determined a reduction in food and caloric intake, body mass gain and adiposity. Despite the increase in blood glucose, the combination of diet and exercise reduced serum insulin, and attenuated peripheral resistance indicators. Isolated exercise increased glucagon expression and reduced insulin in the pancreas, increased IR, GLUT4 and GSK3β expression in muscle. Diet and exercise altered indicators of systemic inflammation, serum lipid profile and liver function markers, in addition to reducing macrovesicular lipid deposition in the liver, improving the oxidative profile in the pancreas and attenuating lipid peroxidation in the kidney. The high-protein ketogenic diet, by itself, promoted changes in kidney function and inflammation, without inducing significant structural changes. Conclusion: The high-protein ketogenic diet associated or not with exercise for 12 weeks had a positive impact on the reduction of body mass and adiposity and fat deposition in the liver, but did not allow confirmation of the improvement in glycemic homeostasis, insulin response and lipid profile. Pancreatic and hepatorenal alterations seemed discreet and insufficient to determine positive or negative effects, thus, further investigations are required, especially of the related metabolic pathways. In this sense, caution is suggested in the use of this diet, even when associated with exercise, since some gaps were evidenced, especially with regard to glycemic homeostasis and impairment of renal function due to proteinuria.

  • INTRODUÇÃO/OBJETIVO: A combinação de crescentes riscos metabólicos e envelhecimento da população, provavelmente, continuará a impulsionar as tendências crescentes de Doenças Crônicas não Transmissíveis (DCNT) em nível global. Devido à alta mortalidade na população idosa, a hipertensão arterial sistêmica (HAS) e o diabetes mellitus tipo 2 (DM2) são DNCT de grande relevância. Essas doenças, apesar dos diferentes órgãos e sistemas envolvidos, têm um fator de risco comum: em geral, inflamação clinicamente silenciosa de baixo grau. Esta inflamação tem como fator preponderante o excesso de tecido adiposo. Nesse sentido, este trabalho teve como finalidade o estudo ampliado de marcadores antropométricos, bioquímicos e inflamatórios de homens idosos com HAS e DM2. MÉTODOS: O estudo envolveu a avaliação de 49 homens com idades entre 60-80 anos, portadores de HAS e DM2. Os grupos foram divididos de acordo com o IMC, em idosos com eutrofia (IE, n= 24) ou idosos com sobrepeso (IS, n= 25). Dados antropométricos foram coletados com a análise de bioimpedância, enquanto os dados inflamatórios e bioquímicos foram realizados a partir da coleta sanguínea. Os soros obtidos foram avaliados por ensaios enzimáticos. RESULTADOS: Apesar da maioria dos idosos realizarem tratamento medicamentoso para diabetes e hipertensão, eles não tinham os níveis glicêmico e pressórico controlados. Idosos com sobrepeso apresentaram maior índice glicêmico (p=0,0201). Todos os dados antropométricos analisados apresentaram diferenças estatísticas significativas, quando comparados entres os grupos. Houve aumento da maioria dos marcadores antropométricos em idosos com sobrepeso. Em contrapartida, nesse mesmo grupo, houve diminuição nos valores de massa magra (p<0,0001), massa óssea (p<0,0001), água (p<0,0001) e metabolismo basal (p=0,001). Idosos com excesso de peso apresentaram maiores médias para todas as citocinas inflamatórias analisadas. Estas incluem IL-17A (p=0,0154), IL-1β (p=0,0376), INF-y (p=0,0442) e TNF-α (p=0,0064). Em contrapartida, houve diminuição da citocina anti-inflamatória IL-10 (p=0,0367) em idosos com sobrepeso. Além disso, observamos o aumento da relação IL-10/IL-17A em idosos com eutrofia em relação aos idosos com sobrepeso. Os padrões de correlações paras as variáveis bioquímicas, inflamatórias e de composição corporal são distintas entre os grupos. Em um contexto amplo, as correlações são mais intensas em idosos com eutrofia, principalmente no que concerne as variáveis inflamatórias e a variável de massa gorda. CONCLUSÃO: Apesar dos idosos com sobrepeso terem uma exacerbação dos parâmetros inflamatórios, vimos que a intensidade de suas correlações com os parâmetros antropométricos e bioquímicos se torna menos evidente. Além disso, constatamos que o padrão de influência dessas citocinas é bem distinto em relação a composição corporal.

  • INTRODUCTION/OBJECTIVE: The combination of increasing metabolic risks and an aging population will likely continue to drive the growing trends in Chronic Non-Communicable Diseases (NCDs) globally. Systemic arterial hypertension (SAH) and type 2 diabetes mellitus (DM2) are highly relevant NCDs due to the high mortality in the elderly population. Despite the different organs and systems involved has a common risk factor: clinically inflammation silence low-grade. This inflammation has as its principal factor the excess of adipose tissue. In this sense, this work aimed at the expanded study of anthropometric, biochemical, and inflammatory markers of elderly with SAH and DM2. METHODS: The study involved the evaluation of 49 men aged 60-80 years with SAH and DM2. The divided groups according to BMI into eutrophic elderly (EE, n = 24) or overweight elderly (OE, n = 25). Bioimpedance analysis was used to collect anthropometric data. Blood collection was performed to analyze inflammatory and biochemical data. The serum obtained was evaluated by enzymatic assays. RESULTS: Although most of the elderly undergo drug treatment for diabetes and hypertension, they did not have their blood glucose and blood pressure levels controlled. Overweight elderly had a higher glycemic index (p = 0.0201). All the anthropometric data analyzed showed statistically significant differences when compared between groups. There was an increase in most anthropometric markers in overweight elderly. In contrast, in this same group, there was a decrease in the values of lean mass (p <0.0001), bone mass (p <0.0001), water (p <0.0001) and basal metabolism (p = 0.001). Overweight elderly had higher averages for all analyzed inflammatory cytokines. These include: IL-17A (p = 0.0154), IL-1β (p = 0.0376), INF-y (p = 0.0442) and TNF-α (p = 0.0064). In contrast, there was a decrease in the anti-inflammatory cytokine IL-10 (p = 0.0367) in overweight elderly. Also, we observed an increase in the IL-10/IL-17A ratio in eutrophic elderly about overweight elderly. The correlation patterns for biochemical, inflammatory, and body composition variables are different between groups. The correlations are more intense in the eutrophic elderly, especially concerning inflammatory variables and the fat mass variable. CONCLUSION: Although the overweight elderly has an increase in inflammatory parameters, the intensity of their correlations with anthropometric and biochemical parameters are less evident. Also, we found that the pattern of influence of these cytokines is quite different about body composition.

  • Gardnerella vaginalis é uma bactéria comensal da microbiota vaginal e o principal agente da vaginose bacteriana (VB), uma infecção vaginal estabelecida devido ao desequilíbrio nesse ecossistema. A patogênese da vaginose bacteriana não está bem estabelecida, no entanto é considerada a principal desordem vaginal que acomete mulheres em idade reprodutiva o que pode hipotetizar a influência dos hormônios sexuais femininos em sua patogênese. Objetivo: Avaliar a influência do 17β-estradiol (E2) na resposta imunológica induzida por Gardnerella vaginalis em modelos in vitro de macrófagos peritoneais murinos (MPMs) fêmeas e monócitos humanos do sangue periférico (hPMs) de mulheres, com ou sem atividade ovariana fisiológica. Metodologia: Realizado experimentos in vivo e in vitro. Para os experimentos in vivo nos animais, foram realizados: peso corporal, índice uterino e dosagem de estradiol e em humanos, foram realizados exames laboratoriais como hemograma, bioquímicos e hormonais. Para os experimentos in vitro os macrófagos peritoneais de fêmeas BALB/C cirurgia simulada (SHAM) ou ovariectomizadas (OVX) e os monócitos humanos do sangue periférico (hPMs) de mulheres em período fértil e menopausa foram subdivididos em três grupos: células inoculadas com G. vaginalis, células com salina estéril, ambas por 6 horas, e células pré-tratadas com 17β-Estradiol (E2), por 6 horas, e inoculadas com G. vaginalis, também por 6 horas. Marcadores inflamatórios como: citocinas GM-CSF, IL-1β, TNF-α, IL-6, IL-12, IL-23, IL-27 e IL-10, nitritos totais, peróxido de hidrogênio (H2O2), foram mensurados em sobrenadante de cultura e a expressão dos genes TLR-2, NF-kB e ERα e ERβ Além disso, uma curva de crescimento foi realizada em dois grupos. O grupo um, apenas com G. vaginalis e o grupo dois, G. vaginalis com 17β-estradiol, durante um período de 24 horas. Resultados: Para a análise da curva de crescimento, pode ser constatada a atividade inibitória do E2 sobre a G. vaginalis. Para os experimentos em animais: In vivo, fêmeas SHAMs apresentaram maior peso uterino e maior concentração de estradiol, enquanto fêmeas OVX apresentaram maior peso corporal. In vitro, a concentração das citocinas, IL-6, IL-8, IL-10 e os marcadores inflamatórios, nitritos totais e o peróxido de hidrogênio (H2O2), apresentaram níveis elevados em MPMs de fêmeas SHAM e OVX inoculadas com G. vaginalis quando comparados com salina estéril. Ao tratamento com E2, os MPMs das fêmeas OVX apresentaram maiores níveis de IL-10 e peróxido de hidrogênio quando com parados com OVX sem tratamento. Entretanto, apenas o peróxido de hidrogênio se equiparou aos MPMs das fêmeas SHAM. Para as expressões gênicas de TLR-2, NF-kB e ERα e ERβ resultado similar foi observado nos modelos SHAM e OVX entre infectados e salina estéril. As comparações entre as células infectadas dos modelos SHAM, OVX e OVX tratados com E2 também apresentaram maiores níveis de citocinas e marcadores inflamatórios e expressões gênicas para células do modelo SHAM, com exceção apenas para expressão do ERβ. Para os experimentos em humanos: In vivo, todos os sujeitos de pesquisa selecionados estavam saudáveis, porém foi observado maior concentração de glicose, colesterol total, LDL e TGP nas mulheres na fase de menopausa e maior concentração de leucócitos nas mulheres em período fértil. Para a dosagem sérica dos hormônios sexuais foi observado que mulheres em período fértil apresentaram maiores concentrações de 17β-estradiol e progesterona, enquanto as mulheres na menopausa, concentrações elevadas dos hormônios folículo estimulante (FSH) e luteinizante (LH). In vitro, os hPMs infectados com G. vaginalis de mulheres no período reprodutivo e na menopausa apresentaram maiores níveis de nitritos totais, H2O2 e citocinas quando comparado com salina. Entretanto, os hPMs infectados de mulheres na menopausa apresentaram maiores níveis de H2O2 em comparação com hPMs de mulheres no período reprodutivo. Em se tratando das citocinas as células tratadas com E2 não se equipararam às com E2 endógeno, exceto IL-10. Conclusão: Aos experimentos em animais e humanos, em relação à influência do E2 na resposta inflamatória aguda ao desafio, in vitro, por G. vaginalis, observa-se que o hormônio apresenta característica imunomodulatória, mas não podendo ser, exclusivamente, definida como pró ou antiinflamatória. Neste estudo, para o E2 endógeno, todas as citocinas, marcadores inflamatórios e expressões gênicas apresentaram maiores níveis, tanto em MPMs quanto para hPMs. Ao tratamento com E2, os MPMs de fêmeas OVX e hPMS de mulheres na menopausa apresentaram similaridade de resposta para os níveis de IL-10 em relação as células com E2 endógeno, indicando uma ação anti-inflamatória. Porém, os genes de reconhecimento e desencadeamento de inflamação aguda também apresentaram similaridade entre MPMs tratadas com E2 em relação às células com E2 exógeno. Influências fisiológicas, como a progesterona endógena, podem contribuir para que células apresentem características de resposta inflamatória aguda, e o tratamento de células, exclusivamente com E2, não tenha resultado em uma equiparação de resposta de células com hormônios sexuais endógenos. Entretanto, o E2 apresentou característica bacteriostática em relação a proliferação G. vaginalis, podendo este hormônio modular fatores de virulência deste microrganismo, assim, contribuindo para uma ausente ou discreta resposta imunológicas no curso da vaginose bacteriana.

  • Gardnerella vaginalis is a commensal bacterium of the vaginal microbiota and the main agent of bacterial vaginosis (BV), a vaginal infection established due to an imbalance in this ecosystem. The pathogenesis of bacterial vaginosis is not well established; however, it is considered the main vaginal disorder that affects women of reproductive age, which may hypothesize the influence of female sex hormones in its pathogenesis. Objective: To evaluate the influence of 17β-estradiol (E2) on the immune response induced by Gardnerella vaginalis in in vitro models of female murine peritoneal macrophages (MPMs) and human peripheral blood monocytes (hPMs) from women, with or without physiological ovarian activity. Methodology: in vivo and in vitro experiments were carried out. For the in vivo experiments in animals, the following were performed: body weight, uterine index and estradiol dosage; and in humans, laboratory tests such as blood count, biochemical and hormonal tests were performed. For the in vitro experiments, peritoneal macrophages from BALB/C sham surgery (SHAM) or ovariectomized (OVX) females and human peripheral blood monocytes (hPMs) from fertile and menopausal women were subdivided into three groups: cells inoculated with G. vaginalis, cells with sterile saline, both for 6 hours, and cells pretreated with 17β-Estradiol (E2), for 6 hours, and inoculated with G. vaginalis, also for 6 hours. Inflammatory markers such as: cytokines GM-CSF, IL-1β, TNFα, IL-6, IL-12, IL-23, IL-27 and IL-10, total nitrites, hydrogen peroxide (H2O2), were measured in culture supernatant, and the expression of TLR-2, NF-kB and ERα and ERβ genes. In addition, a growth curve was performed in two groups. Group one, with only G. vaginalis and group two, G. vaginalis with 17β-estradiol, for a period of 24 hours. Results: For the analysis of the growth curve, the inhibitory activity of E2 on G. vaginalis can be verified. For animal experiments: In vivo, SHAM females had higher uterine weight and higher estradiol concentration, while OVX females had higher body weight. In vitro, the concentration of cytokines, IL-6, IL-8, IL-10 and inflammatory markers, total nitrites and (H2O2), showed high levels in MPMs of SHAM and OVX females inoculated with G. vaginalis when compared to G. vaginalis with sterile saline. When treated with E2, the MPMs of OVX females showed higher levels of IL-10 and hydrogen peroxide, when compared to untreated OVX. However, only the hydrogen peroxide matched the MPMs of the SHAM females. Similar results were observed for gene expressions of TLR-2, NF-kB and ERα and ERβ in the SHAM and OVX models between infected and sterile saline. Comparisons between infected cells from the SHAM, OVX and OVX treated with E2 models also showed higher levels of cytokines and inflammatory markers and gene expressions for cells from the SHAM model, with the exception only for the expression of ERβ. For the human experiments: In vivo, all selected research subjects were healthy, but a higher concentration of glucose, total cholesterol, LDL and TGP was observed in menopausal women and a higher concentration of leukocytes in women in their fertile period. For the serum dosage of sex hormones, it was observed that women in the fertile period had higher concentrations of 17β-estradiol and progesterone, while women in menopause had high concentrations of follicle-stimulating (FSH) and luteinizing (LH) hormones. In vitro, hPMs infected with G. vaginalis from women in the reproductive period and menopause showed higher levels of total nitrites, H2O2 and cytokines when compared to saline. However, infected hPMs from menopausal women had higher levels of H2O2 compared to hPMs from women in the reproductive period. With regard to cytokines, cells treated with E2 did not match those with endogenous E2, except for IL-10. Conclusion: In animal and human experiments, in relation to the influence of E2 on the acute inflammatory response to G. vaginalis, it is observed that the hormone has an immunomodulatory characteristic, but it cannot be exclusively defined as pro or anti-inflammatory. In this study, for endogenous E2, all cytokines, inflammatory markers and gene expressions showed higher levels, both in MPMs and hPMs. When treated with E2, MPMs from OVX females and hPMS from menopausal women showed similar response for IL-10 levels in relation to cells with endogenous E2, indicating an anti-inflammatory action. However, acute inflammation recognition and triggering genes also showed similarity between MPMs treated with E2 and cells with exogenous E2. Physiological influences, such as endogenous progesterone, may contribute for cells to present characteristics of an acute inflammatory response, and the treatment of cells, exclusively with E2, has not resulted in an equivalence of cell response with endogenous sex hormones. However, E2 showed a bacteriostatic characteristic in relation to G. vaginalis proliferation, and this hormone can modulate virulence factors of this microorganism, thus contributing to an absent or discrete immune response in the course of bacterial vaginosis.

  • Objetivo: Avaliar os efeitos dos exercícios preventivo e terapêutico sobre a morfologia e função pancreática e hepática de ratos Wistar em um modelo de envelhecimento e obesidade induzida por dieta hiperlipídica. Métodos: Foram utilizados 24 ratos Wistar machos (idade inicial = 4 meses e idade final = 14 meses) que foram randomicamente distribuídos em três grupos experimentais envelhecidos e obesos (n = 8/grupo): sedentário, exercício terapêutico e exercício preventivo. Avaliou-se parâmetros de adiposidade corporal, concentração plasmática e imunomarcação pancreática de insulina, marcadores de inflamação teciduais, peroxidação lipídica, atividade e imunomarcação de enzimas antioxidantes e parâmetros de morfologia pancreática e hepática. Resultados: Somente o exercício preventivo melhorou os parâmetros de adiposidade corporal e concentração plasmática de insulina. Os exercícios terapêutico e preventivo aumentaram a densidade de ilhotas pancreáticas, mas não exerceram efeitos sob a área e diâmetro menor das ilhotas. A infiltração de macrófagos no pâncreas foi reduzida apenas nos animais submetidos ao exercício preventivo enquanto que no fígado, este efeito resultou tanto do exercício preventivo, quanto do terapêutico. Além disso, os animais submetidos aos exercícios preventivo e terapêutico apresentaram menor imunomarcação para insulina, Fator Nuclear Kappa B (NF-kB) e Fator de Transformação do Crescimento b (TGF-b) no parênquima pancreático, bem como, menor peroxidação lipídica tecidual pancreática e hepática, com o efeito sendo maior no grupo de exercício preventivo. Ambos os protocolos de exercício promoveram menor área de fibrose, aumento da atividade das enzimas antioxidantes catalase e Glutationa Peroxidase (GPx) e aumento da imunomarcação para Hemeoxigenase-1 (HO-1), tanto no pâncreas, quanto no fígado, sendo que o exercício preventivo também apresentou maior efeito sobre estas variáveis. A esteatose hepática macrovesicular foi menor nos animais submetidos aos dois protocolos de exercício, no entanto, a esteatose microvesicular não diferiu entre os grupos experimentais. Conclusão: Os efeitos benéficos do exercício físico, principalmente do preventivo, sobre a adiposidade corporal, insulinemia, morfologia, inflamação e estresse redox nos parênquimas hepático e pancreático sob condições combinadas de envelhecimento e obesidade revelam-se promissores na prevenção de processos degenerativos nestes órgãos sob ambas as condições.

  • Objectives: To evaluate the effects of preventive and therapeutic exercises on pancreatic and hepatic morphology and function of Wistar rats in an aging and obesity model. Methods: Twenty four male Wistar rats (initial age = 4 months and final age = 14 months) were randomly assigned to three experimental aged and obese groups (n = 8 / group): sedentary, therapeutic exercise, preventive exercise. Body composition parameters, plasma concentration and immunostaining of pancreatic insulin, markers of tissue inflammation, lipid peroxidation, activity and immunostaining of antioxidant enzymes and parameters of pancreatic and hepatic structure were evaluated. Results: Only preventive exercise improved the parameters of body composition and plasma insulin concentration. Pancreatic histology showed that therapeutic and preventive exercises increased the density of pancreatic islets, but had no effect on the islet area and smaller diameter. Analysis of pancreatic and hepatic parameters indicated that only preventive exercise reduced macrophage infiltration into the pancreas and both therapeutic and preventive exercise reduced macrophage infiltration in the liver. In addition, preventive and therapeutic exercise animals had lower immunostaining for insulin Factor Nuclear Kappa B (NF-kB) and Transforming Growth Factor Beta (TGF-b) in the pancreatic parenchyma, as well as lower hepatic and pancreatic tissue lipid peroxidation, with the effect being significantly higher in the preventive exercise group. Both exercise protocols promoted smaller area of fibrosis, increased activity of catalase and Glutathione peroxidase (GPx) enzymes and increased immunostaining for Heme oxygenase 1 (HO-1) in both pancreas and liver, and preventive exercise had a greater effect on these variables. Macrovesicular steatosis was lower in animals submitted to both types of exercise protocol; however, microvesicular steatosis was not different between experimental groups. Conclusion: The beneficial effects of exercise, especially preventive exercise, on body adiposity, insulinemia, morphology, inflammation and redox stress in the liver and pancreatic parenchyma under combined conditions of aging and obesity are promising in preventing degenerative processes in these organs under both conditions.

  • As doenças crônicas não transmissíveis (DCNT) são responsáveis por cerca de 70% de todas as mortes no mundo. Neste cenário, a hipertensão arterial sistêmica (HAS) e o diabetes mellitus tipo 2 (DM2) são doenças que apresentam vários aspectos em comum. Nelas biomarcadores oferecem uma compreensão dos processos de doenças e podem beneficiar a detecção precoce, o tratamento e o seu manejo. Desse modo, o objetivo deste estudo foi avaliar as concentrações de biomarcadores inflamatórios no soro de pacientes com diferentes gravidades de DM2 e HAS bem como analisar as correlações entre interleucinas e a gravidade da doença. Inicialmente, pacientes previamente diagnosticados com DM2 e HAS foram estratificados em diferentes graus das doenças, visando detectar os fatores que mais influenciam nas complicações das doenças supracitadas. Os grupos foram divididos em baixo risco (1), risco moderado (2), risco alto (2), risco muito alto (4), de acordo com a Sociedade Europeia de Cardiologia, a qual correlaciona os valores da pressão arterial com a presença de fatores de risco. Para realização desse trabalho, voluntários foram recrutados para aferição da pressão e foi realizada análise da composição corporal através da bioimpedância. Foram coletados sangue desses pacientes para dosagens bioquímicas e para realização de ELISA, visando detectar as concentrações das citocinas IL-17, IL-1β, IFN-γ, TNF-α e IL-10 no soro. As análises estatísticas foram realizadas utilizando o teste Kruskal-Wallis, e as correlações foram analisadas através do software R Studio. Observamos que quanto maior a gravidade, maior a concentração sérica de citocinas pró-inflamatórias, entretanto, não foi observada diferença significativa entre os sexos ou período de climatério. Correlações positivas fortes foram encontradas entre IL-17, IL-1β e IFN-γ do grupo de baixo risco, entretanto, essas correlações são perdidas conforme aumenta o grupo de risco. Esses achados sugerem que a inflamação está presente contribuindo para o agravamento da HAS e DM2. Através de análises bayesianas encontramos a hiperglicemia exercendo um papel central na modulação do estado pró-inflamatório. Esse estado pode ser alterado com a prática de atividade física, confirmando o seu efeito benéfico e protetor. Este é o primeiro trabalho que mostra a complexa interação entre diferentes biomarcadores no DM2 e na HAS, principalmente a hiperglicemia como centro indutor da inflamação, e como a interação desses marcadores aumenta a gravidade das doenças.

  • Noncommunicable chronic diseases (NCDs) account for about 70% of all deaths worldwide. In this scenario, systemic arterial hypertension (SAH) and type 2 diabetes mellitus (DM2) are diseases that have several aspects in common. These biomarkers provide an understanding of disease processes and can benefit early detection, treatment and management. Thus, the aim of this study was to evaluate serum biomarker concentrations in serum of patients with different severities of DM2 and SAH as well as to analyze the correlations between interleukins and disease severity. Initially, patients previously diagnosed with DM2 and hypertension were stratified into different degrees of disease, aiming to detect the factors that most influence the complications of the aforementioned diseases. The groups were divided into low risk (1), moderate risk (2), high risk (2), very high risk (4), according to the European Society of Cardiology, which correlates blood pressure values with the presence of of risk factors. To perform this work, volunteers were recruited for pressure measurement and body composition analysis was performed through bioimpedance. Blood was collected from these patients for biochemical measurements and ELISA to detect serum concentrations of cytokines IL-17, IL-1β, IFN-γ, TNF-α and IL-10. Statistical analyzes were performed using the Kruskal-Wallis test, and correlations were analyzed using the R Studio software. We observed that the higher the severity, the higher the serum concentration of proinflammatory cytokines, however, no significant difference was observed between sexes or climacteric period. Strong positive correlations were found between IL-17, IL-1β and IFN-γ of the low risk group, and these correlations are lost as the risk group increases. These findings suggest that inflammation is present contributing to the worsening of hypertension and DM2. Through Bayesian analyzes we found hyperglycemia playing a central role in modulating the proinflammatory state. This state can be changed with the practice of physical activity, confirming its beneficial and protective effect. This is the first paper that shows the complex interaction between different biomarkers in DM2 and SAH, especially hyperglycemia as inflammation inducer, and how the interaction of these markers increases disease severity.

  • Peltogyne pauciflora Benth (Leguminosae), é uma planta de caatinga no Brasil, conhecida popularmente como buranhê e imburanhê, sendo a infusão da casca utilizada para o tratamento da “inflamação e dor na coluna”. O objetivo deste estudo foi avaliar o extrato etanólico da casca de P. pauciflora e suas frações quanto  a sua constituição química, atividade anti-inflamatória, antinociceptiva, antioxidante e toxicológica. As cascas da planta foram coletadas na Floresta Nacional Contendas do Sincorá e foram maceradas em etanol, obtendo o extrato etanólico (EEPP, doses 50, 100 e 200 mg/kg), sendo posteriormente particionado, originando as frações hexano (H), diclorometano (DM), acetato de etila (AE) e butanol (B), todas na dose 50 mg/kg. O EEPP e as frações H e DM foram analisados por CG-EM, sendo identificados carboidratos, ácidos graxos e compostos fenólicos, destacando as substâncias bioativas identificadas: beta-sitosterol, campesterol, lupenona, lupeol, ácido 3-metoxi-4 hidroxibenzoico, fitol, ácido oleico, linoleato de etila e o ácido palmítico. O EEPP e as frações H, DM e AE contém alto teor de fenólicos totais. Para os ensaios anti-inflamatório, antinociceptivo e antioxidante foram utilizados camundongos Balb/c machos adultos. No teste de Contorção abdominal induzido por ácido acético 0,6%, o EEPP (50 e 100 mg/kg) e as frações H, DM e AE reduziram as respostas nociceptivas em relação ao grupo veículo (VH)(p < 0,05). Quanto ao teste de Nocicepção induzida pela injeção intraplantar de formalina 1,5%, o EEPP (50 e 100 mg/kg) e as frações H e DM reduziram o número de “flinches” em ambas as fases (neurogênica e inflamatória), e a fração AE somente na fase inflamatória em relação ao VH (p < 0,05), e o edema de pata só foi reduzido por EEPP (100 mg/kg). No teste de Von Frey, EEPP (50 e 100 mg/kg) e as frações DM e AE reduziram a hipernocicepção em relação a VH (p < 0,05). A produção de mieloperoxidase pelos neutrófilos diminuiu com EEPP (50, 100, 200 mg/kg) e com as frações H, DM e AE comparados com VH (p < 0,05), assim como a peroxidação lipídica, determinado pelo ensaio de substâncias reativas ao ácido tiobarbitúrico (TBARS) (p < 0,05). O EEPP (50, 100 e 200 mg/kg) e as frações H, DM e AE diminuíram a expressão dos genes TNF-α e IL-6, e apenas EEPP (200 mg/kg) reduziu a expressão do gene IL-17 (p < 0,05). O EEPP (100 mg/kg) e as frações H e DM reduziram a permeabilidade vascular no teste de azul de Evans (p < 0,05). EEPP (50, 100 e 200 mg/kg) e as frações H e DM reduziram a migração de neutrófilos para a cavidade peritoneal (p < 0,05), assim como reduziram os níveis de produção de nitrito (p < 0,05). No teste de letalidade frente à Artemia salina L., EEPP apresentou moderada toxicidade e as frações foram consideradas inativas. Este é o primeiro relato de estudo fitoquímico e biológico do EEPP da casca de P. pauciflora e suas frações H, DM e AE, que apresentaram atividade antioxidante, antinociceptiva e anti-inflamatória, sendo uma promissora fonte natural para uso terapêutico e identificação de novos compostos. Futuramente, serão realizados estudos de identificação de compostos bioativos e mecanismos de ação, além da avaliação toxicológica com determinação da dose letal in vivo.

  • Peltogyne pauciflora Benth (Leguminosae), is a caatinga plant in Brazil, popularly known as buranhê and imburanhê, and the infusion of the bark is used to treat "inflammation and pain in the spine". The objective of this study was to evaluate the ethanolic extract of P. pauciflora bark and its fractions regarding its chemical constitution, anti-inflammatory, antinociceptive, antioxidant and toxicological activity. The bark was collected in the Contendas Sincorá National Forest and were macerated in ethanol, obtaining the ethanolic extract (EEPP, doses 50, 100 and 200 mg/kg), being later partitioned, originating the hexane (H), dichloromethane ( DM), ethyl acetate (AE) and butanol (B) fractions, all the doses at 50 mg/kg. EEPP, H and DM fractions were analyzed by GC-MS, and carbohydrates, fatty and phenolic compounds were identified, being highlighted the bioactive substances: beta-sitosterol, campesterol, lupenone, lupeol, 3-methoxy-4 acid hydroxybenzoic acid, phytol, oleic acid, ethyl linoleate and palmitic acid. EEPP, H, DM and AE fractions contain high content of total phenolics. For anti-inflammatory, antinociceptive and antioxidant tests, adult male Balb/c mice were used. In abdominal contortion test induced by 0.6% acetic acid, EEPP (50 and 100 mg/kg), H, DM and AE fractions reduced the nociceptive responses in relation to the vehicle group (VH) (p <0.05). In the nociception test induced by intraplantar injection of 1.5% formalin, EEPP (50 and 100 mg/kg), H and DM fractions reduced the number of flinches in both phases (neurogenic and inflammatory), and AE fraction reduced only in the inflammatory phase in relation to VH (p <0.05), and besides, the paw edema was reduced by EEPP (100 mg/kg). In the Von Frey test, EEPP (50 and 100 mg/kg), DM and AE fractions reduced hypernociception in relation to VH (p <0.05). Myeloperoxidase production by neutrophils decreased with EEPP (50, 100, 200 mg / kg), H, DM and AE fractions compared to VH (p <0.05), as well as the lipid peroxidation determined by the test of reactive substances thiobarbituric acid (TBARS) (p <0.05). EEPP (50, 100 and 200 mg/kg) and H, DM and AE fractions decreased the expression of the TNF-α and IL-6 genes, and only EEPP (200 mg/kg) reduces the expression of the IL-17 gene (p <0.05). EEPP (100 mg/kg), H and DM fractions reduced the vascular permeability in the Evans blue test (p <0.05). EEPP (50, 100 and 200 mg/kg), H and DM fractions reduced the levels of neutrophils in the peritoneal cavity (p <0.05), as well as reducing the levels of nitrite production (p <0.05). In the lethality test against Artemia salina L., EEPP presented moderate toxicity and the fractions were considered inactive. This is the first phytochemical and biological study report of the bark ethanolic extract of P. pauciflora and its H, DM and AE fractions, which have antioxidant, antinociceptive and anti-inflammatory activities, being a promising natural source for clinical use and identification of new compounds. In the future, studies will be carried out to identify bioactive compounds and mechanisms of action, in addition to toxicological assessment with determination of the lethal dose in vivo.

  • Uma das principais alterações do diabetes mellitus tipo 1 (DM1) é a redução progressiva das células β pancreáticas, e consequentemente da produção de insulina. A peroxidação lipídica, citocinas pró-inflamatórias, e o fator de transcrição nuclear (NF-kB), desempenham papel importante na disfunção e morte de células β durante o DM1. Além disso, o fator de crescimento transformante (TGF-β1) e a fibrose participa do desenvolvimento do diabetes mellitus (DM). Já foi demonstrado os efeitos benéficos do exercício físico sobre complicações em diversos órgãos decorrentes da DM. No entanto, não há estudos que trate seus efeitos sobre ilhotas de langerhans de ratas diabéticas ovariectomizadas.  Para isso, ratas wistar foram anestesiadas e submetidas à ovariectomia bilateral. Após a recuperação cirúrgica, os animais receberam injeção de 40 mg/kg de estreptozotocina (STZ) para a indução do DM1 ou de tampão citrato 0,1M, pH 4,5, e foram divididos em quatro grupos experimentais: 6 ratas controles sedentárias OVX (CSO); 6 controles previamente treinadas OVX (CPTO); 6 diabéticas sedentárias OVX (DSO); e 6 diabéticas previamente treinadas OVX (DPTO). Os animais dos grupos DPTO e CPTO foram submetidos a quatro semanas de treinamento aeróbico previamente à indução do DM1. Após as quatro semanas foi realizada indução do DM1 nos animais dos grupos DSO e DPTO. Em seguida, os grupos CPTO e DPTO foram submetidos ao mesmo protocolo de exercício físico aeróbico regular de intensidade moderada, o qual consistiu em corrida em esteira motorizada por um período de oito semanas. O exercício físico não aumentou o conteúdo de insulina e massa de células β nas ilhotas de langerhans do grupo DPTO. Contudo, diminuiu o conteúdo de glucagon e massa de células α, mostrando o efeito benéfico do exercício sobre o hormônio contra-regulador da homeostasia da glicose no sangue.  Além disso, a glicose do sangue do das ratas DPTO foi menor que a das ratas DSO. O exercício físico também diminuiu a expressão de TGF-β1 nas ratas DPTO em relação as ratas DSO, bem como também diminuiu a fibrose nessas ilhotas. O exercício físico reduziu o estresse nitrativo pela redução da marcação para nitrotirosina no grupo DPTO. Além disso, o exercício físico exerceu efeito fundamental na redução da expressão do fator de transcrição NF-kB. Esses resultados mostram o efeito benéfico do exercício físico previamente a indução do diabetes sobre a expressão de marcadores inflamatórios e oxidativos, estrutura das ilhotas e conteúdo do hormômio contra-regulador da concentração da glicose sanguínea.

  • One of the main changes in type 1 diabetes mellitus (T1DM) is the progressive reduction of the pancreatic β cells, and consequently insulin production. Lipid peroxidation, pro-inflammatory cytokines, and nuclear transcription factor (NF-kB) play an important role in β cell dysfunction and death during T1DM. In addition, the transforming growth factor (TGF-β1) and fibrosis participate in the development of diabetes mellitus. The beneficial effects of physical exercise on complications in various organs resulting from DM have been demonstrated. However, there are no studies evaluating the effects of exercise on islet of langerhans of diabetic ovariectomized rats. For this, wistar rats were anesthetized and submitted to bilateral ovariectomy. After surgical recovery, the animals were injected with 40 mg/kg streptozotocin (STZ) for the induction of DM1 or 0.1M citrate buffer, pH 4.5, and were divided into four experimental groups: 6 sedentary control rats OVX (SCO); 6 previously trained OVX controls (PTCO); 6 sedentary diabetic OVX (SDO); and 6 previously trained diabetic OVX (PTDO). The animals of the PTDO and PTCO groups were submetted four weeks of aerobic training previous to induction of T1DM. After four weeks, T1DM induction was performed in the animals of the SDO and PTDO groups. Then, the PTCO and PTDO groups were submitted to the same protocol of regular aerobic physical exercise of moderate intensity, which consisted of running on a motorized treadmill for a period of eight weeks. Physical exercise did not increase the insulin content and mass of β cells in the islets of langerhans in the PTDO group. However, it decreased the content of glucagon and α cell mass, showing the beneficial effect of exercise on the counter-regulating hormone of blood glucose homeostasis. In addition, blood glucose from PTDO rats was lower than that of SDO rats. Physical exercise also decreased TGF-β1 expression in PTDO rats compared to SCO rats, as well as decreased fibrosis in these islets. Physical exercise reduced nitrative stress by reduced nitrotyrosine expression in the PTDO group. In addition, physical exercise had a fundamental effect in reducing the expression of the transcription factor NF-kB. These results show the beneficial effect of physical exercise prior to the induction of diabetes on the expression of inflammatory and oxidative markers, islet structure and content of the counter-regulating hormone of blood glucose concentration.

  • Schinopsis brasiliensis Engler, popularmente conhecida como “Braúna”, é vastamente utilizada na medicina popular para tratamento de dor, febre, tosse entre outras doenças. Este estudo teve como objetivo avaliar a atividade anti-inflamatória, antinociceptiva e cicatrizante de S. brasiliensis Engler do semiárido da Bahia, coletadas na região da Floresta Nacional Contendas do Sincorá (FLONA), segundo dados etnofarmacológicos da região. Testes anteriores realizados com o extrato bruto (etanólico) e frações já comprovaram a ação antinociceptiva e anti-inflamatória presentes na folha da Braúna. Deste modo, o objetivo deste estudo foi avaliar a atividade antinociceptiva e anti-inflamatória das subfrações da folha de S. brasiliensis, iniciar o processo de identificação dos compostos bioativos, bem como elucidar os possíveis mecanismos de ação dessas subfrações. Os resultados foram obtidos por meio da coleta e identificação da espécie vegetal, análise fitoquímica, preparo do extrato etanólico, frações e subfrações, isolamento e identificação dos compostos. As subfrações testadas foram de hexano (H1, H2, H3, H4) e diclorometano (D1, D2, D3, D4 e D5). Para avaliar a atividade antinociceptiva e anti-inflamatória, foram realizados os testes de contorções abdominais induzidas por ácido acético, recrutamento de leucócitos para a cavidade peritoneal, avaliação do extravasamento peritoneal azul de Evans, nocicepção induzida pela injeção intraplantar de formalina. Para inflamação crônica, foi utilizado o teste de granuloma induzido pelo implante subcutâneo de pellet de algodão. Foi realizado o modelo experimental de ferida excisional para avaliar atividade cicatrizante do extrato bruto. Para a avaliação toxicológica, foram realizados testes de toxicidade aguda em ratos Wistar e de toxidade frente à Artemia salina (TAS). A análise estatística foi realizada por teste ANOVA de uma ou duas vias, abordagem paramétrica, seguida pelo teste de comparação múltipla de Bonferroni, com p < 0,05. As folhas de S. brasiliensis possuem polifenóis, taninos, saponinas, flavonoides, alcaloides e esteroides tanto no extrato bruto quanto nas frações estudadas. A análise por cromatografia gasosa acoplada a espectrofotometria de massa identificou 36 substâncias na fração hexânica e diclorometânica, dentre elas o Galato de Etila, substância de elevado interesse farmacológico. No experimento de contorção abdominal induzida por ácido acético, as subfrações H2, H3, H4 e D1 (50 mg/kg) reduziram significativamente a nocicepção (p < 0,05). Já o extrato etanólico (100 mg/kg) apresentou atividade antinociceptiva com ação opioide e inibitória na síntese de óxido nítrico (p < 0,05). No teste de nocicepção induzida pela injeção intraplantar de formalina, as subfrações H2, H3, H4, D1, D2, D3, D4 e D5 (50 mg/kg) reduziram o número de “flinches” induzidos pela formalina na fase 1 do experimento, (p > 0,05) e na fase 2, as subfrações H3, H4, D1, D2, D4 e D5 diminuíram significativamente a nocicepção quando comparado com o grupo veículo (p < 0,05). Nenhuma das subfrações diminuiu o edema de pata (p > 0,05). As subfrações D2, D3, D4 e D5 foram capazes de aumentar o limiar de dor dos animais no experimento de Von Frey (p < 0,05). As subfrações H2, H3, H4, D1 e D5 (50 mg/kg) apresentaram atividade anti-inflamatória reduzindo a migração de neutrófilos para a cavidade peritoneal (p < 0,05), sendo que o possível mecanismo de ação anti-inflamatório das subfrações H3, H4, D3, D4 e D5 seja pela diminuição da citocina TNF-a nos animais tratados e elevação da citocina IL-10 quando os animais foram tratados com a subfração D1. Não houve diminuição da permeabilidade vascular pelas subfrações avaliadas no teste de extravasamento azul de Evans (p > 0,05). O extrato etanólico (50 e 100 mg/kg) e todas as subfrações testadas reduziram o peso do tecido granulomatoso formado em relação ao grupo controle (p < 0,05), semelhante à redução promovida no grupo dexametasona (p > 0,05). O extrato etanólico não reduziu a área de contração das feridas por excisão em ratos em relação ao grupo veículo (p > 0,05). A avaliação da toxicidade aguda revelou alterações em padrões hematológicos, hepáticos e renais nos animais tratados com uma única dose de 2000 mg/kg do extrato etanólico de Braúna. A citotoxicidade foi avaliada frente ao teste de Artemia salina e a dose média encontrada foi de 93 μg/ml, sendo considerado muito ativa. Desta maneira, pode-se concluir que as folhas de S. brasiliensis Engler possuem potencial terapêutico relevante caracterizado pela presença de compostos com atividade comprovada, bem como por possuir atividades antinociceptivas e anti-inflamatórias significativas, e por se apresentar segura nas doses terapêuticas avaliadas. Sendo assim, a folha de Braúna é uma importante fonte natural para identificação de novas moléculas e compostos com fins terapêuticos, podendo futuramente ser utilizada pela população de forma sustentável, ajudando na proteção ambiental da espécie.

  • Schinopsis brasiliensis Engler, popularly known as “Braúna”, is widely used in folk medicine to treat pain, fever, cough and other diseases. This study aimed to evaluate the antinociceptive, anti-inflammatory and healing activity of S. brasiliensis Engler from Bahia semi-arid, collected in the region of the National Forest Contendas do Sincorá (FLONA), according to ethnopharmacological data of the region. Previous tests carried out with the crude extract (ethanolic) and fractions have already proven the antinociceptive and anti-inflammatory action present in Brauna leaf. Thus, the aim of this study was to evaluate the antinociceptive and anti-inflammatory activity of S. brasiliensis leaf subfractions, to initiate the identification process of bioactive compounds, as well as to elucidate the possible mechanisms of action of these subfractions. The results were obtained through the collection and identification of plant species, phytochemical analysis, ethanolic extract preparation, fractions and subfractions, isolation and identification of compounds. The subfractions tested were hexane (H1, H2, H3, H4) and dichloromethane (D1, D2, D3, D4 and D5). To evaluate antinociceptive and anti-inflammatory activity, tests for abdominal acetic acid-induced writhing, leukocyte recruitment into the peritoneal cavity, evaluation of Evans blue peritoneal extravasation, nociception induced by formalin injection were performed. For chronic inflammation, the granuloma test induced by subcutaneous cotton pellet implantation was used. An experimental excisional wound model was performed to evaluate the healing activity of the leaf extract. For toxicological evaluation, acute toxicity tests were performed in Wistar rats and toxicity against Artemia salina (TAS). Statistical analysis was performed by one-way or two-way ANOVA, parametric approach, followed by Bonferroni's multiple comparison test, with p <0.05. The leaves of S. brasiliensis have polyphenols, tannins, saponins, flavonoids, alkaloids and steroids in both the crude extract and the studied fractions. Gas chromatographic analysis coupled with mass spectrophotometry identified 36 substances in the hexane and dichloromethane fraction, among them Ethyl Galate, a substance of high pharmacological interest. In the acetic acid induced abdominal writhing experiment, subfractions H2, H3, H4 and D1 (50 mg/kg) significantly reduced nociception (p <0.05). The ethanolic extract (100 mg/kg) presented antinociceptive activity with opioid action and inhibitory synthesis of nitric oxide (p <0.05). In the intraplantar formalin injection induced nociception test, subfractions H2, H3, H4, D1, D2, D3, D4 and D5 (50 mg/kg) reduced the number of formalin-induced flinches in phase 1 of the experiment, (p> 0.05) and in phase 2, the subfractions H3, H4, D1, D2, D4 and D5 significantly decreased nociception compared to the vehicle group (p <0.05). None of the subfractions decreased paw edema (p> 0.05). The subfractions D2, D3, D4 and D5 were able to increase the pain threshold of the animals in the Von Frey experiment (p <0.05). The subfractions H2, H3, H4, D1 and D5 (50 mg/kg) showed anti-inflammatory activity reducing the neutrophil migration to the peritoneal cavity (p <0.05), and the possible mechanism of anti-inflammatory action of the H3, H4, D3, D4 and D5 subfractions were due to decreased TNF-a cytokine in treated animals and elevation of IL-10 cytokine when animals were treated with D1 subfraction. There was no decrease in vascular permeability by subfractions evaluated in the Evans blue extravasation test (p> 0.05). Ethanolic extract (50 and 100 mg/kg) and all subfractions tested reduced the weight of granulomatous tissue formed in relation to the control group (p <0.05), similar to the reduction promoted in the dexamethasone group (p> 0.05). The ethanolic extract did not reduce the area of excision wound contraction in rats relative to the vehicle group (p> 0.05). Acute toxicity evaluation revealed changes in hematological, hepatic and renal patterns in animals treated with a single dose of 2000 mg/kg of Braúna ethanolic extract. The cytotoxicity was evaluated against the Artemia saline test and the average dose found was 93 μg/ml, being considered very active. Thus, it can be concluded that S. brasiliensis Engler leaves have relevant therapeutic potential characterized by the presence of compounds with proven activity, as well as having significant antinociceptive and anti-inflammatory activities, and for being safe in the evaluated therapeutic doses. Thus, Brauna leaf is an important natural source for the identification of new molecules and compounds for therapeutic purposes, and can be used by the population in a sustainable way, helping in the environmental protection of the species.

  • A Pimpinella anisum Lineu é utilizada para aumentar a produção de leite materno, porém de forma empírica por mães com hipogalactia, sendo que sua eficácia, segurança, dose efetiva e mecanismos de ação devem ser comprovados. Este estudo avaliou o perfil fitoquímico e a atividade galactagoga do extrato etanólico (EEPA), frações das sementes de P. anisum e do composto isolado anetol, bem como a toxicidade de EEPA. Ratas Wistar lactantes amamentando sete a oito filhotes cada, foram divididas em grupos tratadas diariamente por via intraperitoneal com (n=6): a) 0,4 ml de água destilada; b) EEPA nas doses de 12,5; 25 e 50 mg/Kg; c) FH nas doses de 12,5; 25 e 50 mg/Kg; d) FD nas doses de 12,5; 25 e 50 mg/Kg, e) FAE nas doses de 12,5; 25 e 50 mg/Kg; f) FB nas doses de 6,25; 12,5 e 25 mg/Kg e apenas o g) anetol nas doses de 12,5; 25 e 50 mg/Kg; por via oral.  Foram determinados o ganho de peso diário e total da ninhada, a estimativa da produção de leite em 1 h de amamentação, a perda de peso da rata lactante e o tempo de amamentação do 4º ao 18ª dia de vida dos filhotes. No 19º dia, as ratas foram ordenhadas para determinar o teor energético e os constituintes majoritários do leite, sendo eutanaziadas no 20º dia e removidas as glândulas mamárias para a análise histológica. No 2º experimento, as concentrações de prolactina e ocitocina foram dosadas na adenohipófise ou neurohipófise, soro e glândula mamária nas ratas no 14º e 21º dia de lactação, divididas nos seguintes grupos (n=6): a) 0,4 mL água destilada, b) EEPA (12,5 mg/Kg) e c) anetol (50 mg/Kg). Toxicidade frente à Artemia salina (12,5; 25; 50, 100 e 200 μg/mL) e toxicidade aguda com determinação de DL50 com ratos machos e fêmeas Wistar foram analisadas para os grupos (n: 3): a) etanol 10%, b) 5, 50, 300 e 2000 mg/Kg de EEPA e c) 0,4 ml água destilada. Atividade galactagoga foi observada apenas com o EEPA (12,5 mg/Kg), fração de diclorometano (25 mg/Kg) e anetol (50 mg/Kg), pois houve maior ganho de peso diário (24,8%, 11,6%, 10,2%) e total (28,1%, 11,37%, 9,7%), e estimativa de produção de leite (32,6%, 40%, 22,4%) (p < 0,05), sem alterações na perda de peso da rata lactante e no tempo de amamentação (p > 0,05) em relação ao controle. EEPA aumentou a concentração de proteínas totais e colesterol (84,7% e 185%), a FD aumentou a concentração de lactose, lipídios totais e colesterol (27,6%, 140,4% e 140%) e o anetol aumentou todos os constituintes em relação ao controle (p < 0,05). Embora as glândulas mamárias histologicamente estivessem em processo de involução, o menor número de lóbulos e alvéolos do EEPA e o menor número de alvéolos da FD e anetol indicam que estavam distendidos e a glândula estava mais ativa anteriormente. As maiores concentrações de prolactina adenohipofisária, sérica e na glândula mamária, aos 21 dias foram encontradas com EEPA e anetol (p < 0,05). Aumento da concentração de ocitocina ocorreu ao 14º dia na neurohipófise, e no soro do EEPA e apenas na neurohipófise do anetol. EEPA não apresentou toxicidade aguda nem morte dos animais. Assim, as sementes de P. anisum possuem atividade galactagoga atribuída a presença de compostos com atividade estrogênica (β-sitosterol, estigmasterol, anetol e seus polímeros) e antagonistas de dopamina (anetol) que estimulam a maior produção de prolactina, levando às alterações histológicas da glândula mamária, constituintes e quantidade de leite, refletindo no maior crescimento da ninhada.

  • Pimpinella anisum Lineu is used to increase the production of breast milk, but is empirically used by mothers with hypogalactia, and its effectiveness, safety, effective dose and mechanisms of action must be proven. This study evaluated the phytochemical profile and galactagogue activity of the ethanolic extract (EEPA), fractions of the seeds of P. anisum and the isolated compound anethole, as well as the toxicity of the EEPA. Lactating Wistar rats breastfeeding seven to eight puppies each, were divided into groups treated daily intraperitoneally with (n = 6): a) 0.4 ml of distilled water; b) EEPA in doses of 12.5; 25 and 50 mg/kg; c) FH doses at 12.5; 25 and 50 mg/kg; d) FD in doses of 12.5; 25 and 50 mg/Kg, e) FAE at doses of 12.5; 25 and 50 mg/kg; f) FB at doses of 6.25; 12.5 and 25 mg/kg; and only g) anethole in doses of 12.5; 25 and 50 mg/kg; orally. The daily and total weight gain of the litter was determined, the estimated milk production in 1 h of breastfeeding, the weight loss of the lactating rat and the breastfeeding time from the 4th to the 18th day of life of the puppies. On the 19th day, rats were milked to determine the energy content and the major constituents of milk, being euthanized on the 20th day and removed the mammary glands for histological analysis. In the 2nd experiment, the prolactin and oxytocin diets were measured in the adenohypophysis or neurohypophysis, serum and mammary gland in the rats on the 14th and 21st day of lactation, divided into the following groups (n = 6): a) 0.4 mL distilled water, b) EEPA (12.5 mg/kg) and c) anethole (50 mg/kg). Toxicity to Artemia salina (12.5; 25; 50, 100 and 200 μg/mL) and acute toxicity with determination of LD50 with male and specific Wistar rats were analyzed for groups (n: 3): a) ethanol 10%, b) 5, 50, 300 and 2000 mg/kg of EEPA; and c) 0.4 ml distilled water. Galactagogue activity was observed only with EEPA (12.5 mg/kg), fraction of dichloromethane (25 mg/kg) and anethole (50 mg/kg), as there was greater daily weight gain (24.8%, 11, 6%, 10.2%) and total (28.1%, 11.37%, 9.7%), and estimated milk production (32.6%, 40%, 22.4%) (p < 0.05), without changes in the weight loss of the lactating rat and without breastfeeding time (p> 0.05) in relation to the control. EEPA increased the concentration of total proteins and cholesterol (84.7% and 185%), FD increased the concentration of lactose, total lipids and cholesterol (27.6%, 140.4% and 140%) and anethole increased all constituents in relation to the control (p <0.05). Although the mammary glands were histologically in process of involution, the smaller number of lobes and alveoli of the EEPA and the smaller number of alveoli of the FD and anethole indicate that they were distended and the gland was more active previously. The highest levels of adenohypophyseal, serum and mammary gland prolactin at 21 days were found with EEPA and anethole (p <0.05). Increased oxytocin concentration occurred on the 14th day in the neurohypophysis, and in the EEPA serum and only in the anethole neurohypophysis. EEPA has no acute toxicity or animal death. Thus, as seeds of P. anisum have galactagogue activity attributed to the presence of compounds with estrogenic activity (β-sitosterol, stigmasterol, anethole and their polymers) and dopamine antagonists (anethole) that stimulate a greater production of prolactin, when it comes to histological changes of the mammary gland, constituents and quantity of milk, reflecting the greater growth of the litter.

  • Introdução/Objetivos: Nas mulheres, o envelhecimento é marcado por alterações hormonais fisiológicas que acarretam alterações na composição corporal, desenvolvimento de inflamação crônica de baixo grau e maior predisposição ao desenvolvimento de algumas doenças crônicas como a Hipertensão Arterial Sistêmica (HAS) e o Diabetes Mellitus tipo 2 (DMII). Embora sejam distúrbios com forte influência pró-inflamatória, pouco se sabe sobre como a composição corporal interfere nos marcadores inflamatórios em idosas diabéticas e hipertensas. Portanto, o objetivo deste estudo foi avaliar a inflamação em mulheres idosas diabéticas e hipertensas e sua associação com a massa gorda. Materiais e métodos: Este estudo consistiu na avaliação de 144 mulheres com idade entre 60 e 80 anos, com diagnóstico de HAS e DMII, assistidas por Unidades de Saúde da Família em Vitória da Conquista, Bahia, Brasil. De acordo com o índice de massa corporal (IMC), essas mulheres foram classificadas como eutróficas ou com sobrepeso. Em seguida, foram realizadas avaliações antropométricas de diferentes marcadores, além de coleta de sangue periférico para a mensuração das concentrações séricas dos marcadores bioquímicos e citocinas. Por fim, comparou-se os grupos através do teste não paramétrico de Mann-Whitney e realizou-se o teste de correlação de Spearman, através do pacote de análises Corrplot do software estatístico R, avaliando-se de forma ampliada a influência da composição corporal sobre os fatores analisados.  Resultados: Nas mulheres idosas eutróficas, a relação IL-10/IL-17A foi aumentada quando comparada àquelas com sobrepeso. Além disso, a IL-17 correlacionou-se negativamente com o HDL e positivamente com o colesterol total, LDL, triglicérides, VLDL e glicose nas idosas em eutrofia, mas não nas idosas com excesso de peso. A IL-10 também apresentou diferenças no padrão de correlação observado entre os grupos, pois nas eutróficas esta citocina apresentou correlação positiva com a circunferência da cintura, relação cintura/estatura e associação negativa com IL-17, enquanto em mulheres idosas com excesso de peso, não foi observado tal padrão. Conclusões: Assim, este trabalho demonstra como a massa gorda interfere no balanço de citocinas, principalmente IL-10 e IL-17, e como ela pode exercer um efeito sobre parâmetros bioquímicos e fatores de risco antropométrico associados ao DMII e à HAS.

  • Introduction/Objective: In women, aging is evidenced by physiological hormonal alterations that trigger changes in body composition, emergence of chronic low-grade inflammation, which is an important pre-disposition to the development of chronic diseases such as Systemic Arterial Hypertension (SAH) and Type 2 Diabetes Mellitus (T2DM). Although it is a strong pro-inflammatory nature disorder, little is known about how body composition interferes with inflammatory markers in diabetic and hypertensive elderly women. Therefore, this study’s objective was to evaluate the inflammation in diabetic and hypertensive elderly women, and their association with fat mass. Methods: It comprised the evaluation of 144 women aged between 60 and 80 years old, diagnosed with SAH and T2DM, assisted by Family Health Units in Vitória da Conquista, Bahia, Brazil. According to body mass index (BMI), these women were classified as either eutrophic or overweight. Then, anthropometric evaluations of different markers were performed, as well as peripheral blood collection for the measurement of serum concentrations of biochemical markers and cytokines. Finally, the groups were compared through the Mann-Whitney non-parametric test and the Spearman correlation test was performed through the Corrplot analysis package of the statistical software R, with an increased evaluation of the influence of body composition on the factors analyzed. Results: In the eutrophic females, the IL-10/IL-17A ratio was increased when compared to those overweight. In addition, IL-17 correlated negatively with HDL and positively with total cholesterol, LDL, triglycerides, VLDL, and glucose in the eutrophic ones, but not in the overweight individuals. IL-10 also showed differences in the correlation pattern observed between the groups, since for this eutrophic cytokine, it presented positive correlation with the waist circumference, waist/height ratio, and negative association with IL-17, while in overweight elderly women, it was not observed such pattern. Conclusion: Hence, this work demonstrates how the amount of fat mass interferes with the balance of cytokines, especially IL-10 and IL-17, and how it may exert an effect on biochemical parameters and risk factors associated with T2DM and SAH.

  • INTRODUÇÃO/OBJETIVO: Hipertensão arterial sistêmica (HAS) e diabetes mellitus do tipo 2 (DMT2) são importantes doenças crônicas não transmissíveis (DCNT) responsáveis por elevada mortalidade associada ao risco cardiometabólico. O envelhecimento é marcado pelo amplo espectro de modificações fisiológicas que configuram os idosos como grupo de risco para o desenvolvimento de DCNT. A terapia medicamentosa da HAS e DMT2 frequentemente é acompanhada pela prática de atividade física, e esta abordagem é associada à redução do risco cardiometabólico. Entretanto, pouco se sabe sobre a existência de perfis de benefícios terapêuticos distintos entre homens idosos e adultos hipertensos e diabéticos praticantes de atividade física. Com isso, este trabalho teve por objetivo comparar a influência da pratica atividade física entre homens adultos ou idosos, hipertensos e diabéticos, sobre parâmetros bioquímicos, antropométricos e inflamatórios relacionados a patogênese da HAS e DMT2. MÉTODOS: Neste estudo, 94 homens portadores de HAS e DMT2 foram incluídos. O trabalho contou com a aplicação de uma versão reduzida e validada do questionário internacional de atividade física (IPAQ) e do questionário de avaliação de qualidade de vida e saúde (QVS-80). Os indivíduos praticantes de atividade física foram selecionados pela prática de pelo menos 150 minutos de atividade física por semana por um período de pelo menos três meses. Indivíduos com idade igual ou superior a 60 anos foram classificados como idosos. Em seguida, foi realizada a avaliação antropométrica dos participantes, onde foram coletados dados de massa corpórea, composição corpórea, altura e circunferência de cintura. Após a sugestão de jejum de pelo menos 8h, uma amostra de sangue foi coletada de cada participante para dosagem sérica de marcadores bioquímicos (HDL, LDL, VLDL, triglicérides e glicose), inflamatórios (IL-1β, IL-17, IFN-γ, TNF-α e IL-10) e hormônios sexuais (testosterona total e estradiol. RESULTADOS: Homens adultos e idosos praticantes de atividade física apresentaram mais massas magra e óssea, bem como menor massa gorda do que indivíduos sedentários. A massa magra participou de um eixo de múltiplas correlações negativas com marcadores antropométricos, correlacionando-se negativamente com a razão entre a circunferência de cintura e estatura, massa gorda e índice de massa corporal. Em idosos praticantes de atividade física, a massa magra ainda se correlacionou negativamente com marcadores inflamatórios sistêmicos e positivamente com a IL-10 e com o estradiol sérico. Homens adultos e idosos praticantes de atividade física também apresentaram menores concentrações séricas de LDL, VLDL, triglicérides e de glicose em jejum do que indivíduos sedentários. Os idosos praticantes de atividade física exibiram menores concentrações séricas de IL-1β e TNF-α do que indivíduos de adultos sedentários, bem como menores valores para as concentrações séricas de IL-17, IL-1β, TNF-α e IFN-γ do que a sua contraparte sedentária. Neste estudo não detectamos diferenças entre as concentrações séricas de testosterona e estradiol entre os grupos. CONCLUSÃO: A prática de atividade física mostrou-se benéfica tanto para homens adultos como para homens idosos diabéticos e hipertensos, configurando, nestes, um menor risco cardiovascular associado à patogênese destas doenças. No entanto, os nossos resultados sugerem que, para homens portadores de HAS e DMT2, a prática de atividade física pode ser mais valiosa do que a idade na modulação de marcadores fisiopatológicos destas comorbidades, principalmente por participar da regulação de das concentrações séricas de marcadores da inflamação sistêmica relacionados ao risco cardiometabólico.

  • INTRODUCTION / OBJECTIVE: Systemic arterial hypertension (SAH) and type 2 diabetes mellitus (T2DM) are two important chronic noncommunicable diseases (NCDs) responsible for a high worldwide mortality associated with a high cardiometabolic risk. Aging is marked by a broad spectrum of physiological changes that configure the elderly as a risk group for the development of NCDs. Drug therapy for SAH and DMT2 is often accompanied by physical activity, and this approach is associated with the reduction of cardiometabolic risk associated with these comorbidities. However, little is known about the existence of distinct therapeutic benefit profiles of physical activity among elderly and middle-aged hypertensive and diabetic men. Therefore, the objective of this study was to compare pathophysiological markers of SAH and DMT2 among middle-aged and elderly individuals, while physical activity practitioners or sedentary. METHODS: In this study, 94 men with SAH and DMT2 were included. The work involved the application of the International Physical Activity Questionnaire (IPAQ) and the Quality of Life and Health Assessment Questionnaire (QVS-80). A blood sample was collected from the participants and an anthropometric evaluation was performed. the practice of physical activity was defined by at least 150 minutes of physical activity per week for a period of at least three months. Individuals older than or equal to 60 years were classified as elderly. Blood samples were submitted to biochemical (HDL, LDL, VLDL, triglyceride and fasting glucose), hormonal (total testosterone and estradiol) and immunological analysis (IL-1β, IL-17, IFN-γ, TNF-α and IL-10). RESULTS: Physical activity practitioners presented higher lean mass and bone mass, as well as lower fat mass than sedentary individuals. The lean mass participated in an axis of multiple negative correlations with pathological markers of SAH and DMT2, correlating negatively with the ratio between waist circumference and height, fat mass, and body mass index. In physically active elderly, lean mass still correlated negatively with systemic inflammatory markers and positively with IL-10 and serum estradiol. Physical activity subjects also had lower serum concentrations of LDL, VLDL, triglycerides and fasting glucose. Physically active elderly exhibited lower serum concentrations of IL-1β and TNF-α than sedentary middle-aged men, as well as lower serum concentrations of IL-17, IL-1β, TNF-α and IFN-γ than its sedentary counterpart. In this study we detected differences between the serum concentrations of testosterone and estradiol between the groups. CONCLUSION: Together, our results suggest that, for men with SAH and DMT2, the practice of physical activity is more valuable than the aging phenom in the modulation of pathophysiological markers of these SAH and DMT2, mainly because it participates in the regulation of the systemic inflammation observed in these comorbidities. However, the practice of physical activity has shown to be beneficial for both middle-aged and elderly diabetic and hypertensive men, being associated with have a lower cardiovascular risk in the pathogenesis of these diseases.

  • Objetivo: Investigar os efeitos do treinamento físico sobre alterações no desempenho físico, capacidade antioxidante e expressão gênica de marcadores inflamatórios no músculo esquelético de camundongos expostos à fumaça do cigarro (FC). Métodos: Foram utilizados 32 camundongos C57/BL6 machos com oito semanas de idade divididos aleatoriamente em quatro grupos (n = 8 / grupo), que foram submetidos ou não à FC e exercício físico: sedentário (SED), SED+FC, treinado (TRE) e TRE+FC. Todos os animais foram submetidos ao teste progressivo máximo de natação para avaliação da capacidade aeróbica e ao teste de tela invertida para avaliação da resistência muscular. A massa corporal total (MCT) e o consumo alimentar foram verificados semanalmente ao longo de todo o experimento. Após a eutanásia, os depósitos de tecido adiposo visceral (TAV) e os músculos gastrocnêmios foram coletados e pesados. O TAV foi utilizado para calcular o índice de adiposidade e os músculos para ensaios de atividade da glutationa peroxidase e catalase e expressão dos genes que codificam marcadores proteicos de inflamação. Resultados: O consumo alimentar aumentou entre as medidas iniciais (1ª a 4ª semana) e finais (5ª a 8ª semana) em todos os grupos. A comparação entre os grupos indicou que os camundongos expostos à fumaça do cigarro apresentaram consumo alimentar menor, quando comparados ao grupo SED, não havendo diferença estatística em relação ao grupo TRE. Houve redução da MCT entre as medidas iniciais e finais apenas nos grupos expostos a fumaça do cigarro (SED+FC e TRE+FC). A comparação da MCT entre grupos não mostrou diferença significativa entre estes. O peso da carcaça e o índice de adiposidade foram menores no grupo SED+FC, em relação aos demais grupos. O desempenho no teste de nado forçado melhorou entre as medidas iniciais e finais em todos os grupos. A comparação entre os grupos mostrou que o grupo TRE apresentou melhor desempenho quando comparado aos demais grupos; de forma oposta, o grupo SED+FC apresentou pior desempenho quando comparado aos demais grupos, enquanto os grupos SED e TRE+FC não apresentaram diferença estatística entre si. O desempenho no teste da tela invertida melhorou entre as medidas iniciais e finais apenas no grupo TRE. A comparação entre os grupos mostrou que o grupo TRE apresentou melhor desempenho quando comparado aos demais grupos, enquanto os grupos SED, SED+FC e TRE+FC não apresentaram diferença estatística entre si. A atividade da enzima catalase foi estatisticamente maior no grupo TRE em relação aos demais grupos. Já para a enzima GPx, não foi observada diferença significativa entre os grupos. A análise de expressão gênica indicou que, comparado aos demais grupos, os animais do grupo SED+FC expressaram mais TNFα e MCP-1 e menos HO-1. A expressão do gene da citocina IL-10 foi maior no grupo TRE, quando comparado aos grupos SED e SED+FC, enquanto a do gene TGF-β foi maior no grupo SED+FC, quando comparado ao grupo TRE. Não houve diferença estatística entre os grupos na expressão dos genes relacionados às citocinas IL-1β e IL-6. Conclusão: Quatro semanas de exposição à fumaça induziram à redução do consumo alimentar, acompanhada de maior comprometimento da massa e composição corporal, além de redução do desempenho físico e alteração na expressão de genes associados a manutenção ou controle da inflamação. Em contrapartida, a prática regular de exercício físico protegeu os animais expostos à fumaça contra alterações na composição corporal, desempenho físico e expressão dos genes TNF-α, MCP-1 e HO-1.

  • Objective: To investigate the effects of physical training on changes in physical performance, antioxidant capacity and gene expression of inflammatory markers in skeletal muscle of mice exposed to cigarette smoke (CS). Methods: Thirty-eight eightweek-old male C57 / BL6 mice were randomly divided into four groups (n = 8 / group), submitted or not to CS and physical exercise: sedentary (SED), SED + CS, trained (TRE). and TRE + CS. All animals underwent the maximum progressive swimming test to assess aerobic capacity and inverted screen test for muscle endurance assessment. Total body mass (TBM) and food intake were verified weekly throughout the experiment. After euthanasia, visceral adipose tissue (TAV) deposits and gastrocnemius muscles were collected and weighed. TAV was used to calculate adiposity index and muscle were for glutathione peroxidase and catalase activity assays and expression of genes encoding protein markers of inflammation. Results: Food intake increased between the initial (1st to 4th week) and final (5th to 8th week) measurements in all groups. The comparison between the groups indicated that the mice exposed to cigarette smoke had lower food consumption when compared to the SED group, with no statistical difference compared to the TRE group. There was a reduction in TBM between the initial and final measurements only in the groups exposed to cigarette smoke (SED + CS and TRE + CS). Comparison of TBM between groups showed no significant difference between them. Carcass weight and adiposity index were lower in the SED + CS group compared to the other groups. Forced swim test performance improved between initial and final measurements in all groups. The comparison between the groups showed that the TRE group presented better performance when compared to the other groups; On the other hand, the SED + CS group presented worse performance when compared to the other groups, while the SED and TRE + CS groups did not present statistical difference. Performance in the inverted screen test improved between initial and final measurements in the TRE group only. The comparison between the groups showed that the TRE group presented better performance when compared to the other groups, while the SED, SED + CS and TRE + CS groups did not present statistical difference. Catalase enzyme activity was statistically higher in the TRE group compared to the other groups. For GPx enzyme, no significant difference was observed between the groups. Gene expression analysis indicated that, compared to the other groups, animals from the SED + CS group expressed more TNF-α and MCP-1 and less HO-1. The expression of the IL-10 cytokine gene was higher in the TRE group when compared to the SED and SED + CS groups, while the TGF-β gene expression was higher in the SED + CS group when compared to the TRE group. There was no statistical difference between groups in the expression of IL-1β and IL-6 cytokine-related genes. Conclusion: Four weeks of exposure to cigarette smoke induced a reduction in food intake, accompanied by greater impairment of body mass and composition, as well as reduced physical performance and altered gene expression associated with maintaining or controlling inflammation. In contrast, regular exercise protected animals exposed to smoke from changes in body composition, physical performance, and expression of the TNF-α, MCP-1, and HO-1 genes.

  • Evidências clínicas e experimentais apoiam a hipótese de que os esteroides sexuais regulam a resposta imunológica e, consequentemente, exercem efeitos sobre condições patológicas adversas, como as infecções bacterianas. De um modo geral, o paradigma da influência hormonal sobre a resposta imunológica estipula que o estrógeno aumenta essa resposta, enquanto que a testosterona suprime. Contudo, dados na literatura ainda são controversos. Ainda, há uma lacuna no que diz respeito ao papel do estrógeno nas infecções por bactérias Gram-positivas, como S. aureus. O objetivo deste estudo foi avaliar a influência da ovariectomia e do 17β-estradiol na resposta imunológica induzida por S. aureus em um modelo in vitro de macrófagos peritoneais murinos (MPMs) e monócitos humanos do sangue periférico (hPMs) e em um modelo in vivo. Para isso, camundongos fêmeas BALB/c foram ovariectomizadas (OVX) ou sham-operadas (Sham). No modelo animal, as fêmeas foram inoculadas intraperitonealmente com S. aureus ATCC 25923 ou salina estéril. Os animais foram eutanasiados em 7 períodos diferentes com intervalo de 24 horas cada e 10 fêmeas (n = 5 Sham/n = 5 OVX) foram agrupadas para cada período. Após a eutanásia, amostras de sangue foram coletadas para contagem total e diferencial de leucócitos e quantificação de S. aureus por RT-PCR. O útero e o baço foram removidos e pesados. O soro foi destinado para avaliação dos níveis de estradiol. Os pulmões foram removidos e fracionados para realização da análise imuno-histoquímica para detecção de macrófagos (anti-CD68) e expressão gênica relativa de IL-6, IL-1β e TNF-α por RT-PCR. Os resultados mostraram que fêmeas OVX apresentaram esplenomegalia e resposta tardia de monócitos em comparação com às shams. A inoculação intraperitoneal causou bacteremia em ambos os grupos, e as fêmeas OVX apresentaram um recrutamento pulmonar tardio de macrófagos. Além disso, as fêmeas OVX apresentaram maior expressão gênica de IL-1β, TNF-α e IL-6. A maior expressão de IL-6 em comparação às fêmeas sham foi observada mesmo na ausência da infecção. No modelo in vitro foi realizado o isolamento de macrófagos peritoneais (2 x 105 ), de camundongos fêmeas shams (n=3) e OVX (n=3) e machos (n=3), induzidos por tioglicolato e que foram inoculados com S. aureus por 6 horas. Os macrófagos obtidos das fêmeas OVX e dos machos foram previamente tratados por 24 horas com 17β-estradiol (E2) (10-7 M). Os macrófagos foram coletados e destinados para avaliação da expressão gênica relativa de TNF-α, IL-1β, IL-6, IL8 e TLR2. Os resultados mostraram que o tratamento com E2 diminuiu a expressão gênica das citocinas, com exceção de IL-8, em que os MPMs de machos apresentaram um aumento da expressão. Ainda, E2 diminui a expressão gênica do receptor TLR2. No modelo in vitro de hPMs, foram selecionados seis homens e seis mulheres e os hPMs foram isolados a partir de amostras do sangue periférico dos voluntários. Nas mulheres, o sangue foi coletado tanto no período menstrual quanto no período fértil. Após 24h em estufa de CO2, os hPMs foram inoculados por S. aureus por 6 horas. Após esse período, o sobrenadante foi coletado para análise de citocinas por Luminex e os hPMs removidos para análise de 84 genes envolvidos na resposta do hospedeiro a infecções bacterianas por RT-PCR array. Observou-se que, em comparação aos hPMs masculinos, os hPMs masculinos tratados com E2 produziram menos TNF-α, IL-1 e IL-6 e mais IL-23, IL-27 e GM-CSF, e os hPMs de mulheres no período fértil produziram menos TNF-α, IL-1 e produziram mais IL-10, IL-12, IL-23 e IL-27. Em comparação aos hPMs de mulheres no período menstrual, os hPMs de mulheres no período fértil produziram mais IL-12. hPMs masculinos tratados com E2 e hPMs de mulheres no período fértil expressaram menos TRL2 que hPMs masculinos. De acordo a análise da expressão gênica foi possível inferir que o E2 inibiu a via do NFκB. Com isso, conclui-se que o estrógeno atua como um imunoprotetor por modular uma ação anti-inflamatória na resposta imunológica induzida por S. aureus.

  • Clinical and experimental evidence supports the hypothesis that sex steroids regulate the immune response and thus exert effects on adverse pathological conditions such as bacterial infections. In general, the hormonal influence paradigm on the immune response stipulates that estrogen increases this response, while testosterone suppresses it. However, data in the literature are still controversial. Still, there is a gap regarding the role of estrogen in infections with Gram-positive bacteria such as S. aureus. The aim of this study was to evaluate the influence of ovariectomy and 17β-estradiol on S. aureus-induced immune response in an in vitro model of murine peritoneal macrophages (MPMs) and human peripheral blood monocytes (hPMs) and in an in vivo model. For this, female BALB / c mice were ovariectomized (OVX) or sham operated (Sham). In the animal model, females were inoculated intraperitoneally with S. aureus ATCC 25923 or sterile saline. The animals were euthanized at 7 different periods with 24-hour interval each and 10 females (n = 5 Sham / n = 5 OVX) were grouped for each period. After euthanasia, blood samples were collected for total and differential leukocyte count and quantification of S. aureus by RT-PCR. The uterus and spleen were removed and weighed. The serum was intended for evaluation of estradiol levels. The lungs were removed and fractionated for immunohistochemical analysis for macrophage detection (anti-CD68) and relative gene expression of IL-6, IL-1β and TNF-α by RT-PCR. The results showed that OVX females had splenomegaly and late monocyte response compared to shams. Intraperitoneal inoculation caused bacteremia in both groups, and OVX females presented late pulmonary recruitment of macrophages. In addition, OVX females showed higher gene expression of IL-1β, TNF-α and IL-6. Higher IL-6 expression compared to sham females was observed even in the absence of infection. In the in vitro model, thioglycolate-induced peritoneal macrophages (2 x 105 ) of female shams (n = 3), OVX (n = 3) and male (n = 3) mice were inoculated with S. aureus for 6 hours. Macrophages obtained from OVX females and males were previously treated for 24 hours with 17βestradiol (E2) (10-7 M). The macrophages were collected and destined to evaluate the relative gene expression of TNF-α, IL-1β, IL-6, IL-8 and TLR2. Results showed that E2 treatment decreased cytokines gene expression, except for IL-8, in which male MPMs showed increased expression. Also, E2 decreases TLR2 receptor gene expression. In the in vitro model of hPMs, six men and six women were selected and hPMs were isolated from peripheral blood samples of the volunteers. In women, blood was collected in both menstrual and fertile periods. After 24h in a CO2 oven, the hPMs were inoculated by S. aureus for 6 hours. After this period, the supernatant was collected for Luminex cytokine analysis and the hPMs removed for analysis of 84 genes involved in host response to bacterial infections by RT-PCR array. Compared to male hPMs, it was observed that E2-treated male hPMs produced less TNF-α, IL-1 and IL-6 and more IL-23, IL-27 and GM-CSF, and women's hPMs in the fertile period produced less TNF-α, IL-1 and produced more IL-10, IL-12, IL-23 and IL-27. Compared to hPMs from women in menstrual period, the hPMs from women in the fertile period produced more IL-12. Male hPMs treated with E2 and hPMs of women in the fertile period expressed less TRL2 than male hPMs. According to the analysis of gene expression it was possible to infer that E2 inhibited the NFκB pathway. It is concluded that estrogen acts as an immunoprotectant by modulating an antiinflammatory action on the immune response induced by S. aureus. Keywords: Sexual dimorphism. 17β-estradiol.

  • Infecções bacterianas são um problema de saúde pública devido a grande variedade de microrganismos patogênicos. Entre estes, Staphylococcus aureus é o principal agente causador de infecções bacterianas no mundo e diversas cepas deste agente já são capazes de criar resistência a terapia medicamentosa convencional.  Nesse contexto, a terapia fotodinâmica antimicrobiana (antimicrobial photodynamic therapy; aPDT) aparece como uma ferramenta promissora por meio da inativação microbiana com o uso da luz. Em geral, a aPDT é aplicada em tratamentos envolvendo fotossensibilizadores capazes de gerar radicais livres de oxigênio que vão levar a depuração bacteriana. Assim, este trabalho propõe de maneira pioneira o uso do Resveratrol e do extrato de Myrciaria cauliflora como fotossensibilizadores na terapia antimicrobiana contra S. aureus. Métodos: Testes in vitro foram realizados para determinar a atividade antibacteriana dos fotossensibilizadores fotoativados com luz LED azul, bem como experimentos com ácido úrico para verificação da formação de oxigênio singlete . Possíveis alterações estruturais do Resveratrol foram avaliadas por HPLC. Nos ensaios in vivo, o modelo de bolsão de ar foi realizado em camundongos C57Bl/6 para avaliar a atividade antimicrobiana do Resveratrol fotoativado, além da avaliação da migração celular por técnicas histológicas e produção de citocinas pela técnica de ELISA. Em outra análise, um modelo de infecção intradérmica, na orelha esquerda foi realizado para avaliação dos dois fotossensibilizadores, separadamente, em camundongos Balb/c. Após a infecção e eutanásia, a orelha foi coletada e utilizada a técnica de imunohistoquímica para a marcação de Mieloperoxidase (MPO) e E-caderina. O linfonodo dos animais foi retirado para avaliação da carga bacteriana e produção de citocinas. Resultados e discussão: O Resveratrol fotoativado exibiu um aumento na marcação para MPO sendo sua ação antibacteriana, possivelmente causada pela formação de oxigênio singlete . No modelo de bolsão de ar, foram produzidas citocinas TNF-α e IL-17A, diminuindo a carga bacteriana e, consequentemente, diminuindo inflamação após 24 horas de infecção. A diminuição do número de celulas no ambiente inflamatório foi associada à resolução da inflamação, juntamente com maior produção de IL-10. Além disso, foi observado na derme da orelha dos animais, que o Resveratrol fotoativado promoveu um aumento produção de MPO com redução da carga bacteriana no linfonodo drenante. Em relação a Myrciaria cauliflora foi observada a  redução na carga bacteriana nos testes in vitro, com aumento da produção de TNF-α, 17A no linfonodo drenate e MPO no sítio indradérmico de infecção. Conclusão: A partir dos dados analisados, é possível predizer que o Resveratrol e o extrato de Myrciaria cauliflora  são fotossensibilizadores promissores para a terapia fotodinâmica antimicrobiana, sobretudo para o controle da carga bacteriana em infecções intradérmicas causadas por S. aureus.

  • Bacterial infections are a public health problem due to the wide variety of pathogenic microorganisms. Among these, Staphylococcus aureus is the main causative agent of bacterial infections in the world and several strains of this agent are already able to resist conventional drug therapy. In this context, antimicrobial photodynamic therapy (aPDT) appears as a promising tool through microbial inactivation with the use of light. In general, aPDT is applied in treatments involving photosensitizers capable of generating oxygen free radicals that will lead to bacterial clearance. Thus, this work has pioneered the use of Resveratrol and Myrciaria cauliflora extract as photosensitizers in antimicrobial therapy against S. aureus. Methods: In vitro tests were performed to determine the antibacterial activity of blue LED light-activated photosensitizers, as well as experiments with uric acid to verify singlet oxygen formation. Possible structural changes of Resveratrol were evaluated by HPLC. In in vivo assays, the air pocket model was performed in C57Bl/6 mice to evaluate the antimicrobial activity of photoactivated Resveratrol, as well as to evaluate cell migration by histological techniques and cytokine production by ELISA. In another analysis, a model of intradermal infection in the left ear was performed to evaluate the two photosensitizers separately in Balb/c mice. After infection and euthanasia, the ear was collected and the immunohistochemistry technique was used to mark Myeloperoxidase (MPO) and E-cadherin. The lymph node of the animals was removed to evaluate the bacterial load and cytokine production. Results and Discussion: Photoactivated Resveratrol exhibited an increase in MPO labeling and its antibacterial action, possibly caused by singlet oxygen formation. In the air pocket model, TNF-α and IL-17A cytokines were produced, decreasing bacterial load and, consequently, decreasing inflammation after 24 hours of infection. Decreased number of cells in the inflammatory environment was associated with resolution of inflammation along with higher IL-10 production. In addition, it was observed in the ear dermis that photoactivated Resveratrol promoted an increase in MPO production with reduction of bacterial load in the draining lymph node. Regarding Myrciaria cauliflora, a reduction in bacterial load was observed in in vitro tests, with increased production of TNF-α, 17A in the lymph node drainage and MPO in the indradermal site of infection. Conclusion: From the analyzed data, it is possible to predict that Resveratrol and Myrciaria cauliflora extract are promising photosensitizers for antimicrobial photodynamic therapy, especially for the control of bacterial load in intradermal infections caused by S. aureus.

  • Investigar a influência dos parâmetros: intensidade e frequência semanal durante 8 semanas de exercício físico aeróbico, sobre as adaptações da defesa antioxidante e alterações funcionais e estruturais nos rins de camundongos em um modelo de Injúria Renal Aguda induzida pela cisplatina. Foram utilizados 40 camundongos Swiss machos com quatro meses de idade (16 semanas) que foram randomicamente distribuídos em cinco grupos experimentais (n = 8/grupo): 1.Sedentário, 2. (BI_3X) Treinado em baixa intensidade 3 vezes por semana, 3. (AI_3X) Treinado em alta intensidade 3 vezes por semana, 3. (BI_5X) Treinado em baixa intensidade 5 vezes por semana e (AI_5X) Treinado em alta intensidade 5 vezes por semana. Praticaram exercício físico, nado forçado, por 8 semanas e ao término do período de treinamento, foi administrado antineoplásico Cisplatina em todos os grupos experimentais para indução da Injúria Renal Aguda. Após 96 horas da administração os animais foram eutanasiados e os rins, músculos e tecidos adiposos viscerais retirados para posterior análise. Avaliou-se parâmetros de composição corporal, estrutura e função renal, marcadores de peroxidação lipídica e atividade de enzimas antioxidantes, bem como a expressão de genes envolvidos nas vias de balanço redox, inflamatória e apoptóticas. Os resultados obtidos revelaram que a frequência semanal de 3 vezes por semana promoveu melhores resultados com relação a melhora da estrutura renal, do balanço redox e dos processos apoptóticos. Observou-se que o grupo treinado em frequência 3 vezes por semana, independente da intensidade, apresentou maior massa de tecido adiposo marrom quando comparado aos demais grupos experimentais. Estruturalmente houve melhores resultados relacionados a essa mesma frequência com menor porcentagem de túbulos necróticos o que pode ter refletido numa melhora funcional com menor concentração sérica de ureia. Não houve diferença significativa na concentração sérica de creatinina entre os grupos experimentais. A peroxidação lipídica foi maior no grupo sedentário quando comparados aos grupos exercitados, independentemente da frequência e intensidade. Entretanto a atividade da catalase foi maior em todos os grupos exercitados em comparação ao grupo sedentário. O grupo treinado BI_3X apresentou
    maior atividade da GPx e menor expressão gênica do NFkB e Caspase-3. Não houve diferenças significativas na expressão gênica do Nrf2 em nenhum dos grupos experimentais. Nossos dados demonstram que a frequência semanal de treinamento foi o parâmetro de maior influência sobre o balanço redox e apoptose e inflamação, na injúria renal aguda induzida pela cisplatina. O resultados indicam que o aumento da capacidade antioxidante no tecido renal atenuam o processo inflamatório e vias de sinalização de morte celular, eventos estes que constituem a fisiopatologia da injúria
    renal aguda induzida pela cisplatina.

  • To investigate the influence of the parameters: intensity and weekly frequency during 8 weeks of aerobic exercise, on the adaptations of the antioxidant defense and functional and structural alterations in the kidneys of mice in a model of acute renal injury induced by cisplatin. Fourty old male Swiss mice at four months of age (16 weeks) were randomly assigned to five experimental groups (n = 8 / group): 1.
    Sedentary, 2. (LI_3X) low intensity training 3 times per week, 3. (HI_3X) high intensity training 3 times per week, 4. (LI_5X) low intensity training 5 times per week and 5. (HI_5X) high intensity training 5 times per week. They practiced forced swimming for 8 weeks and at the end of the training period the antineoplastic Cisplatin was administered in all the experimental groups for induction of acute renal injury. After 96 hours of administration the animals were euthanized and the kidneys, muscles and visceral adipose tissues were removed for further analysis. Body composition parameters, renal structure and function, lipid peroxidation markers and antioxidant enzymes activity, as well as the expression of genes involved in the redox, inflammatory and apoptotic balance pathways were evaluated. The results showed that the weekly frequency of 3 times per week promoted better results in relation to the improvement of renal structure, redox balance and apoptotic processes. It was observed that the group trained in the frequency of 3 times per week, regardless of the intensity, had a greater mass of brown adipose tissue when compared to the other experimental groups. Structurally there were better results related to this same
    frequency with a lower percentage of necrotic tubules, which may have reflected a functional improvement with lower serum urea concentration. There was no significant difference in serum creatinine concentration between the experimental groups. The lipid peroxidation was higher in the sedentary group when compared to the exercised groups, regardless of frequency and intensity. However, catalase activity was higher in all groups compared to the sedentary group. The trained group BI_3X presented higher GPx activity and lower gene expression of NFkB and Caspase-3. There were no significant differences in the gene expression of Nrf2 in any of the experimental groups. Our data demonstrate that weekly training frequency was the parameter of greatest influence on redox balance and apoptosis and inflammation in acute renal injury induced by cisplatin. The results indicate that increased antioxidant capacity in
    renal tissue attenuates the inflammatory process and cell death signaling pathways, which constitute the pathophysiology of acute renal injury induced by cisplatin.

  • A espécie vegetal a ser estudada, Aeschynomene martii Benth, do gênero Aeschynomene, família das Leguminosae, possuem compostos bioativos com grande potencial farmacológico a serem usados. O estudo teve como objetivo avaliar as atividades antinociceptiva, anti-inflamatória, antioxidante e toxicológica do caule e casca da A. martii Benth, coletada na região da Floresta Nacional Contendas do Sincorá (FLONA). O extrato etanólico foi preparado por maceração e submetido à extração por partição. Após a evaporação, foram obtidas as quatro frações do extrato bruto com os seguintes rendimentos: 13,21% (hexano), 40,92% (diclorometano), 30,68% (acetato de etila) e 8,10% (butanol) em relação ao peso do extrato bruto 17,03 g. A prospecção fitoquímica das frações foi realizada por CCD com reveladores específicos. As frações de hexano e diclorometano tiveram seus perfis cromatográficos avaliados por CG-EM com destaque para a identificação de: estigmasterol, -sitosterol, lupenona, 3,3',4',5,5',7-hexametoxiflavona, além dos ácidos graxos. A atividade antioxidante foi avaliada por teste do sequestro do radical livre DPPH, o extrato etanólico e as frações não apresentaram atividade antioxidante significativa (p < 0,05), além de serem classificadas quanto à concentração inibitória (boa, média e baixa, respectivamente). Para os ensaios antinociceptivo e antiinflamatório foram utilizados camundongos machos adultos Balb/c. O efeito antinociceptivo foi avaliado pelos testes: Contorção abdominal induzido por ácido acético 0,6%, onde todas as frações (25 mg/kg) promoveram redução significativa das respostas nociceptivas (p < 0,05). Injeção intraplantar de formalina 1,5%, na fase 1 e 2, as frações (25 mg/kg) reduziram o número de “flinches”, em comparação ao controle negativo (p < 0,05), quando comparadas ao grupo controle positivo morfina, as frações de diclorometano e butanol não apresentaram diferença estatística (p < 0,05), o edema de pata formado reduziu significativamente nas frações (25 mg/kg) de hexano, diclorometano e acetato de etila (p < 0,05). No Von Frey eletrônico por injeção intraplantar de carragenina (Cg), todas as frações (25 mg/kg) foram capazes de reduzir significativamente a hipernocicepção (p < 0,05). Para a determinação da atividade anti-inflamatória foram realizados testes de: Migração de neutrófilos para a cavidade peritoneal, onde todas as frações (25 mg/kg) apresentaram redução significativamente a migração de neutrófilos em (p < 0,05); Determinação dos níveis de nitrito, todas as frações (25 mg/kg) apresentaram redução significativa (p < 0,05). Avaliação da permeabilidade vascular por azul de Evans, que mostrou uma significativa redução do extravasamento do corante no líquido peritoneal (p < 0,05). Para determinação da toxicidade frente à Artemia salina Leach, as classificações de toxicidade das frações foram ativa no extrato etanólico e hexano, moderada em diclorometano e inativa em acetato de etila e butanol. Os resultados permitem considerar que a A. martii Benth é uma promissora fonte natural para a identificação de novos compostos bioativos e agentes terapêuticos, bem como uma espécie promissora para uso quanto fitoterápico.

  • The plant species to be studied, Aeschynomene martii Benth, of the genus Aeschynomene, family of Leguminosae, possess bioactive compounds with great pharmacological potential to be used. The objective of this study was to evaluate the antinociceptive, anti-inflammatory, antioxidant and toxicological activities of A. martii Benth stem and bark, collected in the Contendas do Sincorá National Forest region (FLONA). The ethanolic extract was prepared by maceration and subjected to partition extraction. After evaporation, the four fractions of the crude extract were obtained in the following yields: 13.21% (hexane), 40.92% (dichloromethane), 30.68% (ethyl acetate) and 8.10% (butanol) relative to the weight of the crude extract 17.03 g. The phytochemical exploration of the fractions was performed by CCD with specific developers. Fractions of hexane and dichloromethane had their chromatographic profiles evaluated by GC-MS, with the emphasis on the identification: stigmasterol, b-sitosterol, lupenone, 3,3',4',5,5',7-hexamethoxyflavone, besides the acids fatty acids. The antioxidant activity was evaluated by the DPPH free radical sequestration test, the ethanolic extract and the fractions showed no significant antioxidant activity (p < 0.05), besides being classified as inhibitory concentration (good, medium and low, respectively). For the antinociceptive and anti-inflammatory assays adult male Balb/c mice were used. The antinociceptive effect was evaluated by the following tests: 0.6% acetic acid-induced abdominal contortion, where all fractions (25 mg/kg) promoted a significant reduction in nociceptive responses (p < 0.05). Intraplantar injection of formalin 1.5%, in phases 1 and 2, the fractions (25 mg/kg) reduced the number of flinches in comparison to the negative control (p < 0.05), when compared to the positive control group morphine, dichloromethane and butanol fractions did not present statistical difference (p < 0.05), the paw edema formed significantly reduced in the fractions (25 mg/kg) of hexane, dichloromethane and ethyl acetate (p < 0.05). In the electronic Von Frey by intraplantar injection of carrageenan (Cg), all fractions (25 mg/kg) were able to significantly reduce hypernociception (p < 0.05). For the determination of anti-inflammatory activity, tests were performed: Neutrophil migration to the peritoneal cavity, where all fractions (25 mg/kg) showed a significant reduction in neutrophil migration in (p < 0.05); Determination of nitrite levels, all fractions (25 mg/kg) presented a significant reduction (p < 0.05). Evaluation of vascular permeability by Evans blue, which showed a significant reduction of dye extravasation in the peritoneal fluid (p < 0.05). To determine the toxicity to Artemia saline Leach, the toxicity classifications of the fractions were active in ethanolic extract and hexane, moderate in dichloromethane and inactivated in ethyl acetate and butanol. The results allow us to consider that A. martii Benth is a promising natural source for the identification of novel bioactive compounds and therapeutic agents, as well as a promising species for use as a herbal remedy.
